Oral administration of beer and peptone meal is known to stimulate gastric acid secretion in vagally-innervated and denervated dogs, most probably by inducing an increased gastrin release. To confirm the role of circulating gastrin, it was examined whether or not YM022 (a CK-B/gastrin-receptor antagonist) and cimetidine inhibit the gastric acid secretion stimulated by beer, peptone and peptone-beer in denervated Heidenhain pouch dogs. Both YM022 and cimetidine markedly inhibited the gastric acid secretion stimulated by the above three secretagogues. The underlying mechanism for acid inhibition by YM022 appears to involve CCK-B/gastrin receptors in both parietal cells and ECL cells. In the case of cimetidine, the mechanism underlying acid inhibition appears to involve the direct action on histamine H2-receptors in parietal cells.
