Modulation of Calcium Currents by Nociceptin/Orphanin FQ in TRIGEMINAL NEURONS

We examined if nociceptin modulates calcium currents in sensory neurons. Nociceptin inhibited high voltage-activated Ca channel (HVACC) currents in a dose-dependent manner in subsets of trigeminal ganglion (TG) neurons acutely isolated from neonatal rat. The subpopulation of TG neurons responsive to nociceptin also responded to DAMGO, the representative p-opioid receptor (μOR) agonist, indicating that nociceptin preferentially inhibited HVACC in a subpopulation of small nociceptive neurons expressing μOR. The HVACC current inhibition was voltage-dependent, exhibited prepulse facilitation, and was blocked by N-ethylmaleimide, indicating that it was mediated by Gi/Go protein. The calcium current inhibition was blocked by an opioid receptor like-1 (ORL-1) antagonist UFP-101, but not by non-specific opioid receptor antagonist naloxone. These data suggest that the HVACC inhibition by nociceptin through the activation of ORL-1 may contribute to its analgesic actions and modulate neuronal signaling in trigeminal system.

Objectives: This study investigated quality among three herb medicine extract granules(DSGOST) which were made from different companies to check quality control of herb medicine extract granules. Methods: we selected three DSGOST extract granules which were made from different companies. And we experimented extract granules by method from K.P(Korean Pharmacopoeia), K.H.P(Korean Herbal Pharmacopoeia) of KFDA. Results: In qualitative analysis of DSGOST, we indentified Akebiae Caulis (木通), Asari Herba Cum Radix (細辛), Evodiae Fructus (吳茱萸) in three different DSGOST extract granules. In quantitative analysis of DSGOST, Medication A,B,C contained similar content of Paeoniflorin & Glycyrrhizic acid. However Medication B contains especially lowest value of Cinnamic acid & total Decursin. Conclusions: Herb medicine extract granules have different contents of ingredients although those were made by same prescription. And these differences may influence medicinal effect to patients. So we need to make system of quality control with various research of quantitative & qualitative analysis about herb medicine extract granules.

본 연구는 아이스플랜트의 기능성 소금 대체제로서의 가능성을 조사하고자 천연 및 정제소금과의 NaCl, D-pinitol, 총 페놀, 총 플라보노이드 및 DPPH 라디칼 소거능에 대한 기초자료를 비교 조사하였다. 아이스플랜트 동결건조분말의 NaCl 함량을 분석한 결과 19%이었고 D-pinitol의 함량은 777 mg/100 g으로 천일염 및 정제염에 비해 NaCl 함량은 유의적으로 낮으나 D-pinitol을 함유하고 있어 건강기능성에는 도움이 될 것으로 사료된다. 또한, 아이스플랜트 동결건조분말의 총 페놀 및 플라보노이드 함량은 각각 115 mg/100 g 및 985 mg/100 g을 함유하고 있어 DPPH라디칼 소거능의 항산화효과를 갖는 것으로 나타났다. 따라서 아이스플랜트 분말의 bioactive compound을 함유한 대체 소금 천연소재로서 가능성을 확인한 기초 연구를 제공하였다. This study investigated the suitability of freeze-dried ice plant (Mesembryanthemum crystallinum) as the source of an edible healthy salt alternative by examining the content of NaCl, D-pinitol, total phenols, total flavonoids, and DPPH radical scavenging activity compared with those of natural and purified salts. Our results showed that the NaCl content of freeze-dried ice plant, natural and purified salts was 19, 94 and 99%, respectively. The D-pinitol content of freeze-dried ice plant was 777 mg/100 g, whereas D-pinitol was not detected in either natural or purified salts. The total phenol and flavonoid contents of freeze-dried ice plant were 115 mg GAE/100 g and 985 mg RE/100 g, respectively. The DPPH radical scavenging activity of freeze-dried ice plant was markedly higher than that of natural and purified salts. Altogether, these results show that freeze-dried ice plant contains both NaCl and bioactive compounds and can be used as a source of edible salt with positive health effects.

The effects of extremely low frequency electromagnetic fields (EMF) on intracellular Ca<sup>2+</sup> mobilization and cellular function in RBL 2H3 cells were investigated. Exposure to EMF (60 Hz, 0.1 or 1 mT) for 4 or 16 h did not produce any cytotoxic effects in RBL 2H3 cells. Melittin, ionomycin and thapsigargin each dose-dependently increased the intracellular Ca<sup>2+</sup> concentration. The increase of intracellular Ca<sup>2+</sup> induced by these three agents was not affected by exposure to EMF (60 Hz, 1 mT) for 4 or 16 h in RBL 2H3 cells. To investigate the effect of EMF on exocytosis, we measured beta- hexosaminidase release in RBL 2H3 cells. Basal release of beta-hexosaminidase was 12.3±2.3% in RBL 2H3 cells. Exposure to EMF (60 Hz, 0.1 or 1 mT) for 4 or 16 h did not affect the basal or 1ՌM melittin-induced beta-hexosaminidase release in RBL 2H3 cells. This study suggests that exposure to EMF (60 Hz, 0.1 or 1 mT), which is the limit of occupational exposure, has no influence on intracellular Ca<sup>2+</sup> mobilization and cellular function in RBL 2H3 cells.

<P>The effects of bond coat nature in thermal barrier coating (TBC) systems on the delamination or fracture behavior of the TBCs with different bond coats prepared using two different processes-air plasma spray (APS) and high velocity oxyfuel (HVOF)-were investigated by cyclic thermal fatigue tests. The TBCs with the HVOF bond coat were delaminated or fractured after 3-6 cycles, whereas those with the APS bond coat were delaminated after 10 cycles or show a sound condition. These results indicate that the TBC system with the APS bond coat has better thermal durability than the system with the HVOF bond coat under long-term cyclic thermal exposure. The hardness values of the TBCs (top coats) in both systems are dependent on applied loads, irrespective of the hardness of the bond coats and the substrate. The values are not responded to the bond coat nature or the exposure time. Thermally grown oxide (TGO) layers in both cases consist of two regions with the inner TGO layer containing only Al2O3 and the outer TGO layer of mixed-oxide zone containing Ni, Co, Cr, Al in Al2O3 matrix. The outer TGO layer has a more irregular shape than the inner TGO layer, and there are many pores within the outer layer. At failure, the TGO thickness of the TBC system with the HVOF bond coat is 9-13 m, depending on the total exposed time, and that of the TBC system with the APS bond coat is about 20 m. The both TBC systems show the diffusion layer on the side of substrate in the interface between the bond coat and the substrate. The relationship between the delamination or fracture behavior and the bond coat nature has been discussed, based on the elemental analysis and microstructural evaluation.</P>

<P>Corn is a major staple food, along with rice and wheat, in many parts of the world. There are several reports of hypersensitivity to maize pollen. However, cases of occupational allergic rhinitis induced by inhalation of maize pollen are very rare. We herein report the case of a 67-year-old male with occupational rhinitis caused by occupational exposure to maize pollen in a cornfield. He showed positive responses to maize pollen, as well as grass pollens, in skin prick tests. A high level of serum immunoglobulin E (IgE) specific to maize pollen extracts was detected by an enzyme-linked immunosorbent assay (ELISA). Laboratory tests showed a high serum level of total IgE (724 kU/L) and a high level of IgE specific to maize pollen (8.32 kU/L) using the Immuno-CAP system. Occupational rhinitis was confirmed by a nasal provocation test with maize pollen extracts. IgE ELISA inhibition tests showed antibody cross-reactivity between maize pollen and grass pollen extracts. IgE immunoblotting using maize pollen extracts demonstrated a 27 kDa IgE-binding component. These findings suggest that maize pollen can induce IgE-mediated occupational rhinitis in exposed workers.</P>
