        High-Speed Milling of Hardened Mold Steel P20 with Minimum Quantity Lubrication

        Strong thermal–mechanical coupling and rapid tool failure easily occur in the milling process of hardened steel with solid end mills. This work investigates the effectiveness of minimum quantity lubrication (MQL) during high-speed milling of hardened mold steel. Three kinds of spraying modes were designed in the experiment. They are namely the spraying ways to the flank face (MQL-F), to the rake face (MQL-R) and to the rake and flank faces (MQL-FR). The cutting forces, tool wear, and chips when using the MQL modes and compressed air cooling mode (CAIR-F) were compared, and the wear mechanism and lubrication mechanism were further analyzed. The results showed that the tool wear values and milling forces of several MQL modes were significantly lower than those with CAIR-F. The milling force and tool wear values were successively reduced using MQL-R, MQL-F, and MQL-FR, with better lubrication of the rake face resulting in a smaller curvature radius for the chip. Abrasion wear, adhesion/attrition wear, and flaking/chipping were the main wear mechanisms. The MQL-FR method was the optimal method of the MQL modes. It could effectively reduce the cutting contact stress and unit cutting energy, and obviously reduced wear phenomena such as adhesion and chipping.

        Soluble Axl Is a Novel Diagnostic Biomarker of Hepatocellular Carcinoma in Chinese Patients with Chronic Hepatitis B Virus Infection

        Purpose The purpose of this study was to evaluate the diagnostic value of soluble Axl (sAxl) in hepatocellular carcinoma (HCC) in comparison with serum "-fetoprotein (AFP). Materials and Methods Eighty HCC patients, 80 liver cirrhosis patients (LC), 80 patients with hepatitis B virus (HBV) infection, and 80 healthy controls (HC) were enrolled. sAxl levels were measured by an enzyme-linked immunosorbent assay, serum AFP levels were measured by an electrochemiluminescence immunoassay. Receiver operating characteristic (ROC) curves were used to evaluate diagnostic performances. Results The results show that levels of sAxl were high expression in patients with HCC (p < 0.05), varied with disease state as follows: HCC > LC > HC > HBV. Logistic regression and ROC curve analysis identified the optimal cut-off for sAxl in differentiating all HCC and non-HCC patients was 1,202 pg/mL (area under the receiver operating characteristic [AUC], 0.888; 95% confidence interval [CI], 0.852 to 0.924) with sensitivity 95.0%, specificity 73.3%. Furthermore, differential diagnosis of early HCC with non-HCC patients for sAxl showed the optimal cut-off was 1,202 pg/mL (AUC, 0.881; 95% CI, 0.831 to 0.931; sensitivity, 94.1%; specificity, 73.3%). Among AFP-negative HCC patients with non-HCC patients, the cut-off was 1,301 pg/mL (AUC, 0.898; 95% CI, 0.854 to 0.942) with a sensitivity of 84.6%, a specificity of 76.3%. The optimal cut-off for sAxl in differentiating all HCC and chronic liver disease patients was 1,243 pg/mL (AUC, 0.840; 95% CI, 0.791 to 0.888) with sensitivity 93.8%, specificity 61.9%. The combination of AFP and sAxl increased diagnostic value for HCC. Conclusion sAxl outperforms AFP in detecting HCC, especially in early HCC and in AFP-negative HCC. Combination sAxl with AFP improved the specificity for early HCC diagnosis. In summary, sAxl is a candidate serum marker for diagnosing HCC.

        Combining Ultrasound-Mediated Intracellular Delivery with Microfluidics in Various Applications

        Ultrasound-mediated intracellular delivery is one of the popular technologies based on membrane rupture at present. To date, ultrasound directly acts on a large number of cells to achieve cargo delivery and has been widely used in drug deliv-ery, disease therapy and other fields. However, the existing macroscopic methods can no longer meet the requirements of accurate tracking and analysis and are prone to extensive cell damage and even death. With the rapid advancements in microfl uidic technologies, the combination of ultrasound and microfluidics (CUM) technology can effectively improve the delivery efficiency and cell survival rates. This new technology has rapidly become a new direction and focus of research. Thus, we analysed the mechanism of sonoporation and the effect of acoustic waves in a microfluidic channel. In addition, we reviewed the application of these new technologies in terms of structure and fabrication of ultrasound transducers and microfl uidic devices. As regards our main objective, we hope to help researchers better understand the future developments and the challenges of new technologies. With this review, researchers can promote the development of new technologies to solve the current challenges of intracellular delivery and advance clinical applications.
