A microchip electrophoresis (ME) method was developed using a programmed field strength gradients (PFSG) for the single nucleotide polymorphism (SNP) based fast identification of cattle breeds. Four different Korean cattle (Hanwoo) and Holstein SNP markers amplified by allele-specific polymerase chain reaction were separated in a glass microchip filled with 0.5% poly(ethyleneoxide) ($M_r$ = 8 000 000) by PFSG as follows: 750 V/cm for 0 - 14 s, 166.7 V/cm for 14 - 31 s, 83.3 V/cm for 31 - 46 s, and 750 V/cm for 46 - 100 s. The cattle breeds were clearly distinguished within 45 s. The ME-PFSG method was 7 times and 5 times faster than the constant electric field ME method and the capillary electrophoresis- PFSG method, respectively, with a high resolving power ($R_s$ = 5.05 - 9.98). The proposed methodology could be a powerful tool for the fast and simultaneous determination of SNP markers for various cattle breeds with high accuracy.

<P>The fruit of <I>Vaccinium bracteatum</I> Thunb. (VBF) is commonly known as the oriental blueberry in Korea. The aim of this study was to evaluate the antidepressant-like effects of water VBF extract (VBFW) in a mouse model of chronic restraint stress (CRS) and to identify the underlying mechanisms of its action. The behavioral effects of VBFW were assessed in the forced swim test (FST) and open field test (OFT). The levels of serum corticosterone (CORT), brain monoamines, in addition to the extracellular signal-regulated kinases (ERKs)/protein kinase B (Akt) signaling pathway were evaluated. VBFW treatment significantly reduced the immobility time and increased swimming time in FST without altering the locomotor activity in unstressed mice. Furthermore, CRS mice treated with VBFW exhibited a significantly decreased immobility time in FST and serum CORT, increased locomotor activity in OFT, and enhanced brain monoamine neurotransmitters. Similarly, VBFW significantly upregulated the ERKs/Akt signaling pathway in the hippocampus and PFC. In addition, VBFW may reverse CORT-induced cell death by enhancing cyclic AMP-responsive element-binding protein expression through the up-regulation of ERKs/Akt signaling pathways. In addition, VBFW showed the strong antagonistic effect of the 5-HT<TEX>$ _{2\text{A}}$</TEX> receptor by inhibiting 5-HT-induced intracellular Ca<TEX>$ ^{2+}$</TEX> and ERK1/2 phosphorylation. Our study provides evidence that antidepressant-like effects of VBFW might be mediated by the regulation of monoaminergic systems and glucocorticoids, which is possibly associated with neuroprotective effects and antagonism of 5-HT<TEX>$ _{2\text{A}}$</TEX> receptor.</P>

<P>The present study evaluates the anti-oxidative stress activity of <I>Vaccinium bracteatum</I> Thunb. fruit extract (VBFW) to identify the mechanisms responsible for its antidepressant-like effects. To evaluate the antidepressant and anti-oxidant effects of VBFW, malondialdehyde (MDA), serotonin transporter (SERT), and monoamine oxidase A (MAO-A) levels were measured in a mouse model of chronic restraint stress (CRS). The underlying mechanisms preventing oxidative stress and neuronal apoptosis were investigated using <I>in vitro</I> models of hydrogen peroxide (H<SUB>2</SUB>O<TEX>$ _{2})$</TEX>-induced neuronal damage. The results showed that VBFW treatment (200<TEX>$ \,$</TEX>mg/kg) significantly reduced MDA, SERT, and MAO-A levels in the prefrontal cortex of CRS mice. Furthermore, VBFW (30<TEX>$ \,$</TEX><TEX>$ \mu $</TEX>g/mL) exhibited protective effects against H<SUB>2</SUB>O<SUB>2</SUB>-induced cell death via inhibition of the H<SUB>2</SUB>O<SUB>2</SUB>-induced increase in Bax and decrease in Bcl-2 levels within the mitochondria of SH-SY5Y cells. Furthermore, VBFW (10 and 30<TEX>$ \,$</TEX><TEX>$ \mu $</TEX>g/mL) exerted protective effects against H<SUB>2</SUB>O<SUB>2</SUB>-induced cell death through inhibition of key mitochondria-associated apoptotic proteins such as cytochrome c, caspase-3 and PARP. Additionally, VBFW (10 and 30<TEX>$ \,$</TEX><TEX>$ \mu $</TEX>g/mL) could improve the activity of anti-oxidant enzymes (such as SOD and catalase) in H<SUB>2</SUB>O<SUB>2</SUB>-treated SH-SY5Y cells. These results suggest that the antidepressant and anti-oxidant effects of VBFW might be mediated by the regulation of SERT and MAO-A, and possibly associated with regulation of oxidative stress-induced apoptosis.</P>

Septo-optic dysplasia is a congenital anomaly with diverse phenotypes from normal to mixtures of visual abnormality, endocrine dysfunction, psychomotor retardations and epileptic seizures. It is characterized by optic atrophy, pituitary dysfunction and midline structure abnormalities in corpus callosum or septum pellucidum. Diagnosis of septo-optic dysplasia plus is made when cortical malformations accompanied. Here we report a middle-aged woman with septo-optic dysplasia plus having unilateral optic atrophy, agenesis of septum pellucidum and cortical malformations.

<P><B>Abstract</B></P><P>Hydroxy fatty acids are used as starting materials for the production of secondary metabolites and signalling molecules as well as in the manufacture of industrial fine chemicals. However, these compounds are usually difficult to produce from renewable biomass by chemical means. In this study, linoleate double bond hydratases of <I>Lactobacillus acidophilus</I> NBRC 13951 were cloned for the first time. These enzymes were highly specific for the hydration of the C‐9 or the C‐12 double bond of unsaturated fatty acids (e.g., linoleic acid). Thereby, the enzymes allowed the selective production of hydroxy fatty acids such as 13‐hydroxy‐<I>cis</I>‐9‐octadecenoic acid and 10‐hydroxy‐<I>cis</I>‐12‐octadecenoic acid from linoleic acid. In addition, the hydroxy fatty acids were further converted into industrially relevant carboxylic acids (e.g., 12‐hydroxy‐<I>cis</I>‐9‐dodecenoic acid, α,ω‐tridec‐9‐enedioic acid) and lactones (i.e., δ‐decalactone, γ‐dodecelactone) <I>via</I> whole‐cell biocatalysis using an enzyme cascade. This study thus contributes to the preparation of hydroxy fatty acids, unsaturated carboxylic acids, and lactones from renewable unsaturated fatty acids.</P>
Fracture and long-term aging characteristics of GFRP clamping plates for improved movable weirs

<P> In this study, the durability of movable weirs was improved by replacing steel clamping plates with glass fiber-reinforced polymer (GFRP) material. Because the clamping plates of movable weirs are always in contact with water, the service life of weirs is reduced due to corrosion. Other environmental conditions also degrade movable weirs, such as exposure to continuous inundation, dry environments, repeated dry and wet conditions, and chemical environments. This study evaluated the absorption, fracture, and long-term aging properties of GFRP clamping plates for improved movable weirs. Absorption increased with immersion time in 60°C tap water, sodium sulfate (Na<SUB>2</SUB>SO<SUB>4</SUB>) solution, or calcium chloride (CaCl<SUB>2</SUB>) solution. However, the total absorption for 50 days and 100 days exposure was low, less than 0.50% and 0.62%, respectively. In fracture load tests, the GFRP clamping plates showed the largest reduction in strength after exposure to the CaCl<SUB>2</SUB> solution; 70% and 67% of the control load were retained for exposures of 50 days and 100 days, respectively. After exposure to both tap water and the Na<SUB>2</SUB>SO<SUB>4</SUB> solution, the GFRP clamping plates showed residual strengths of 77% and 69% after 50 days and 100 days exposure, respectively. The GFRP clamping plates showed 74% and 71% residual strength after exposure to repeated freeze-thaw environments for 50 days and 100 days, respectively, and 80% residual strength after exposure to all other environments for 50 days or 100 days. Both vertical and horizontal cracks were generated before fracture after exposure to an environment involving direct contact with moisture. Without moisture, only horizontal cracks were generated before fracture. </P>

Lactic acid bacteria have been reported their beneficial roles on host including reduction of infectious diarrhea problems. In this study, preventive effect of Lactobacillus (L.) reuteri HY25101 and L. johnsonii HY25103 on porcine epidemic diarrhea virus (PEDV) was investigated in suckling piglets. Two groups of one day old PEDV naïve piglets were orally administered L. reuteri HY25101 and L. johnsonii HY25103 for three days respectively before challenge with lethal dose of PEDV. In second experiment, passive immunized one day old piglets using colostrums containing PEDV specific IgA were used. The survival rates of the L. reuteri HY25101 administered group were significantly higher than that of L. johnsonii HY25103 administered group and viral shedding was rapidly diminished in L. reuteri HY25101 administered group. Interestingly piglets born from the sow immunized with attenuated PEDV vaccine were not completely protected from PEDV challenge, however coadministeration of L. reuteri HY25101 and colostrums containing PEDV specific IgA were more effectively prevent PEDV infection. These results suggested that dietary treatment using L. reuteri HY25101 could reduce diarrheal problem and mortality rate caused by PEDV in suckling pigs. In addition, L. reuteri HY25101 could be used as one of effective compensation treatment with attenuated live vaccine for PED.
