        분산형 집단에너지 정책변동 연구: 에너지 데이터 기반의 국가 열지도 사업을 중심으로

        전 세계적인 에너지·기후 위기는 각국의 이해관계가 복잡하게 얽혀있어 글로벌 대응이 필수적인 아젠다임이 최근 다시금 환기되고 있다. 특히, 우리나라는 에너지 수입의존도가 높고 에너지 다소비·저효율 소비 및 온실가스 多 배출구조가 지속되고 있어 에너지효율 및 탄소중립 달성을 위한 혁신적이고 실효성 있는 에너지정책이 절실하다고 하겠다. 본 연구에서는 우리나라에 1980년대 중반 집단에너지 방식이 도입된 이후의 분산형 집단에너지 정책변동 중 “국가 열(熱) 지도 사업” 정책 추진사례를 수정 다중흐름모형으로 분석하고자 한다. 연구 대상 기간인 이명박-박근혜 정부의 10년간은 에너지정책의 주된 패러다임이 “분산형 에너지플랫폼”으로 전환하는 데 있어 정책의제 설정과 정책 표류, 정책수정의 전환이 이루어졌던 기간으로서 연구에 의미가 있을 것이다. As the global energy and climate crisis has complicated interests of each country, the agenda that requires a global response has recently been revived. In particular, Korea is highly dependent on energy imports and continues to have high energy consumption, low efficiency of energy consumption, and high greenhouse gas emissions, so innovative and effective energy policies are urgently needed to achieve energy efficiency and carbon neutrality. In this study, among the changes in distributed district energy policy after the integrated energy method was introduced in Korea in the mid-1980’s, the case of the “National Heat Map Project” policy implementation is analyzed with a modified multi-flow model. The 10 years of the Lee Myung-bak and Park Geun-hye administrations, the period of study, was a period in which the main paradigm of energy policy shifted to a “distributed energy platform” and policy transitions such as policy agenda setting, policy drift, and policy revision were made. A study on the process would be meaningful.

      • Molecular cloning and characterization of soybean (Glycine max) NAC transcription factor genes induced by abiotic stress

        The plant-specific NAC (NAM, ATAF, and CUC)-domain proteins play important roles in plant development and stress responses. Comparative time-course expression analyses were carried out to analyze the expression levels of 62 soybean NAC genes during drought stress in order to search for the stress-inducible NAC genes. Ten GmSNAC (Glycine max stress-inducible NAC) genes having the significant differential expression in response to the drought stress and abscisic acid (ABA) hormone application were further investigated for their expression profiles with various stresses such as drought, high salinity, cold and with ABA treatments by the quantitative real-time PCR analyses. In this research, the full-length cDNAs of eight GmSNAC were isolated for the further studies. Eight GmSNAC proteins were tested for their transcription activation in the yeast assay system. Two GmSNAC proteins showed the very high transcriptional activities and the other two GmSNAC proteins displayed moderate levels of transactivation while the remaining four GmSNAC proteins lacked transactivation in yeast. Subcellular localization of eight GmSNAC proteins was analyzed via the green fluorescent protein-GmSNAC fusion protein in tobacco plant cell. Three GmSNAC proteins with the C-terminal transmembrane domain were localized to the nucleus and cytoplasmic fractions. The other five GmSNAC proteins were targeted to the nucleus. The function of GmSNAC49 gene was further investigated using the overexpression transgenic Arabidopsis. Germination rate in transgenic plants over-expressing GmSNAC49 was delayed in the media supplemented with mannitol or ABA compared with that of wild-type (WT) plants. The 35S:GmSNAC49 transgenic Arabidopsis displayed improved tolerance to drought stress compared to the WT. The results of this systematic analysis of the GmSNAC family responsive to abiotic stress will provide novel tools and resources for the development of improved drought tolerant transgenic soybean cultivars

      • Estrogen receptor-related receptor γ regulates testicular steroidogenesis through direct and indirect regulation of steroidogenic gene expression

        <P><B>Abstract</B></P> <P>Biosynthesis of testosterone, which mainly occurs in testicular Leydig cells, is controlled by steroidogenic proteins, such as StAR and P450c17. Although estrogen-related receptor gamma (ERRγ), an orphan nuclear receptor, is expressed in the testis, its role is not well understood. In this study, we investigated the expression of ERRγ in Leydig cells and its molecular action on testicular steroidogenesis. ERRγ is expressed in mouse Leydig cells from pre-pubertal stages. ERRγ overexpression in primary Leydig cells elevated the production of testosterone with a marked increase of P450c17 expression at both mRNA and protein levels, albeit decreased expression of StAR. Promoter-reporter analyses showed that ERRγ directly regulated the P450c17 promoter. Further deletion mutant analyses of the P450c17 promoter revealed that ERRγ activated expression of the P450c17 gene by binding to an ERRγ response element within the P450c17 promoter. Meanwhile, ERRγ suppressed cAMP-induced activation of the StAR promoter, which was likely due to ERRγ-mediated inhibition of the transcriptional activity of Nur77, which is induced by cAMP and regulates StAR gene expression in Leydig cells. Interestingly, ERRγ coexpression also decreased the protein level of Nur77, which occurred through proteasomal degradation, suggesting ERRγ-mediated regulation of steroidogenesis at another level. Taken together, these findings suggest that ERRγ regulates testicular steroidogenesis, both directly controlling and indirectly fine-tuning the expression of steroidogenic genes.</P> <P><B>Highlights</B></P> <P> <UL> <LI> ERRγ elevates testosterone production in testicular Leydig cells. </LI> <LI> ERRγ directly regulates the P450c17 promoter. </LI> <LI> ERRγ inhibits Nur77 transactivation and reduces the Nur77 protein stability; thus regulates cAMP/Nur77 regulated promoters. </LI> </UL> </P>

        NUP50 is necessary for the survival of primordial germ cells in mouse embryos

        <P>Nucleoporin 50 kDa (NUP50), a component of the nuclear pore complex, is highly expressed in male germ cells, but its role in germ cells is largely unknown. In this study, we analyzed the expression and function of NUP50 during the embryonic development of germ cells using NUP50-deficient mice. NUP50 was expressed in germ cells of both sexes at embryonic day 15.5 (E15.5), E13.5, and E12.5. In addition, NUP50 expression was also detected in primordial germ cells (PGCs) migrating into the genital ridges at E9.5. The gonads of <I>Nup50</I><I>−</I><I>/</I><I>−</I> embryos of both sexes contained few PGCs at both E11.5 and E12.5 and no developing germ cells at E15.5. The migratory PGCs in <I>Nup50</I><I>−</I><I>/</I><I>−</I> embryos at E9.5 showed increased apoptosis but a normal rate of proliferation, resulting in the progressive loss of germ cells at later stages. Taken together, these results suggest that NUP50 plays an essential role in the survival of PGCs during embryonic development.</P>

      • Ectopic expression of mungbean ubiquitin conjugating enzyme E2 enhances resistance to osmotic stress in Arabidopsis

        The ubiquitin conjugating enzyme E2 (UBC E2) mediates selective ubiquitination, acting with E1 and E3 enzymes to designate specific proteins for subsequent degradation. In the present study, we characterized the function of the mung bean VrUBC1 gene (Vigna radiata UBC 1). RNA gel-blot analysis showed that VrUBC1 mRNA expression was induced by either dehydration, high salinity or by the exogenous abscisic acid (ABA), but not by low temperature or wounding. Biochemical studies of VrUBC1 recombinant protein and complementation of yeast ubc4/5 by VrUBC1 revealed that VrUBC1 encodes a functional UBC E2. To understand the function of this gene in development and plant responses to osmotic stresses, we overexpressed VrUBC1 in Arabidopsis (Arabidopsis thaliana). The VrUBC1-overexpressing plants displayed highly sensitive responses to ABA and osmotic stress during germination, enhanced ABA- or salt-induced stomatal closing, and increased drought stress tolerance. The expression levels of a number of key ABA signaling genes were increased in VrUBC1-overexpressing plants compared to the wild-type plants. Yeast two-hybrid and bimolecular fluorescence complementation demonstrated that VrUBC1 interacts with AtVBP1 (A. thaliana VrUBC1 Binding Partner 1), a C3HC4-type RING E3 ligase. Overall, these results demonstrate that VrUBC1 plays a positive role in osmotic stress tolerance through transcriptional regulation of ABA-related genes and possibly through interaction with a novel RING E3 ligase.

        Differential regulation of steroidogenic enzyme genes by TRα signaling in testicular Leydig cells.

        <P>Thyroid hormone signaling has long been implicated in mammalian testicular function, affecting steroidogenesis in testicular Leydig cells. However, its molecular mechanism is not well understood. Here, we investigated the molecular action of thyroid hormone receptor-α (TRα) on mouse testicular steroidogenesis. TRα/thyroid hormone (T3) signaling differentially affected the expression of steroidogenic enzyme genes, mainly regulating their promoter activity. TRα directly regulated the promoter activity of the cytochrome P450 17α-hydroxylase/C17-20 lyase gene, elevating its expression in the presence of T3. TRα also indirectly regulated the expression of steroidogenic enzyme genes, such as steroidogenic acute regulatory protein and 3β-hydroxysteroid dehydrogenase, by modulating the transactivation of Nur77 on steroidogenic enzyme gene promoters through protein-protein interaction. TRα enhanced Nur77 transactivation by excluding histone deacetylases from Nur77 in the absence of T3, whereas liganded TRα inhibited Nur77 transactivation, likely due to interfering with the recruitment of coactivator such as the steroid receptor coactivator-1 to Nur77. Together, these findings suggest a role of TRα/T3 in testicular steroidogenesis and may provide molecular mechanisms for the differential regulation of steroidogenic enzyme genes by thyroid hormone.</P>

      • Inhibitory Effect of Yongdamsagan-Tang Water Extract, a Traditional Herbal Formula, on Testosterone-Induced Benign Prostatic Hyperplasia in Rats

        <P>Yongdamsagan-tang, a traditional herbal formula, is used widely for the treatment of inflammation and viral diseases. In this study, we investigated whether Yongdamsagan-tang water extract (YSTE) affects testosterone propionate- (TP-) induced benign prostatic hyperplasia (BPH) in a rat model. To induce BPH, rats were injected subcutaneously with 10 mg/kg of TP every day. YSTE was administrated daily by oral gavage at doses of 200 and 500 mg/kg along with the TP injection. After 4 weeks, prostates were collected, weighed, and analyzed. The relative prostrate weight was significantly lower in both YSTE groups (200 and 500 mg/kg/day) compared with the TP-induced BPH group. YSTE administration reduced the expression of proliferation markers PCNA, cyclin D1, and Ki-67 and the histological abnormalities observed in the prostate in TP-induced BPH rats. YSTE attenuated the increase in the TP-induced androgen concentration in the prostate. The YSTE groups also showed decreased lipid peroxidation and increased glutathione reductase activity in the prostate. These findings suggest that YSTE effectively prevented the development of TP-induced BPH in rats through antiproliferative and antioxidative activities and might be useful in the clinical treatment of BPH.</P>

        A Study on the Development of a Blended Learning Model : Based on Brummelen’s Biblical Model for Learning

        The purpose of this research is to make a blended learning model in order to supply instructional learning methods and strategies for diverse learners who prefer different learning styles based on Brummelen’s learning model. This model introduces the Biblical view on diverse learners and instructional learning methods. The research will be applied to college courses to find out the effectiveness of the model. For this, the research explores Brummelen’s learning model based on Bernice and McCarthy’s research on learning styles. Students’ diversity is emphasized through a Biblical view and the four steps of the learning model, which was designed to draw out diverse learners’ personal responses from their learning with a Christian perspective. This research also explores the concept and characteristics of Felder and Silverman’s learning styles in order to give understanding about diverse learners and different learning methods to suit each learner’s learning style.
