      • c-Cbl-Mediated Neddylation Antagonizes Ubiquitination and Degradation of the TGF-β Type II Receptor

        Transforming growth factor β (TGF-β) is a potent antiproliferative factor in multiple types of cells. Deregulation of TGF-β signaling is associated with the development of many cancers, including leukemia, though the molecular mechanisms are largely unclear. Here, we show that Casitas B-lineage lymphoma (c-Cbl), a known proto-oncogene encoding an ubiquitin E3 ligase, promotes TGF-β signaling by neddylating and stabilizing the type II receptor (TβRII). Knockout of c-Cbl decreases the TβRII protein level and desensitizes hematopoietic stem or progenitor cells to TGF-β stimulation, while c-Cbl overexpression stabilizes TβRII and sensitizes leukemia cells to TGF-β. c-Cbl conjugates neural precursor cell-expressed, developmentally downregulated 8 (NEDD8), a ubiquitin-like protein, to TβRII at Lys556 and Lys567. Neddylation of TβRII promotes its endocytosis to EEA1-positive early endosomes while preventing its endocytosis to caveolin-positive compartments, therefore inhibiting TβRII ubiquitination and degradation. We have also identified a neddylation-activity-defective c-Cbl mutation from leukemia patients, implying a link between aberrant TβRII neddylation and leukemia development.

        The Expression of Porcine Adiponectin and Stearoyl Coenzyme a Desaturase Genes in Differentiating Adipocytes

        The gene expression of porcine adiponectin and stearoyl coenzyme A desaturase (SCD) was investigated in this study. The partial gene sequences for adiponectin and SCD were amplified by RT-PCR from subcutaneous adipose tissue and cloned by TA cloning techniques. Sequences of these genes were determined and found to be highly homologous to that of other species, suggesting similar function of these genes as in other species. The transcripts of these adipocyte-related genes in pig tissues were measured by Northern analysis. The transcripts for adiponectin and SCD were highly expressed in porcine subcutaneous adipose tissue; the transcripts for SCD were also barely detected in the liver, but the greatest concentrations were in the adipose tissue. In porcine stromalvascular cells (S/V cells) cultured in vitro, transcripts for adiponectin and SCD increased gradually during adipocyte differentiation. The level of adipocyte adiponectin mRNA was associated with late adipocyte differentiation, indicating the gene may not be involved in adipocyte differentiation but has great importance in porcine adipocyte functions. The SCD transcripts were not detectable until 2 d after induction of adipocyte differentiation. It was highly expressed in differentiating porcine adipocytes (2 to 10 d after the induction of adipocyte differentiation), indicating a significant role of SCD in adipocytes.

        The Expression of Genes Related to Egg Production in the Liver of Taiwan Country Chickens

        The purpose of this study was to detect expression of genes related to egg production in Taiwan Country chickens by suppression subtractive hybridization. Liver samples of mRNA extraction from two Taiwan Country chicken strains (L2 and B), originated from the same population but with very distinct egg production rates after long-term selection for egg and meat production respectively. Two-way subtraction was performed. The hepatic cDNA from the low egg production chickens (B) was subtracted from the hepatic cDNA from the high egg production strain (L2). The reversed subtraction (L2 from B) was also performed. The resulting differentially expressed gene fragments were cloned and sequenced. We sequenced 288 clones from the forward subtraction and 96 clones from the reverse subtraction. These genes were subjected to further screening to confirm the differential expression between the two genetic breeds of chickens. The apolipoprotein B (apoB) was expressed to a greater extent in the liver of the L2 than in the B line chickens. The 5-aminoimidazole- 4-carboxamide ribonucleotide formyltransferase/IMP cyclohydrolase (PURH) was expressed to a greater extent in the liver of the B than in the L2 strain chickens. We demonstrated that both apoB and PURH were more highly expressed in the liver than that in other tissues (muscle, ovary, and oviduct) in laying Taiwan Country chickens. Taken together, these data suggest that after the selection for egg production, expression of apoB and PURH genes were also changed. Whether the changed expression of these genes is directly related to egg production is not known, but these two genes may be useful markers for egg laying performance in Taiwan Country chickens.

        THE EFFECT OF DIETARY MAGNESIUM LEVEL ON THE MAGNESIUM METABOLIZABILITY IN LAYING TSAIYA DUCK AND LEG HORN HEN

        The study was undertaken to determine the effect of various levels of dietary magnesium on the metabolizability of magnesium in laying Tsaiya ducks and Leghorn hens. Twenty-five Tsaiya ducks and 25 Leghorn hens were raised in individual cages. The basal diet was mainly consisted of corn starch and isolated soybean protein. At the beginning of the experiments, birds were fed the basal diet added with 500 mg/kg Mg (1070 mg/kg in total by analysis) for 10 days in order to make the birds adapt to the new diet. Then, they were randomly divided into five treatments with five birds for each treatment and fed the experimental diets containing 690, 1070, 1690, 2150 or 2380 mg/kg Mg for 21 days and during the last five days excreta were collected. The experiment was repeated three times each at the age of 25, 31 and 36 weeks. The metabolizability of magnesium in the diet was determined by the indicator method. The metabolizabilities of magnesium for Tsaiya duck and Leghorn hen were 59.9% and 62.5%, respectively as the diet contained 690 mg/kg magnesium, and were significantly decreased (p<0.05) to 39.7 and 30.7%, respectively when the dietary magnesium content was increased to 2380 mg/kg.

        THE EFFECT OF DIETARY MAGNESIUM LEVEL ON THE EGGSHELL QUALITY IN LAYING TSAIYA DUCK AND LEGHORN HEN

        The study was undertaken to determine the effect of dietary magnesium on the eggshell quality and other performance in laying Tsaiya ducks and Leghorn hens. Twenty-five Tsaiya ducks and 25 Leghorn hens were raised in individual cages. The basal diet was mainly consisted of corn starch and isolated soybean protein. At the beginning of the experiments, birds were fed for 10 days with the basal diet supplemented with 500 mg/kg Mg (1070 mg/kg in total by analysis) in order to allow the birds adapting to the new diet. Both Tsaiya ducks and Leghorn hens were then each randomly divided into five groups and each group of five birds were fed with the experimental diets containing 690, 1070, 1690, 2150 or 2380 mg/kg Mg, respectively for 21 days. Eggs were collected in order to measure eggshell quality, Mg and Ca content of the eggshell. At the end of the experiments, blood samples of all birds were taken from their brachial veins for measuring the concentration of Mg and Ca in the plasma. Experimental results appeared that the dietary Mg content did not significantly affect egg production, egg weight, eggshell breaking strength and thickness in both Tsaiya ducks and Leghorn hens. In Tsaiya ducks, however, the plasma Mg concentration was affected by the dietary Mg content, but the plasma Mg almost reached a plateau (4.66 mg/dl) as long as the dietary Mg level was over 1070 mg/kg. In Leghorn hens, the plasma Mg level was significantly increased from 1.66 mg/dl to 4.03 mg/dl when the dietary Mg content in the diet had been increased from 690 mg/kg to 2380 mg/kg, suggesting that the plasma Mg concentration seems to be directly influenced by the Mg absorbed in the intestine. In the Tsaiya ducks, however, the dietary Mg level did not significantly affect the eggshell Mg content (from 0.113% to 0.123%). Whereas, there was a negative correlation between the eggshell thickness and eggshell Mg content (r = -0.50, p<0.01), revealing that the increase in eggshell Mg content probably associated with the impairment of eggshell quality in Tsaiya ducks. In Leghorn hens, however, there was no significant correlation between eggshell quality and eggshell Mg content, although the Mg content in the eggshell was also increased from 0.279% to 0.427% when the dietary Mg had been elevated from 690 mg/kg to 2380 mg/kg.

        Estimation of growth parameters of Listeria monocytogenes after sublethal heat and slightly acidic electrolyzed water (SAEW) treatment

        <P>Time to detection experiments (TTD) based on turbidometry using an automatic Bioscreen C is a useful and straightforward method for estimating microbial growth parameters (lag time (lambda), growth rate (mu) and 'work to be done' (h(0))) at constant temperature. This study investigated the effects of slightly acidic electrolyzed water (SAEW) and heat treatment on Listeria monocytogenes growth at different recovery temperatures (10 degrees C, 15 degrees C, 25 degrees C, and 30 degrees C). Similar surviving and sublethally injured L. monocytogenes populations were obtained by heat treatment (55 degrees C for 10 min) and SAEW treatment (available chlorine concentration of 30 mg/l and ratio of bacteria against SAEW of 8:2 for 30 s). In these experimental conditions, stresses had greater impact on the lambda and h(0) parameter in comparison with recovery temperature while there was no great change in growth rate under isothermal conditions. Larger lambda values and h(0) parameters were observed in sublethal-heat injured L. monocytogenes (the maximum lambda and h(0) parameters are 30.199 h and 1.6492) as compared to SAEW groups (the maximum lambda and h(0) parameters are 22.634 h and 1.4396). The sensitivity analysis of SAEW and heat treatments on h(0) parameter indicated that SAEW treatment showed a higher influence. The collinearity diagnostics of independent variables [recovery temperature (T), mu, lambda] for dependent variable (h(0) parameter) demonstrated that T, mu and lambda had strong collinearity. In addition, the established secondary models in this study have good performances on predicting the effect of recovery temperature on bacterial growth parameters. (C) 2016 Elsevier Ltd.         Modelling of Escherichia coli O157:H7 growth at various storage temperatures on beef treated with electrolyzed oxidizing water

        The influence of storage temperature (4, 10, 15, 20, 25, and 30<SUP>o</SUP>C) on the growth of Escherichia coli O157:H7 in beef untreated (control) and treated by acidic electrolyzed oxidizing water (AcEOW) or slightly acidic electrolyzed oxidizing water (SAcEOW) was examined. A Baranyi model was employed to describe growth parameters such as specific growth rate (SGR) and lag time (LT) as a function of storage temperature. SGR increased and LT declined with rising temperatures in all samples. There were no significant differences between the SGR and LT values obtained from beef treated with AcEOW or SAcEOW. Secondary models were established for SGR and LT to evaluate the effects of storage temperature on the growth kinetics of E. coli O157:H7 in treated and untreated beef. Mathematical evaluation was carried out to validate the performance of the developed models.

        Inhibitory effects of low concentration electrolyzed water and other sanitizers against foodborne pathogens on oyster mushroom

        In this study we investigated the effects of low concentration electrolyzed water (LcEW) and several other sanitizers (strong acid electrolyzed water (SAEW), aqueous ozone (AO), 1% citric acid (CA) and sodium hypochlorite solution (NaOCl)) on the inactivation of natural microflora (total aerobic bacteria counts (TBC) and yeasts and moulds (YM)) and foodborne pathogens (Escherichia coli O157:H7, Listeria monocytogenes, Salmonella Typhimurium and Bacillus cereus) on oyster mushroom. The effects of temperature and treatment time on the antimicrobial activity of LcEW to reduce the populations of foodborne pathogens were also determined. LcEW showed the strongest bactericidal efficacy among all the sanitizers on TBC, YM and pathogens by reductions of 1.35, 1.08 and 1.90-2.16 log CFU/g after 3 min treatment at room temperature (23 +/- 2 <SUP>o</SUP>C), respectively. There was no significant difference between the antimicrobial effects of LcEW and SAEW (P > 0.05). Among those sanitizers, their relative influence of inactivation was LcEW > NaOCl > CA > AO.
