      • Membrane Anchoring of α-Helical Proteins: Role of Tryptophan

        <P>The function of membrane proteins relies on a defined orientation of protein relative to lipid. In apparent correlation to protein anchoring, tryptophan residues are enriched in the lipid headgroup region. To characterize the thermodynamic and structural basis of this relationship in α-helical membrane proteins, we examined the role of three conserved tryptophans in the folding of the heterodimeric integrin αIIbβ3 transmembrane (TM) complex in phospholipid bicelles and mammalian membranes. In the homogenous lipid environment of bicelles, tryptophan was replaceable by residues of distinct polarities. The appropriate polarity was guided by the electrostatic potential of the tryptophan surrounding, suggesting that tryptophan can complement diverse environments by adjusting the orientation of its anisotropic side chain to achieve site-specific anchoring. As a sole membrane anchor, tryptophan made a contribution of 0.4 kcal/mol to TM complex stability in bicelles. In membranes, it proved more difficult to replace tryptophan even by tyrosine, indicating a superior capacity to interact with heterogeneous lipids of biological membranes. Interestingly, at intracellular TM helix ends, where integrin activation is initiated, sequence motifs that interact with lipids via opposing polarity patterns were found to restrict TM helix orientations beyond tryptophan anchoring. In contrast to bicelles, phenylalanine became the least accepted substitute in membranes, demonstrating an increased role of the hydrophobic effect.         Fournier gangrene with concurrent multifocal necrotizing fasciitis: a systematic review and case report

        Purpose: A patient presented to a regional surgical center with Fournier gangrene (FG) and concurrent multifocal necrotizing fasciitis (NF). Given the rarity, it was decided to undertake a systematic review to investigate the incidence and prevalence of FG with multifocal NF and consequently determine the treatment and approach to management of such presentation. Methods: Firstly, the report of the 56-year-old male patient is discussed regarding his surgical management. Secondly, a systematic review was undertaken according to PRISMA guidelines using MEDLINE, Scopus, and Embase databases. Searches used the following MeSH terms: (“fournier’s gangrene”) AND ((necrotising fasciitis) OR (necrotising soft tissue infection)). Once the search results were obtained, duplicate articles were removed. Titles, abstracts, and articles were reviewed by 2 authors. Results: The search strategy using the 3 databases revealed a total of 402 studies. Fifty-seven studies were removed due to duplication. A total of 345 records were screened via title and abstract, of which 115 were excluded. Two hundred and thirty studies were reviewed for eligibility. A total of all 230 studies were excluded; 169 were excluded as they included the incorrect patient population (patients suffered from FG or NF, but not both collectively), 60 studies were excluded due to incorrect study designs, and 1 report occurred in the wrong setting. Conclusion: This highlights that while being a relatively known, uncommon infection both FG and NF are well documented separately within the literature. However, FG with concurrent multifocal NF has not been documented within the literature.

        New method for rapidly estimating population densities of the concealed wood‐borer Monochamus alternatus (Coleoptera: Cerambycidae) in the field

        To rapidly estimate pine sawyer, Monochamus alternatus Hope, population densities in forests, the vertical distributions of M. alternatus oviposition sites and larvae on infested Masson pines (Pinus massoniana Lamb.) were studied. Results showed that the number of oviposition sites on sections of trunks between 0 and 2 m above ground was significantly greater than on sections of trunk above 2 m, and the vertical distribution had a significant logarithmic relationship with trunk height. The larval number of M. alternatus on dead infested trees had a significant difference among heights of trunks. Sections on trunks at 2–4 m usually contained the largest number of M. alternatus larvae, while the number of larvae on trunks above 10 m declined significantly, as well as in the 1 m section of trunk at the base. The vertical distribution of M. alternatus larvae on dead infested pines showed a distinct parabolic relationship with trunk height. The number of oviposition sites of M. alternatus on infested Masson pine trunks revealed a significant exponential relationship with the diameter at breast height (DBH) of trees. A significant positive linear relationship also was observed between M. alternatus larval number and DBH on the host pine trees, as well as between the numbers of larvae and oviposition sites on an individual tree. The total number of larvae in an infested tree could be calculated easily using an established equation, through counting the number of oviposition sites at 3–4 m of trunk aboveground. This study developed a practical method for rapid estimation of M. alternatus populations.
