        Transcriptome Analysis Reveals the Putative Polyketide Synthase Gene Involved in Hispidin Biosynthesis in Sanghuangporus sanghuang

        Hispidin is an important styrylpyrone produced by Sanghuangporus sanghuang. To analyzehispidin biosynthesis in S. sanghuang, the transcriptomes of hispidin-producing and non-producingS. sanghuang were determined by Illumina sequencing. Five PKSs were identifiedusing genome annotation. Comparative analysis with the reference transcriptome showedthat two PKSs (ShPKS3 and ShPKS4) had low expression levels in four types of media. Thegene expression pattern of only ShPKS1 was consistent with the yield variation of hispidin. The combined analyses of gene expression with qPCR and hispidin detection by liquid chromatography-mass spectrometry coupled with ion-trap and time-of-flight technologies(LCMS-IT-TOF) showed that ShPKS1 was involved in hispidin biosynthesis in S. sanghuang. ShPKS1 is a partially reducing PKS gene with extra AMP and ACP domains before the KSdomain. The domain architecture of ShPKS1 was AMP-ACP-KS-AT-DH-KR-ACP-ACP. Phylogenetic analysis shows that ShPKS1 and other PKS genes from Hymenochaetaceaeform a unique monophyletic clade closely related to the clade containing Agaricales hispidinsynthase. Taken together, our data indicate that ShPKS1 is a novel PKS of S. sanghuanginvolved in hispidin biosynthesis.

        DRA0336, Another OxyR Homolog, Involved in the Antioxidation Mechanisms in Deinococcus radiodurans

        A novel OxyR (DR0615) with one conserved cysteine that senses hydrogen peroxide in Deinococcus radiodurans had been identified in our previous work. Comparative genomics revealed that D. radiodurans possesses another OxyR homolog, OxyR2 (DRA0336). In this study, we constructed the deletion mutant of oxyR2 and the double mutant of both the OxyR homologs to investigate the role of OxyR in response to oxidative stress in D. radiodurans. Deletion of oxyR2 resulted in an obviously increased sensitivity to hydrogen peroxide, and the double mutant for oxyR and oxyR2 was significantly more sensitive than any of the two single mutants. The total catalase activity of the double mutant was lower than that of any of the single mutants, and reactive oxygen species (ROS) accumulated to a greater extent. DNA microarray analysis further suggested that oxyR2 was involved in antioxidation mechanisms. Site-direct mutagenesis and complementation analysis revealed that C228 in OxyR2 was essential. This is the first report of the presence of two OxyR in one organism. These results suggest that D. radiodurans OxyR and OxyR2 function together to protect the cell against oxidative stress.

      • THE EFFECT OF ANTHROPOMORPHIC BRAND ROLES AND IMPLICIT THEORIES ON CONSUMER RESPONSES

        Recently, brand managers have widely adopted brand anthropomorphism as positioning strategies to differentiate from other competitors. Aggarwal and McGill (2012) anthropomorphized brand as two specific roles. Brand-as-partner refers its role as “the co-producer of benefits”, a relationship in which the brand and consumers work together to co-create the benefits as equals. However, brand-as-servant represents its role as “the outsourced provider of benefits”, a hierarchical relationship in which the brand works for consumers to create benefits. Our research extends prior literature by investigating how a brand can improve consumers’ responses with the anthropomorphization of being either a collaborative partner or a supportive servant, depending on consumer’s mind-set ? aptly termed implicit theories. Implicit theories are the beliefs that people have about the nature of human characteristics. In specific, entity theorists believe human traits are relatively fixed and stable; and incremental theorists believe human traits are substantially changeable (Dweck & Leggett, 1988). These two implicit theories trigger many distinctive cognitive and behavioral patterns. For example, entity theorists prefer effortless success and receiving assurances to signal their abilities. In contrast, incremental theories emphasize on self-improvement through high effort to realize their self-enhancement and intentions (Park & John, 2014). We generalize this theory to anthropomorphic brand positioning and adopt consumer’s efficacy (confidence) to explain the underlying mechanism. We hypothesize a servant brand is more appealing to entity theorists, because they prefer situations in which they are guaranteed of being satisfied effortlessly (Dweck & Leggett, 1988). Therefore, they are more confident about their abilities when a servant brand acts as an assurance for providing benefits. In contrast, a partner brand may be more attractive to incremental theorists, because they believe their abilities can be improved through working with the partner brand. Thus, they should be more self-assured about their abilities, and increase their perceived efficacy to realize their intentions. Three studies were conducted to test the hypotheses by both manipulating and measuring implicit theories. The consistent results revealed the significant interactions between brand roles and implicit theories on brand evaluation as well as the mediation effect of consumer efficacy. In sum, this research contributes to brand managers by advising them to cultivate appropriate brand role position in accordance with the target and potential consumer’s distinct mind-set.

        Ultrasonic Vibration and Rheocasting for Refinement of Mg–Zn–Y Alloy Reinforced with LPSO Structure

        In this work, ultrasonic vibration (UV) and rheo-squeeze casting was first applied on the Mg alloy reinforced with long periodstacking ordered (LPSO) structure. The semisolid slurry of Mg–Zn–Y alloy was prepared by UV and processed by rheosqueezecasting in succession. The effects of UV, Zr addition and squeeze pressure on microstructure of semisolid Mg–Zn–Yalloy were studied. The results revealed that the synergic effect of UV and Zr addition generated a finer microstructure thaneither one alone when preparing the slurries. Rheo-squeeze casting could significantly refine the LPSO structure and α-Mgmatrix in Mg96.9Zn1Y2Zr0.1alloy without changing the phase compositions or the type of LPSO structure. When the squeezepressure increased from 0 to 400 MPa, the block LPSO structure was completely eliminated and the average thickness ofLPSO structure decreased from 9.8 to 4.3 μm. Under 400 MPa squeeze pressure, the tensile strength and elongation of therheocast Mg96.9Zn1Y2Zr0.1alloy reached the maximum values, which were 234 MPa and 17.6%, respectively, due to its fineα-Mg matrix (α1-Mg and α2-Mg grains) and LPSO structure.
