This study was to investigate the identification of bacteria take part in the aging of fermented vegetable such as Tongkimchi and Dongchimi, the fermented vegetables which are indigenous to Korea were prepared in laboratory scale according to the same method used by private house in Seoul. The analogy of aerobic and anaerobic bacterial flora isolated from Kimchi was found, and their bacterial flora were identified by investigating the cultural, morphological and physiological characteristics. In results obtained are as follows; 1. During the aging of Tongkimchi. bacterial flora found in the early stage were Leuconostoc mesenteriodes, Lactobacillus plantarum, Streptococcus faecails and Pediococcus pentosaceus. In the middle period, however, Lactobacillus plantarum and streptococcus paecalis were predominant, and Lactobacillus brevis and Streptococcus faecalis subsp. liquefaciens were also fund. 2. During the aging of Dongchimi, aeroges found in the early stage were the genus Pseudomonas, and anaerobes such as Leuconostoc mesenteroides, Lactobacillus breyis, Lactobacillus plantarum and Pediococcus pentosaceus were also found. In the middle period, however, Leuconostoc mesenteroides and Pseudomonas denitriflcans were more predominant.

Saccharomyces cerevisiae Rasse O의 배양조건과 trdhalose를 중심으로한 균체성분과의 관계에 대하여

Effects of the sugar content in molasses media and pH on cell constituents of produced yeast adopting Saccharomyces cerevisiae Rasse O as a seed organism were studied, and following results were obtained. 1. Trehalose accumulation of the yeast was reduced at lower range of pH, however protein was increased. 2. Trehalose content of the yeast enriched by feeding increased sugar at suitable pH. 3. There was no significant increase of thehalose content in the cell by feeding concentrated molasses at lower range of pH.
沈菜類 熟成에 따르는 Microflora, 糖, pH 및 Vitamin B_(12)의 變化와 Pseudomonas sp.의 Vitamin B_(12) 生産性에 관하여

This study was to investigate the changes of microflora, sugar, pH, and vitamin B_(12) during the aging of fermented vegetables such as Tongkimchi and Dongchimi and the productivity of vitamin B_(12) by Pseudomonas denitrificans D-2-1 selected as vitamin B_12 producing bacteria from Dongchimi. The main bacteria responsible for Tongkimchi fermertation were Lactobacillus plantarum, Leuconostoc mesenteroides and Streptococcus faecalis, but the main bacteria for Dongchimi fermentation were Leuconostoc mesenteroides and the genus Pseudomonas. In TongKimchi and Dongchimi sugar continuously decreased from ca 3% and pH reduced from 5.8 to 4.0 and less, but increased as the over-ripening progresses. Vitamin B_(12) content of TonKimchi was higher than those of Dongchimi at the early stage, but the opposite result at a later period Vitamin B_(12) content of TongKimchi and Dongchimi reached its maximum at the middle period. Pseudomonas denitrificans D-2- 1 cultured for 15 days on the media of radish juice produced vitamin B_12 up to 274 ng/㎖ and the strain cultured on the media of radish juice containing 1.5% NaCl produced vitamin B_12 of 261 ng/㎖. The strain cultured on the media of radish juice containing 3% NaCl and 0.1% L-glutamic acid as a nitrogen source produced vitamin B_(12) of 272ng/㎖. In contrast to the amount of vitamin B_(12)(245ng/㎖) obtained from the media in which L-glutamic acid was not added, the media containing L-glutamic acid increased the productivity of vitamin B_(12) up to 11%
原發性 肝癌患者의 腹水 r-glutamyl transpeptidase 活性値에 關한 硏究

Gamma-glutamyl transpeptidase (γ-GTP) is a enzyme, cencentrated mainly in kidney, pancreas, liver, spleen, small intestine, brain and heart muscle as a very sensitive indicator of liver cell dysfunction, especially in primary hepatoma. For the purpose of differential diagnosis of ascites with primary hepatoma, the levels of ascitic γ-GTP in various gastrointestinal diseases have been estimated, the correlation between ascitic γ-GTP and serurn alkaline phosphatase, SGOT, SGPT activity, ascitic total protein amount established. Also by peritoneoscopic examination the relationship between γ-GTP levels and extent of involvement of primary hepatoma has been analyzed. The material included 19 cases of primary hepatoma, 18 cases of liver cirrhosis, 12 cases of tuberculous peritonitis, 18 cases of stomach cancer and 6 cases of benign intraabdominal tumor. The results are as follows. 1. The peak age incidence of primary hepatoma was in 6th decade and ratio between male and female was 2.8:1. 2. Mean values of ascitic γ-GTP levels was highest In primary hepatoma (60.2±30.4mU/ml) and they were 28.3±14.2mU/ml in stomach cancer, 14.8±11.6mU/ml in liver cirrhosis, 14.0±6.6mU/ml in benign intraabdominal tumor and 10.2±5.2mU/m1 in tuberculous peritonitis in decreasing order (P<0.05). 3. The correlation between ascitic γ-GTP and serum alkaline phosphatase, SGOT, SGPT activity, ascitic total protein amount were absent in primary hepatoma (P>0.05). 4. Mean values of aecitic γ-GTP levels in primary hepatoma with and without 1iver cirrhosis were 61.2±28.2mU/ml and 59.8± 32.1mU/ml, respectively. The difference of both cases was not statistically significant (P:>0.1). 5. Primary hepatoma was divided into three groups by extent of involvement undel peritoneos-copy. Mean values of γ-GTP levels in group A was 98.6±38.7mU/ml, in group B 49.8±4. OmU/ml and in group C 38.0±3.6mU/ml, respectively (P<0.05). It is concluded that significantly increased ascitic γ-GTP levels in primary hepatoma would be useful for the differential diagnosis of ascites and ascitic γ-GTP levels are related to exten of involvement of primary hepatoma.
Fission yeast 에서 tRNA precursor splicing에 관련된 유전자, ptp3의 연구

We had previously isolated six pre-tRNA^(Val) splicing mutants from a collection of temperature sensitive Schizosaccharomyces pombe strains. The six mutants had been named ptp1 to ptp6. We cloned the PTP3 gene from a genomic library by complementation of the growth defect. One colony showing complementation was isolated and the plasmid was recovered. The plasmid contained 2 4 kb insert and can complement 87% of growth defect. However, the complementation efficiency of 1 9 kb subclone of this plasmid was reduced to 60% There was no significant open reading frame in entire 2 4 kb sequence Instead, the gene contained an incomplete open reading frame and a complete tRNA^(Ser) gene sequence of S. pombe. This result suggest that the tRNA^(Ser) gene or the gene of the incomplete open reading frame may participate in the regulation of pre-tRNA^(Val) splicing.
