Rapid detection of aneuploidy using FISH in uncultured amniocytes for prenatal diagnosis

목적 : 산전진단에 있어 빠른 진단을 위해 그 유용성이 널리 알려져 있는 FISH 방법을 미배양 세포에 적용할 때, 그 민감도를 높이기 위해 본 연구소의 경험과 기준을 소개하고자 한다. 방법 : 1999년 5월부터 2006년 6월까지 본연구소에서 다운증후군 고위험군, 에드워드 증후군 고위험군, 고령산모, 초음파 이상소견 등의 적응증을 주소로 시행한 7,893례의 양수검체를 대상으로 빠른 진단을 위해 8,613례의 미배양 양수세포에 FISH 검사를 시행하였다. 분석은 함춘유전연구소의 기준에 따랐으며, 기존의 세포유전학적 결과와 최종 비교하였다. 결과 : 8613례의 FISH 검사 결과, 30개 이상의 세포관찰이 가능하고, 정상인 경우 정상세포의 비율이 75%, 비정상의 경우 비정상 세포의 비율이 70%에 해당하는 8,502례의 결과를 얻었으며, 세포유전학적 결과와도 일치하였다. 결론 : 산전진단 시 빠른 진단을 위한 FISH검사는 매우 유용하며, 정확한 분석을 위해 그 기준을 마련하는 것은 매우 중요하다 하겠다. 그러나 비용과 인력이 많이 소요되는 한계점을 가지고 있다. Purpose: FISH is suggested as a useful too fer rapid detection of specific aneuploidy in uncultured amniocytes abnormality in inter phase nucleus. In this study, we are going to share our experience using FISH in prenatal diagnosis and suggest the criteria fer the diagnosis of aneuploidy by analyzing the results of FISH test. Methods: From January, 1999 to May, 2006, 8.613 tests in amniotic fluids obtained from 7,893 pregnant women were performed by using FISH for prenatal diagnosis of trisomy 21, trisomy 18 and trisomy 13. The indications of chromosome study were a screen positive for Down syndrome or Edwards syndrome in maternal serum marker screening test and an advanced maternal age (≥ 35 years old). Results: We have the 8.502 informative results from 8.613 tests (98.7 %) which is submitted our criteria and the sensitivity is 98.2 %. Conclusion: FISH on uncultured amniocytes is a rapid, clinically useful too for prenatal diagnosis, with informative specimens being highly accurate. But the limitation of FISH is both expensive and labor-in-tensive.

Flock House virus (FHV), an insect RNA virus, has a bipartite genome. FHV RNA1 can be packaged in turnip yellow mosaic virus (TYMV) as long as the FHV RNA has a TYMV sequence at the 3’-end. The encapsidated FHV RNA1 has four additional nucleotides at the 5’- end. We investigated whether the recombinant FHV RNA1 could replicate in mammalian cells. To address this issue, we prepared in vitro transcribed FHV RNAs that mimicked the recombinant FHV RNA1, and introduced them into baby hamster kidney (BHK) cells. The result showed that the recombinant FHV RNA1 was capable of replication. An eGFP gene inserted into the frame with B2 gene of the FHV RNA1 was also successfully expressed. We also observed that eGFP expression at the protein level was strong at 28℃ but weak at 30℃. Sequence analysis showed that the 3’-ends of the RNA1 and RNA3 replication products were identical to those of the authentic FHV RNAs. This indicates that FHV replicase correctly recognized an internally-located replication signal. In contrast, the 5’-ends of recombinant FHV RNA1 frequently had deletions, indicating random initiation of (+)-strand synthesis.

Thermal cutting processes that can be applied to dismantling nuclear power plants include oxygen cutting, plasma cutting, and laser cutting. According to the global trend, research projects are being carried out in various countries to upgrade laser cutting, and many studies are also being conducted in Korea with plans to apply laser cutting processes when dismantling nuclear power plants. However, with the current technology level of the laser cutting process, the maximum thickness that can be cut is limited to 250 mm. Therefore, in this study, a laser-oxygen hybrid cutting process was implemented by adding a laser heat source to the oxygen cutting process that can cut carbon steel with a thickness of 250 mm or more (RV, beam, column, beam, etc.) when dismantling the nuclear power plant. This has the advantage of improving the cutting speed and reducing the cutting width Kerf compared to conventional oxygen cutting. In this research, the laser-oxygen hybrid cutting process consisted of laser cutting to which Raycus’ 8 kW Fiber Laser power source was applied and oxygen cutting to which hydrogen was applied with Fuel Gas. The oxygen torch was placed perpendicular to the test piece, and the laser head was irradiated by tilting 35° to 70°. The effects of cutting directions on quality and performance were studied, and cutting paths were selected by comparing cutting results. Thereafter, it was confirmed that there is an optimal laser output power according to the cutting thickness by studying the effect on the cutting surface quality by changing only the laser output power under the same cutting conditions. The results of this study are expected to be helpful in the remote cutting process using laser-oxygen hybrid cutting when dismantling domestic nuclear power plants in the future.
Photoprotection effect of Pu’er tea and Curcuma longa L. extracts against UV and blue lights

Plant extracts have been studied due to their potential as photoprotective agents against UV and blue light exposure. Previous studies have revealed that several plant extracts have photoprotection capacities and synergistic effects with synthetic products. However, such results for pu’er tea and Curcuma longa L. have not been reported yet for a cosmetic formulation. Thus, the objective of this study was to evaluate photoprotection capacities of pu’er tea and C. longa L. extracts for a sunscreen compound. The pu’er tea extract improved sun protection factor value of 2-ethyl-hexyl methoxycinnamate (a synthetic sunscreen compound) by 46% and showed a high antioxidant capability that could help skin recover from photo-induced damage. C. longa L. extract also showed a potential to protect skin from blue lightinduced damage because it not only had a maximum absorption peak at the blue light range, but also protected human fibroblasts from blue light-induced damage. The addition of both extracts shifted the critical wavelength of 2-ethyl-hexyl methoxycinnamate from 350 nm to 386 nm, giving it a broad-spectrum feature. Thus, pu’er tea and C. longa L. extracts may enhance the photoprotection ability of synthetic sunscreen products.

The study is about the function of Smart Control Module using SAE J1939 Protocol for the commercial vehicles. Presently, the load factor of CAN bus is increasing because of one CAN network composition and addition of new ECU’s with thc error of CAN communication. Also, development costs and times arc increasing for the compatibility between non-standard ECUs of commercial vehicle. So, commercial automobile vendor appreciates what CAN Integrated Control System. it is to reduce thc load factor of CAN bus by function what is transmission and block of Smart Control Module. And, to secure compatibility between ECUs in the commcreial vehicle by function what is manufacturing of Smart Control Modulc. When Smart Control Module is broken, it will self-diagnose and recovcr what makes CAN bus to one network. This paper describes the features of a Smart Control Module and verification of performance is described.

This study is about the functions of Smart Control Module using SAE J1939 protocol for the commercial vehicles. Presently, the load factor of CAN bus is increasing because of one network composition and addition of new ECUs for intelligence of vehicles. System that achieve integration network control function is needed for development of commercial vehicles because of CAN bus errors by increase of ECU. This study is about the development of SCM(Smart Control Module) that can diagnosis CAN bus errors, fault diagnosis of ECU and basic function that arbitrate CAN bus between ECUs of commercial vehicle. SCM was designed 4channel separation function and Gateway function for load factor decrease. HILS(Hardware in the loop simulation)system that can achieve simulation about CAN Message of all systems on vehicle was applied to evaluate performance and verification of designed function and performance. The load factor on CAN bus was decreased at using functions what is delivery, block and process of SCM. Through this, it enabled systematic architecture organization for gateway.

Twenty years have passed since a prenatal screening for Down syndrome and neural tube defect was applied to obstetric field. The Quad test (AFP, hCG, uE3, Inhibin-A) of the second trimester and the combination test (PAPP-A, hCG, NT) of the first trimester became popular now. The recent trend of prenatal screening is to combine these two screening tests together in order to increase a detection rate of Down syndrome. Three types of screening methods are introduced as follows; integrated test, sequential test and contingent test. In addition to combination of each test, an incorporation of characteristic ultrasound findings of Down syndrome is suggested for its risk calculation. The absence of fetal nasal bone would be a very useful marker especially in the first trimester screening test. According to a change of way calculating risk of Down syndrome, obstetrician's role will be more increased not by passive participation, but by active participation using ultrasound in risk calculation.
