      • Loss of Expression of Cyclin D2 by Aberrant DNA Methylation: a Potential Biomarker in Vietnamese Breast Cancer Patients

        DNA methylation of tumor suppressor gene promoters is the most frequent phenomenon leading to inactivation of function, consequently driving malignant cell transformation. Cyclin D2 is implicated in tumor suppression. In our study, we carried out the MSP assay to evaluation the methylation status at CpG islands in the cyclin D2 promoter in breast cancer cases from the Vietnamese population. The results showed that the frequency of methylation reached 62.1% (59 of 95 breast cancer tumors), but was low in non-cancer specimens at 10% (2 of 20 non-cancer specimens). Additionally, with an RR (relative risk) and OR (odd ratios) of 6.21 and 14.8, DNA hypermethylation of cyclin D2 increased the possibility of malignant transformation. Our results confirmed the cyclin D2 hypermethylation could be used as the potential biomarker which could be applied in prognosis and early diagnosis of Vietnamese breast cancer patients.

      • BRCA1 Promoter Hypermethylation Signature for Early Detection of Breast Cancer in the Vietnamese Population

        Breast cancer, a leading cause of death among women in most countries worldwide, is rapidly increasing in incidence in Vietnam. One of biomarkers is the disruption of the genetic material including epigenetic changes like DNA methylation. With the aim of finding hypermethylation at CpG islands of promoter of BRCA1 gene, belonged to the tumor suppressor gene family, as the biomarker for breast cancer in Vietnamese population, sensitive methyl specific PCR (MSP) was carried out on 115 samples including 95 breast cancer specimens and 20 normal breast tissues with other diseases which were obtained from Ho Chi Minh City Medical Hospital, Vietnam. The result indicated that the frequency of BRCA1 hypermethylation reached 82.1% in the cases (p<0.001). In addition, the DNA hypermethylation of this candidate gene increased the possibility to be breast cancer with high incidence via calculated odd ratios (p<0.05). In conclusion, hypermethylation of this candidate gene could be used as the promising biomarker application with Vietnamese breast cancer patients.

        One-Pot Preparation of Antibacterial Electrospun Polycaprolactone Membrane Embedded with Gamma Irradiation-Induced Silver Nanoparticles

        In this study, we proposed a straightforward electrospun polycaprolactone (PCL) loaded with silver nanoparticles (SNPs)membrane fabrication process, in which SNPs were directly synthesized from silver nitrate (AgNO3) in PCL–acetone mixtureby gamma irradiation. The insolubility of AgNO3in PCL solution was solved using an auxiliary dimethyl sulfoxide solvent. As a physical approach, gamma rays readily converted silver ions into SNPs without the addition of harmful reductionagents, which reduced the cytotoxicity of the synthesized material. By avoiding some processes such as purification, solventremoval, or redispersion of SNPs, this method was more time-saving compared to other related studies. SNPs formation wasconfirmed by both UV–Visible spectrum (UV–Vis) and X-ray diffraction analysis. Scanning electron microscopy (SEM)revealed that the addition of SNPs significantly reduced the fiber diameter of PCL–Ag membranes compared to that of rawPCL. Uniform spherical-shaped SNPs incorporated in PCL fibers were observed under transmission electron microscopy(TEM). The tensile test showed that the electrospun PCL–Ag membranes exhibited good mechanical characteristics. Moistureeasily penetrated the porous microstructure of PCL–Ag, facilitating wound humidity regulation. Inductively coupledplasma-mass spectroscopy (ICP-MS) was employed to study the release profiles of SNPs at different time intervals. Overall,the PCL–Ag 500 ppm sample exerted excellent antibacterial activity against Pseudomonas aeruginosa and Staphylococcusaureus strains and low in vitro cytotoxicity.

        HER2 Status and Its Heterogeneity in Gastric Carcinoma of Vietnamese Patient

        Background: Human epidermal growth factor receptor 2 (HER2) is related to the pathogenesis and poor outcome of numerous types of carcinomas, including gastric carcinoma. Gastric cancer patients with HER2 positivity have become potential candidates for targeted therapy with trastuzumab. Methods: We investigated 208 gastric cancer specimens using immunohistochemistry (IHC), fluorescence in situ hybridization and dual in situ hybridization (ISH). We also investigated the concordance between IHC and ISH. The correlation between HER2 status and various clinicopathological findings was also investigated. Results: In total, 15.9% (33/208) and 24.5% (51/208) of gastric cancers showed HER2 gene amplification and protein overexpression, respectively. A high level of concordance between ISH and IHC analyses (91.3%, κ = 0.76) was found. A significant correlation between HER2 status and intestinal-type (p < .05) and differentiated carcinomas (p < .05) was also noted. The HER2 heterogeneity was high in gastric cancers; we found 68.8% phenotypic heterogeneity and 57.6% genotypic heterogeneity. Heterogeneity in HER2 protein expression and gene amplification showed a close association with diffuse histologic type and IHC 2+. Conclusions: HER2 protein overexpression and gene amplification were detected in 24.5% and 15.9% of gastric cancer specimens, respectively. Intestinal-type showed a higher level of HER2 protein overexpression and gene amplification than diffuse type. HER2 status also showed a significant relationship with well- and moderately-differentiated carcinomas. The ratio of phenotypic and genotypic heterogeneity of HER2 was high in gastric carcinomas and was associated with HER2 IHC 2+ and diffuse histologic type.
