재조합 Saccharomyces cerevisiae에서 Permeabilizing Agent를 이용한 HLY의 분비촉진

배양 말기까지 세포내에 30% 이상 존재하는 유전산물인 HLY를 효과적으로 분비를 유도하기 위해 다섯가지의 permeabilizing agent-CaCl₂, ethanol, Tween 80, Triton X-100, CTAB-를 사용하였다. 그 결과 효모형질전환체에 대한 CaCl₂, Tween 80, ethanol의 처리에서는 HLY의 분비촉진을 확인할 수 있었다. 특히 10% ethanol의 처리에서는 HLY 활성을 38.6% 증가시켰다. 이러한 결과는 ethanol이 직접적으로 세포막과 벽에 변화를 주어 세포내에 존재하는 HLY의 분비를 촉진시켰고, 배지 속에 첨가된 고농도 ethanol은 분비된 HLY의 안정성을 증가시킨 것으로 생각된다. 하지만 기대했던 계면활성제의 효과는 거의 없었는데 이러한 결과는 계면활성제가 유전산물 분비에 미치는 영향이 없다고 단정 내리기보다는 측정방법의 문제인 것으로 보인다. We have intended to accelerate the secretion of human lysozyme(HLY) with permeabilizing agents from the cultivated cells of the recombinant Saccharomyces cerevisiae. The five agents-CaCl₂, Tween 80, ethanol, Triton X-100, and cetyltrimethylammonium bromide(CTAB) were used as permeabilizing agents. Treatments of the yeast cell with CaCl₂, Tween 80, and ethanol were effective to increase the secretion from the yeast cells. Especially, treatment of 10% ethanol increased the extracellular HLY activity by 38.6% at 30℃ for 48 h in culture broth. But Triton X-l00 and CTAB unexpectedly didn′t playa role in increase of HLY secretion. Recovery of a foreign protein by permeabilizing agents is easier than by osmotic shock, and is less expensive than enzymatic digestion.
숙시닐화 및 부분가수분해가 대두단백질 분리물의 기능적 특성과 단백질 - 단백질 상호작용에 미치는 영향

숙시닐화 또는 트립신처리에 의한 대두단백질 분리물의 화학적 변형은 단백질의 함량을 감소시키는 것으로 나타났고, 아미노산 조성에서 tyrosine의 증가가 현저하였으나 lysine은 트립신처리시에만 크게 증가하였다. 화학적 변형은 단백질의 용해성을 증가시키고 pH의존성이 뚜렷하여 등전점 변이시키는 효과를 나타내었다. 단백질의 용해성은 염류의 농도증가에 의해 감소하는 경향을 나타내었으며, 화학적 변형은 유흡수성과 수분흡수성, 유화특성 및 기포성 등을 증가시키는 반면에 기포안정성을 다소 저하시키고 자외선 흡광도와 용적밀도를 감소시켰다. 한편 대두단백질 분리물과 우육단백질 혼합에 따른 상호작용에 의해서는 유화활성, 유화활성지수 및 기포성의 증가를 가져왔으나 유화안정성에 대해서는 현저한 효과가 나타나지 않았다. Soybean protein isolates were acylated with succinic anhydride and partially hydrolyzed with trypsin. Chemical modification decreased protein contents of samples and, in amino acid composition, tyrosine was increased comparatively. And lysine was increased remarkably by partial proteolysis. Succinylation and trypsin treatment increased the aqueous solubility and shifted the isoelectric potint that showed high pH-dependence of protein solubility. Protein solubility was influenced by salt concentration such as NaCl, CaCl₂, NaNO₃ and NaH₂PO₄. Chemical modification increased the absorption of oil and water, emulsification properties and foam capacity, but decreased foam stability, ultraviolet absorbance and bulk density. Protein-protein interaction between soybean protein isolates and beef protein increased the emulsifying activity, emulsifying activity index and foaming properties, but it didn't have any influence on emulsion stability.
