<P>Ticlopidine is a first-generation thienopyridine antiplatelet drug that prevents adenosine 5′-diphosphate (ADP)-induced platelet aggregation. We identified the enzymes responsible for the two-step metabolic bioactivation of ticlopidine in human liver microsomes and plasma. Formation of 2-oxo-ticlopidine, an intermediate metabolite, was NADPH dependent and cytochrome P450 (CYP) 1A2, 2B6, 2C19, and 2D6 were involved in this reaction. Conversion of 2-oxo-ticlopidine to thiol metabolites was observed in both microsomes (M1 and M2) and plasma (M1). These two metabolites were considered as isomers, and mass spectral analysis suggested that M2 was a thiol metabolite bearing an exocyclic double bond, whereas M1 was an isomer in which the double bond was migrated to an endocyclic position in the piperidine ring. The conversion of 2-oxo-ticlopidine to M1 in plasma was significantly increased by the addition of 1 mM CaCl<SUB>2</SUB>. In contrast, the activity in microsomes was not changed in the presence of CaCl<SUB>2</SUB>. M1 formation in plasma was inhibited by EDTA but not by other esterase inhibitors, whereas this activity in microsomes was substantially inhibited by carboxylesterase (CES) inhibitors such as bis-(<I>p</I>-nitrophenyl)phosphate (BNPP), diisopropylphosphorofluoride (DFP), and clopidogrel. The conversion of 2-oxo-ticlopidine to M1 was further confirmed with recombinant paraoxonase 1 (PON1) and CES1. However, M2 was detected only in NADPH-dependent microsomal incubation, and multiple CYP isoforms were involved in M2 formation with highest contribution of CYP2B6. In vitro platelet aggregation assay demonstrated that M2 was pharmacologically active. These results collectively indicated that the formation of M2 was mediated by CYP isoforms whereas M1, an isomer of M2, was generated either by human PON1 in plasma or by CES1 in the human liver.</P>

In this study, dehumidification experiments of curve-patterned copper plates having roughened morphology by blasting and wire-metallizing were assessed. To fabricate specimen with sine-wave pattern, the pattern was fabricated using plastic deformation by RP mold with sine wave (amplitude: 300㎛). Also, aluminum oxide powder (75㎛) was used in the blasting process and copper wire was used in the wire-metallizing process. To assess the wettability of blasting and wire-metallizing specimens, blasting and wire-metallizing were performed at the flat Cu plate. Moreover, the water contact angles were observed at the roughened flat Cu plate. Measurement of the surface roughness and morphology, surface image was observed optical microscope and field emission scanning electron microscope (FESEM), respectively. To assess the dehumidification capability of the fabricated specimen, several types of control Cu plates were analyzed using lab-made dehumidification apparatus.

This study assessed the drug delivery property and mechanical strength of micro needle array (MNA). MNAs were fabricated using polydimethylsiloxane (PDMS) mold having negative morphology of commercialized acupuncture. To manufacture MNAs composed of bio-compatible polymer which is artificial or nature, polymer/herbs mixture were poured in preheated PDMS mold. Subsequently, the polymer/herbs mixture was demolded from the PDMS mold. To find the appropriate mechanical strength for each treatment site, the mechanical strength of MNAs was measured using the universal testing machine (UTM). Also, optical microscope and field emission scanning electron microscope (FE-SEM) were used to find suitable geometry and needle length of each MNAs. Moreover, these drug delivery property and efficacy of MNAs with herbs were evaluated using qualitative and quantitative methods.
Pattern 을 갖는 PDMS 의 변형률에 따른 표면 에너지 변화 실험

In this study, we have suggested a simple method using a single-axis tensile strength by mechanical methods. PDMS (sylgard184) specimens were fabricated for elongation of the specimen. To fabricate the specimens, aluminum molds and acrylic molds were designed and fabricated. And then, to measure contact angles of fabricated specimens, specimens which were stretched by tensile jig were measured using a contact angle measuring instrument. In our previous study, it was observed that the water contact angles of stretched PDMS specimens were increased with respect to tensile strain. Thus, to reveal the reason for the phenomenon, chemical composition of elongated PDMS surfaces were analyzed via XPS and the physical morphology of the elongated PDMS flat surface was observed using SEM. However, we did not analyze this phenomenon. So, we did analysis using XPS, SEM data continuously. Additionally, the patterned PDMS specimens were analyzed using contact angle, XPS, SEM.
