Developmental Potential of Rat Extraembryonic Stem Cells

<P>We have previously found that certain stem cells that are derived from rat blastocysts and named extraembryonic endoderm precursor (XEN-P) cells show a unique molecular signature sharing some of the characteristics of embryonic stem cells (ES), trophoblast stem cells (TS), and extraembryonic endoderm stem cells (XEN). These XEN-P cells are positive for AP, SSEA1, Oct4, and Rex1 markers similar to ES cells and also express signature markers of TS-eomesodermin (Eomes) and XEN-Gata6. Here we show that these cells integrate into the visceral and parietal extraembryonic endoderm lineages as well as into the inner cell mass (ICM), the primitive endoderm, and the polar and mural trophectoderm (TE) of cultured embryos. In addition, we find that the XEN-P cells colonize yolk sac and contribute to trophoblast lineages of postimplantation embryos following transfer to surrogate mothers. We also find that the XEN-P cell culture propagates by shedding cell clusters into the media in addition to typical expansion of colonies. Interestingly, the cell cultures exist as mixed populations of two interconvertible phenotypes of flat and round cells with preferential expression of stem cell markers Oct4 and SSEA1 in round cells. We believe these cells represent a metastable stage during ICM cellular segregation. These results are important for developing hypotheses of cell fate plasticity in the ICM and provide a model for the study of development and differentiation along the extraembryonic lineages.</P>

Objective: Aim of present study was the set up of a fast and reliable protocol using species-specific markers for the quali-quantitative analysis of DNA and the detection of ruminant biological components in dairy products. For this purpose, the promoter of the gene coding for the α-lactoalbumin (LALBA) was chosen as possible candidate for the presence of short interspersed nuclear elements (SINEs). Methods: DNA was isolated from somatic cells of 120 individual milk samples of cattle (30), Mediterranean river buffalo (30), goat (30), and sheep (30) and the gene promoter region (about 600/700 bp) of LALBA (from about 600 bp upstream of exon 1) has been sequenced. For the development of a single polymerase chain reaction (PCR) protocol that allows the simultaneous identification of DNA from the four species of ruminants, the following internal primers pair were used: 5’-CACTGATCTTAAAGCTCAGGTT-3’ (forward) and 5’-TCAGA GTAGGCCACAGAAG-3’ (reverse). Results: Sequencing results of LALBA gene promoter region confirmed the presence of SINEs as monomorphic “within” and variable in size “among” the selected species. Amplicon lengths were 582 bp in cattle, 592 bp in buffalo, 655 in goat and 729 bp in sheep. PCR specificity was demonstrated by the detection of trace amounts of species-specific DNA from mixed sources (0.25 ng/μL). Conclusion: We developed a rapid PCR protocol for the quali-quantitative analysis of DNA and the traceability of dairy products using a species-specific marker with only one pair of primers. Our results validate the proposed technique as a suitable tool for a simple and inexpensive (economic) detection of animal origin components in foodstuffs.
A Case for Using Service Availability to Characterize IP Backbone Topologies

Traditional service-level agreements (SLAs), defined by average delay or packet loss, often camouflage the instantaneous performance perceived by end-users. We define a set of metrics for service availability to quantify the performance of Internet protocol (IP) backbone networks and capture the impact of routing dynamics on packet forwarding. Given a network topology and its link weights, we propose a novel technique to compute the associated service availability by taking into account transient routing dynamics and operational conditions, such as border gateway protocol (BGP) table size and traffic distributions. Even though there are numerous models for characterizing topologies, none of them provide insights on the expected performance perceived by end customers. Our simulations show that the amount of service disruption experienced by similar networks (i.e., with similar intrinsic properties such as average out-degree or network diameter) could be significantly different, making it imperative to use new metrics for characterizing networks. In the second part of the paper, we derive goodness factors based on service availability viewed from three perspectives: Ingress node (from one node to many destinations), link (traffic traversing a link), and network-wide (across all source-destination pairs). We show how goodness factors can be used in various applications and describe our numerical results.
