        Tumor suppressive effect of scavenger receptor class A member 5 overexpression in colorectal cancer by regulating PI3K/AKT/mTOR pathway

        Background Colorectal cancer (CRC) exhibits high risks of morbidity and mortality. Objective To investigate the efect of scavenger receptor class A member 5 (SCRAR5) on CRC and its mechanism on modulation of cancer development. Methods The SCRAR5 expression in four kinds of CRC cell lines (SW620, SW480, HT29, and HCT116) was measured by quantitative PCR and western blotting, respectively. The efects of SCRAR5 abnormal expression on cell proliferation, apoptosis, and migration were analyzed by CCK-8 assay, EdU assay, colony-forming assay, fow cytometry assay, Transwell assay and wound healing assay, respectively. Meanwhile, the involvements of PI3K/AKT/mTOR pathway with the role of SCRAR5 were investigated by western blotting. Afterwards, the in vivo efects of SCRAR5 abnormal expression on CRC xenograft mice were fnally investigated by evaluating tumor volume, apoptosis and Ki67 expression. Results SCRAR5 was lowly expressed in CRC cell lines, especially SW480 cells. Up-regulation of SCRAR5 signifcantly promoted cell apoptosis, reduced cell proliferation and migration in SW480 cells. Notably, SCRAR5 overexpression obviously inhibited the phosphorylation levels of PI3K, AKT, and mTOR. Reversely, SCRAR5 silence exhibited promoting efects on HT29 cells. Consistently, in vivo experiments also revealed that SCRAR5 overexpression remarkably suppressed tumor volume and Ki67 expression, as well as promoted cell apoptosis. Conclusions Overall, up-regulating of SCRAR5 obviously inhibited CRC tumor growth in vitro and in vivo, which might be related to PI3K/AKT/mTOR pathway.

        The purpose of this research is to examine how Wang Hong’s characteristics (expertise and attractiveness) and video content (informativeness and entertainment) affect consumers’ purchase intention in Wanghong Marketing, that is a representative form of live commerce in China. This study investigates whether Wang Hong’s trust plays a mediation role in the influence of Wang Hong’s characteristics and purchase intention and whether the trust in the product has a mediation effect on the characteristics of video contents and purchase intention. Also, we observed whether Wang Hong’s trust has an impact on trust in the product. Especially we examined how the Wang Hong-products’ fitness moderates the influence of Wang Hong’s characteristics on their trust and also paid attention to whether the product type (utilitarian vs. hedonic) can act as a moderation variable in the relationship between the characteristics of video content and the trust in the product. The data consists of 290 consumers who have purchased products through live commerce conducted by famous Wang Hong, and a structural equation model was used to analyze the data. The finding indicates that Wang Hong’s characteristics have a significant effect on Wang Hong’s trust. The characteristics of the video content have a significant effect on trust in the product. Besides, the higher the trust in Wanghong, the higher the reliability of the product. Regarding the moderating effect of the wanghong-products’ fitness, if the attractiveness is higher, then the trust of the wanghong becomes higher, but the expertise cannot play a moderating role. In terms of product type, compared with the hedonic product, when people purchase the utilitarian product, the informativeness of video has a greater impact on products’ trust, while the entertainment, there is no moderation effect.

        MicroRNA-576-3p Inhibits Proliferation in Bladder Cancer Cells by Targeting Cyclin D1

        MicroRNAs (miRNAs) are small, endogenous RNAs that play important gene-regulatory roles by binding to the imperfectly complementary sequences at the 3'-UTR of mRNAs and directing their gene expression. Here, we first discovered that miR-576-3p was down-regulated in human bladder cancer cell lines compared with the non-malignant cell line. To better characterize the role of miR-576-3p in bladder cancer cells, we over-expressed or down-regulated miR-576-3p in bladder cancer cells by transfecting with chemically synthesized mimic or inhibitor. The overexpression of miR-576-3p remarkably inhibited cell proliferation via G1-phase arrest, and decreased both mRNA and protein levels of cyclin D1 which played a key role in G1/S phase transition. The knock-down of miR-576-3p significantly promoted the proliferation of bladder cancer cells by accelerating the progression of cell cycle and increased the expression of cyclin D1. Moreover, the dual-luciferase reporter assays indicated that miR-576-3p could directly target cyclin D1 through binding its 3'-UTR. All the results demonstrated that miR-576-3p might be a novel suppressor of bladder cancer cell proliferation through targeting cyclin D1.

        A Novel Fluorescent Nanoswitch Based on Carbon Dots for Sensitive Detection of Hg2+ and I-

        In this paper, a novel fluorescent nanoswitch based on carbon dots (CDs) was developed for the sensitive and selective determination of Hg2+ and I-. The CDs were obtained by simple hydrothermal process and had a strong fluorescence emission at 440 nm. The fluorescence of the CDs can be selectively quenched by Hg2+ ion, and then the I- was added into the system, which can interact with Hg2+ and recover fluorescence of the CDs. Under optimal conditions, the quenching fluorescence intensity on addition of Hg2+ has obtained a satisfactory linear relationship covering the linear range of 0–50 µM with the linear relationship (R21 = 0.9991), and the limit of detection is 0.047 µM. The additions of I- could lead to the fluorescence intensity of the solution of CDs and Hg2+ (50 µM) recover rapidly, which is linearly related (R22 = 0.9957) to the concentration of I- in the range from 0 to 70 µM, the detection limit for I- was calculated to be 0.084 µM. Moreover, the developed method to detect Hg2+ and I- was evaluated in real examples, and the fluorescence switching can sensitively and selectively detect Hg2+ and I- over some potentially interfering ions, the recoveries were up to 97.8–107.0% and 96.7–106.6%, respectively.

      • Segmentation of the Synthetic Aperture Radar Image Using the Watershed Transformation and Region Merging Technique

        This paper presents an efficient algorithm that segments the synthetic aperture radar (SAR) image into several homogeneous regions by combining edge- and region-based information. A multi-direction ratio edge detector is used to capture the locally directional variation information of a SAR image and construct the ratio edge strength map (RESM) of the SAR image. Watershed transformation is performed on the thresholded RESM to obtain an over-segmentation result. An efficient hierarchical region merging procedure based on the region adjacency graph representation of the image regions is proposed. A novel dissimilarity measurement between the two adjacent regions is derived from the multiplicative noise speckle model of the multi-look SAR image. Experimental results and the performance evaluation show that the proposed method outperforms two widely used SAR image segmentation approaches.

        MicroRNA-409-3p Inhibits Migration and Invasion of Bladder Cancer Cells via Targeting c-Met

        There is increasing evidence suggesting that dysregulation of certain microRNAs (miRNAs) may con-tribute to tumor progression and metastasis. Previous studies have shown that miR-409-3p is dysregulated in some malignancies, but its role in bladder cancer is still unknown. Here, we find that miR-409-3p is down-regulated in human bladder cancer tissues and cell lines. Enforced expression of miR-409-3p in bladder cancer cells significantly reduced their migration and invasion without affecting cell viability. Bioinformatics analysis identified the pro-metastatic gene c-Met as a potential miR-409-3p target. Further studies indicated that miR-409-3p suppressed the expression of c-Met by binding to its 3-untranslated region. Silencing of c-Met by small interfering RNAs phenocopied the effects of miR-409-3p overexpression, whereas restoration of c-Met in bladder cancer cells bladder cancer cells overexpressing miR-409-3p, partially reversed the suppressive effects of miR-409-3p. We further showed that MMP2 and MMP9 may be downstream effector proteins of miR-409-3p. These findings indicate that miR-409-3p could be a potential tumor suppressor in bladder cancer.

        Effects of zinc-bearing palygorskite on rumen fermentation in vitro

        Objective: The aim of the study was to investigate the effect of zinc-bearing palygorskite (Zn-Pal) on rumen fermentation by in vitro gas-production system. Methods: In trial, 90 incubators were evenly divided into five groups: control (0% Zn-Pal), treatment I (0.2% Zn-Pal), treatment II (0.4% Zn-Pal), treatment III (0.6% Zn-Pal), and treatment IV (0.8% Zn-Pal). The contents of zinc for treatments were 0, 49, 98, 147, 196 mg/kg, respectively. The main chemical composition and microstructure of Zn-Pal was investigated by X-ray diffraction. The physicochemical features were evaluated by Zeta potential analysis, cation-exchange capacity, ethylene blue absorption and specific surface area (the Brunauer–Emmett–Teller method). In vitro gas production (GP) was recorded at 3, 6, 9, 12, 18, 24, 36, 48, 60, and 72 h incubation. Incubation was stopped at 0, 6, 12, 24, 48, and 72 h and the inoculants were tested for pH, microbial protein yield (MCP), NH3-N, volatile fatty acids (VFAs), lipopolysaccharide (LPS). Results: The results showed that the GP in the treatment groups was not significantly different from the control groups (p>0.05). Compared to the control group, pH was higher at 24 h, 48 h (p<0.05), and 72 h (p<0.01) (range 6 to 7). The concentration of NH3-N in the three treatment groups was higher than in the control group at 24 h (p<0.01), meanwhile, it was lower at 48 h and 72 h (p<0.01), except in the treatment IV. The concentration of MCP in treatment I group was higher than in the control at 48 h (p<0.01). Compared with control, the LPS concentration in treatment III became lower at 12 h (p<0.05). Total VFAs in treatments were higher than in the control at 24 h, 48 h (p<0.05). Conclusion: These results suggest that the addition of Zn-Pal can improve the rumen fermentation, especially when adding 0.2% Zn-Pal.
