        Engineering a Highly Thermostable and Stress Tolerant Superoxide Dismutase by N-terminal Modification and Metal Incorporation

        Thermophilic or hyperthermophilic SODs (superoxide dismutase) usually offer substantial biotechnological advantages over mesophilic SODs. Previously a 244-amino acid N-terminal domain (NTD) from a heatresistant SOD of Geobacillus thermodenitrificans NG80-2 was discovered and demonstrated to be able to confer thermostability to homologous mesophilic SODs, which revealed a new type of heat resistance mechanism. To further improve the heat resistance and stress tolerance of thermophilic cambialistic superoxide dismutase (Fe/Mn- SODAp) from Aeropyrum pernix K1 through metal incorporation and fusion with the newly found peptide NTD for broadening its industrial application, the wildtype SODAp and NTD-fused ntdSODAp were expressed in E. coli BL21 and incorporated with metal cofactors by two ways. Recombinant fusion SOD obtained by in vitro reconstitution (Mn-rec ntdSODAp) exhibited improved optimum temperature at 70oC and dramatically enhanced thermostability especially at 110oC with enhanced pH stability from 4 to 10 and higher tolerance for denaturants and organic media than Mn-rec SODAp. To the best of our knowledge, Mn-rec ntdSODAp could be the most heat resistant SOD. In addition, metal incorporation of SODAp and ntdSODAp via in vivo modification have been developed and proved to be more practical for industrial use. These results indicate that fusion with NTD along with metal incorporation can generate superimposed effect and be applied to enhance the stability of cambialistic thermophilic SODs, thus providing a universal and convenient bioengineering method for generating extremely stable SODs.

        중국 가계의 부채 보유에 미치는 영향요인 연구

        This study analyzes China's accelerated household debt growth and its influencing factors. We utilize data from the 2022 China Household Panel Survey (CFPS) and conduct empirical analysis using Tobit and Probit models. The findings are as follows: First, as the age of the head of household increases, the probability of household debt holding follows an inverted U-shaped trend, rising in the 30s and falling in the 40s and beyond, supporting the life-cycle theory. Second, male and married heads of households have a higher probability of increasing debt, suggesting that household financial decisions are mainly made by men in China. Third, holding financial and real estate assets increases the likelihood of having debt. This indicates that real estate is seen as an essential investment in China and that households use debt to improve their wealth. Fourth, as households' net income increased, the asset-liability ratio declined. This suggests that as households' net income increases, their expenditure burden decreases, reducing the demand for debt. In Korea, households tend to increase leverage for real estate investment as their net income increases. Still, in China, households strongly prefer "saving" and a culture that prioritizes debt repayment, resulting in a different perception of household debt than in Korea. Households in China have different attitudes toward debt utilization depending on the size of their wealth due to the large gap between rich and poor. The results of this study are based on the life cycle theory, and caution should be exercised in interpreting the results of the asset-related variables.

        중국 신선식품 O2O플랫폼의 서비스 품질요인이 재이용 의도에 미치는 영향 연구 : 경제적 가치와 감성적 가치의 매개효과를 중심으로

        본 연구는 중국의 신선식품 O2O 플랫폼 서비스 품질이 소비자의 지각된 경제적 가치와 감성적 가치에 미치는 영향과 이들이 재이용 의도에 어떻게 작용하는지에 대해 실증적으로 연구하였다. 분석 결과, 서비스 품질의 하위 요인인 유용성, 적시성, 공감성, 용이성, 신뢰성이 소비자가 지각하는 경제적 가치에 긍정적 영향을 미치며, 이 중 유용성의 영향이 가장 높은 것으로 나타났다. 공감성은 지각된 감성적 가치에 가장 큰 영향을 미쳤고, 신뢰성과 용이성이 뒤를 이었다. 재이용 의도와 관련해서는 적시성이 가장 중요한 요인으로 나타났으며, 용이성과 유용성이 중요한 요인으로 확인되었다. 경제적 가치와 감성적 가치는 서비스 품질과 재이용 의도 사이에서 매개 역할을 하며, 경제적 가치는 서비스 품질이 재이용 의도에 미치는 영향을 강화하는 한편, 감성적 가치는 소비자의 긍정적 감정이 장기적인 행동 변화로 이어지는 중요한 요소로 작용함을 확인할 수 있었다. 이러한 결과는 중국의 O2O 플랫폼을 통한 신선식품 판매 확대 전략에 있어서 유용성과 공감성의 중요성을 강조하며, 경제적 가치와 감성적 가치를 지속적으로 제공할 수 있는 서비스 품질의 차별화 전략이 필요함을 시사한다. This study empirically analyzes the impact of service quality of fresh food O2O platforms in China on consumers perceived economic value and emotional value, and how they affect their reuse intention. The results show that the sub-factors of service quality - usefulness, timeliness, fairness, ease, and reliability - have a positive impact on consumers perceived economic value, with usefulness having the highest impact. Fairness had the strongest impact on perceived emotional value, followed by reliability and ease of use. Regarding reuse intentions, timeliness was found to be the most important factor, followed by ease and usefulness. Economic value and emotional value mediated the relationship between service quality and repetition intentions, with economic value strengthening the impact of service quality on repetition intentions, while emotional value was found to be an important factor in consumers positive emotions leading to long-term behavior change. These findings highlight the importance of usefulness and fairness in the strategy of expanding fresh food sales through O2O platforms in China, and suggest the need for a differentiated service quality strategy that can consistently deliver both economic and emotional value.

        LRIG1 Enhances Chemosensitivity by Modulating BCL-2 Expression and Receptor Tyrosine Kinase Signaling in Glioma Cells

        Purpose: Leucine-rich repeats and immunoglobulin-like domains 1 (LRIG1) are an inhibitor of receptor tyrosine kinases (RTKs) that was discovered in recent years, and many studies showed that LRIG1 is a tumor suppressor gene and may be related to tumor drug resistance. In this study, we explored whether LRIG1 protein expression can improve the chemosensitivity of glioma cells and what was its mechanism. Materials and Methods: We collected 93 cases of glioma tissues and detected the expression of LRIG1 and BCL-2. We constructed a multidrugresistance cell line U251/multidrug resistance (MDR) and examined the change of LRIG1 and BCL-2 at mRNA and protein expression levels. LRIG1 expressionwas upregulated in U251/MDR cells and we detected the change of multidrug resistance. Meanwhile, we changed the expression of LRIG1 and BCL-2 and explored the relationship between LRIG1 and BCL-2. Finally, we also explored the relationship between LRIG1 and RTKs. Results: LRIG1 was negatively correlated with BCL-2 expression in glioma tissue and U251/MDR cells, and upregulation of LRIG1 can enhance chemosensitivity and inhibit BCL-2 expression. Furthermore, LRIG1 was negatively correlated with RTKs in U251/MDR cells. Conclusion: These results demonstrated that LRIG1 can improvechemosensitivity by modulating BCL-2 expression and RTK signaling in glioma cells.
