        Purification, characterization and action mechanism of plantaricin JY22, a novel bacteriocin against Bacillus cereus produced by Lactobacillus plantarum JY22 from golden carp intestine

        A novel bacteriocin-producing strain, Lactobacillus plantarum JY22 isolated from golden carp intestine, was screened and identified by its physiobiochemical characteristics and 16S rRNA gene sequence analysis. This bacteriocin, named plantaricin JY22, was purified using ethyl acetate extraction and gel filtration. Its molecular weight was approximately 4.1 kDa by SDS-PAGE analysis. The partial amino acid sequence of plantaricin JY22 was DFGFDIPDEV. It was highly heat-stable and remained active at pH range from 2.5 to 5.5, but was sensitive to protease. Plantaricin JY22 had a bactericidal mode. Scanning electron microscope analysis indicated that plantaricin JY22 damaged the morphology of cells and spores for Bacillus cereus. Moreover, the plantaricin JY22 destroyed cell membrane integrity as confirmed by the leakage of electrolytes, the losses of Na?K?-ATP, AKP, nucleic acids (OD260nm) and proteins. SDS-PAGE of B. cereus proteins further demonstrated that plantaricin JY22 had a remarkable effect on bacterial proteins.

        A CRISPR/Cas9 Cleavage System for Capturing Fungal Secondary Metabolite Gene Clusters

        More and more available fungal genome sequence data reveal a large amount of secondary metabolite (SM) biosynthetic ‘dark matter’ to be discovered. Heterogeneous expression is one of the most effective approaches to exploit these novel natural products, but it is limited by having to clone entire biosynthetic gene clusters (BGCs) without errors. So far, few effective technologies have been developed to manipulate the specific large DNA fragments in filamentous fungi. Here, we developed a fungal BGC-capturing system based on CRISPR/Cas9 cleavage in vitro. In our system, Cas9 protein was purified and CRISPR guide sequences in combination with in vivo yeast assembly were rationally designed. Using targeted cleavages of plasmid DNAs with linear (8.5 kb) or circular (8.5 kb and 28 kb) states, we were able to cleave the plasmids precisely, demonstrating the high efficiency of this system. Furthermore, we successfully captured the entire Nrc gene cluster from the genomic DNA of Neosartorya fischeri. Our results provide an easy and efficient approach to manipulate fungal genomic DNA based on the in vitro application of Cas9 endonuclease. Our methodology will lay a foundation for capturing entire groups of BGCs in filamentous fungi and accelerate fungal SMs mining.

        Nowadays, the audiences' consumption attitudes, consumption patterns and consumer groups are all in the great changes, thus it is necessary to improve the film’s revenue by excellent script selecting, accurate market positioning, effective product marketing, and accurate forecasting of the box office. This paper introduced the application and benefit of big data in the film industry chain in terms of film making and investing, film publicity and distribution, film broadcasting and film audience, pointed out many challenges that big data encountered in China’s film industry and finally provided useful suggestions for the practitioners in the film industry of all aspects.

        Calculation of dynamic stress intensity factors and T-stress using an improved SBFEM

        The scaled boundary finite element method is extended to evaluate the dynamic stress intensity factors and T-stress with a numerical procedure based on the improved continued-fraction. The improved continued-fraction approach for the dynamic stiffness matrix is introduced to represent the inertial effect at high frequencies, which leads to numerically better conditioned matrices. After separating the singular stress term from other high order terms, the internal displacements can be obtained by numerical integration and no mesh refinement is needed around the crack tip. The condition numbers of coefficient matrix of the improved method are much smaller than that of the original method, which shows that the improved algorithm can obtain well-conditioned coefficient matrices, and the efficiency of the solution process and its stability can be significantly improved. Several numerical examples are presented to demonstrate the increased robustness and efficiency of the proposed method in both homogeneous and bimaterial crack problems.

        지식공유-지식창출-기업성과 간 관계에 대한 지식유형의 의미

        이 글의 목적은 두 가지이다. 하나는 지식기반 관점에서 명시적, 암묵적 지식유형에 따른 지식공유가 지식창출, 기업 전사적 성과 및 해외자회사 성과와의 관계에서 나타내는 차이점을 고찰하는 것이다. 다른 하나는 지식창출이 지식공유-성과 간 관계를 매개하는 효과와 강도를 비교하였다. 이를 위해 중국에 진출한 글로벌 기업의 해외자회사 구성원들을 대상으로 수집한 538개 설문자료를 이용하여 구조방정식 모형으로 실증분석을 실시하였다. 분석결과에 따른 시사점은 다음과 같다. 첫째, 암묵적 지식공유와 명시적 지식공유 모두 지식창출에 양(+)의 영향을 미치는 가운데 명시적 지식공유의 성과 개선 효과가 더 큰 것으로 나타났다. 둘째, 명시적 지식공유는 기업 전사적 성과에 대해 양(+)의 영향을 미치고 해외자회사 성과에 대해서는 영향을 미치지 않는 반면 암묵적 지식공유는 기업전사적 성과와 해외자회사 성과 모두에게 영향을 미치지 않는 것으로 나타났다. 셋째, 암묵적 지식공유와 명시적 지식공유가 성과에 미치는 영향은 지식창출의 매개효과를 통해 모두 확대된다. 넷째, 해외자회사 성과에 대한 지식창출의 매개효과는 명시적 지식공유가 더 크고, 기업 전사적 성과에 대한 지식창출의 매개효과는 암묵적 지식공유가 더 큰 것으로 나타났다. This paper examines the different implications of knowledge sharing according to knowledge type in knowledge creation, the performance of MNCs, and foreign subsidiary performance and examines the mediating effect of knowledge creation between them. Then we explored the mediating strength from the Knowledge-Based View (KBV). For this purpose, we conducted a survey of 589 employees of foreign subsidiaries of MNCs that went into China. Except for unfavorable respondents we used 538 to analyze the structural equation model. The key analysis findings are as follows: First, both explicit knowledge sharing and tacit knowledge sharing have a positive effect on knowledge creation, and the effect of explicit knowledge sharing is greater. Second, only the explicit knowledge sharing has a positive effect on the performance of overall MNCs and the tacit knowledge sharing has no effect on it and neither explicit knowledge sharing nor tacit knowledge sharing has on effect on the foreign subsidiary performance. Third, the effect on the performance of overall MNCs and the foreign subsidiary performance of tacit knowledge sharing and explicit knowledge sharing all extend through the mediating effect of knowledge creation. Forth, in the implication of foreign subsidiary performance, the mediating strength of knowledge creation on explicit knowledge sharing is greater than tacit knowledge sharing but in the implication of the performance of overall MNCs, it shows that the mediating strength of knowledge creation on tacit knowledge sharing is greater than explicit knowledge sharing.

      • A Novel Image Encryption Scheme based on the LSM Chaotic System

        The Logistic-Sine map (LSM) chaotic system is introduced by combing the Logistic map (LM) and Sin map (SM). The bifurcation diagrams show that the chaotic range of LSM is much larger than these of the Logistic or Tent maps. Complexity characteristics of Logistic map, Sine map and Logistic-Sine map are analyzed based on C0 algorithm. The results show that C0 complexity value of the LSM is the largest one among the three. Then, a novel image encryption scheme based on the LSM chaotic system was proposed. First, the positions of image pixels are shuffled through swapping positions randomly by using chaotic values. The permutation sequences are ralated to plain-images by introducing the plaintext feedback technique. Second, the diffusion procedure with LSM is introduced to diffuse the image, which is composed of two rounds. The experimental results and analysis by using several security measures show that the proposed image encryption scheme has high security and efficiency.

        Four-Leg Object Recognition for Service Robot Based on Top-hat Transformation

        A method of object recognition for objects with special topological structure is developed based on top-hat transformation for service robot. The objects with four legs are easily detected and recognized by service robot with laser ranger array by this method. First, the top-hat transformation in one dimension is reviewed, then the recognition strategy of self-adapting threshold for objects with special topological structure is proposed, and the general data process for object recognition and position is proposed and analyzed. Experimental results show that the process of object recognition based on top-hat transformation proposed in this article is an effective and accurate application.
