        Fast FCS-MPC-Based SVPWM Method to Reduce Switching States of Multilevel Cascaded H-Bridge STATCOMs

        Finite control set model-predictive control (FCS-MPC) has received increasing attentions due to its outstanding dynamicperformance. It is being widely used in power converters and multilevel inverters. However, FCS-MPC requires a lot ofcalculations, especially for multilevel-cascaded H-bridge (CHB) static synchronous compensators (STATCOMs), since it has totake account of all the feasible voltage vectors of inverters. Hence, an improved five-segment space vector pulse widthmodulation (SVPWM) method based on the non-orthogonal static reference frames is proposed. The proposed SVPWM methodhas a lower number of switching states and requires fewer computations than the conventional method. As a result, it makesFCS-MPC more efficient for multilevel cascaded H-bridge STATCOMs. The partial cost function is adopted to sequentiallysolve for the reference current and capacitor voltage. The proposed FCS-MPC method can reduce the calculation burden of theFCS-MPC strategy, and reduce both the switching frequency and power losses. Simulation and experimental results validate theexcellent performance of the proposed method when compared with the conventional approach.

        Analysis of ESTs from a Normalized cDNA Library of the Rhizome Tip of Oryza longistaminata

        Oryza longistaminata, a perennial wild rice species with an AA genome, is characterized by the presence of rhizomatous stems. The rhizomatous trait in rice was previously shown to be quantitatively controlled by many genes, but the molecular mechanism related to rhizome development is still unknown. In the present study, expressed sequence tags (ESTs) generated from rhizome tips of O. longistaminata were collected and analyzed. A total of 10,283 complimentary deoxyribunucleic acid clones were randomly sequenced, which generated 10,136 raw sequences,and finally, 4,419 unisequences with diverse functional categories were generated. These unisequences were mapped onto the Oryza sativa genome, which revealed that 4,285(96.97%) and 4,151 (93.94%) of the unisequences were alignable to the japonica and indica genomic sequences,respectively, with >80% sequence identity. Additionally, 41unisequences showed four typical types of alternative splicing patterns. More than 600 simple sequence repeats were identified in these unisequences. A subset of unisequences were physically colocalized onto rhizome-related quantitative trait locus intervals in rice and sorghum, and one gene,OLRR1, was further confirmed to be enriched in the rhizome tip and young leaf by real-time polymerase chain reaction and in situ hybridization. Unisequences reported in this study provide valuable data for molecular dissection of the rhizomatous growth habit in O. longistaminata.

        Tissue-Specific CRISPR/Cas9 System of Cotton Pollen with GhPLIMP2b and GhMYB24 Promoters

        CRISPR/Cas9 technology is a powerful tool for improving crop genetic traits. However, CRISPR/Cas9 system for efficient expression of cotton germ cells has still not been established. In this study, we developed a tissue-specific vectors to drive Cas9 expression with GhPLIMP2b and GhMYB24 promoters and established the effective method to transform cotton pollen by Agrobacterium vacuum infiltration. GhPLIMP2b and GhMYB24 promoters of cotton pollen were cloned into Cas9 expression vectors. The sgRNAs targetting to CLA1, ERA1 and GGB (drought-resistant negative regulation genes) were designed and constructed into GhPLIMP2b and GhMYB24 promoter vectors. Cotton pollens were tranformed by Agrobacterium vacuum infiltration with GhPLIMP2b and GhMYB24 promoter vectors. The results of clone sequencing shown that mutation types of the sequence were mainly base substitutions wth the frequency from 3.29 to 6.45%. Eleven potential off-target sites were chosen and two sites were observed. Our results indicated that GhPLIM2bP::Cas9 and GhMYB24P::Cas9 editing vectors achieved targeted edition of endogenous genes in cotton pollen, but there were a few off-target effects. This study provides an effective gene-editing system for the rapid acquisition of cotton mutants using pollen as a transgenic receptor.

        The application of modified isochronal well test in a low-permeability condensate gas field

        The modified isochronal well test is a deliverability test method suitable for gas wells in low-permeability gas reservoirs. Generally, the absolute open flow of gas wells is determined by modified isochronal well test, thereby establishing a reasonable working system for gas wells. It is also possible to obtain gas layer physical parameters, gas reservoir engineering parameters, heterogeneity, boundary characteristics, etc. In this paper, the actual test data of a condensate gas field are applied. By modified isochronal well test analysis, not only the deliverability equation of the gas well is obtained, but also the absolute open flow is determined. At the same time, the formation parameters such as reservoir permeability and skin factor are obtained and the acidification effect is evaluated.
