        Cloning and functional characterizations of an apoptogenic Hid gene in the Scuttle Fly, Megaselia scalaris (Diptera; Phoridae)

        Although the mechanisms of apoptotic cell death have been well studied in the fruit fly, Drosophila melanogaster, it is unclear whether such mechanisms are conserved in other distantly related species. Using degenerate primers and PCR, we cloned a proapoptotic gene homologous to Head involution defective (Hid) from the Scuttle fly, Megaselia scalaris (MsHid). MsHid cDNA encodes a 197-amino acid-long polypeptide, which so far is the smallest HID protein. PCR analyses revealed that the MsHid gene consists of four exons and three introns. Ectopic expression of MsHid in various peptidergic neurons and non-neuronal tissues in Drosophila effectively induced apoptosis of these cells. However, deletion of either conserved domain, N-terminal IBM or C-terminal MTS, abolished the apoptogenic activity of MsHID, indicating that these two domains are indispensable. Expression of MsHid was found in all life stages, but more prominently in embryos and pupae. MsHid is actively expressed in the central nervous system (CNS), indicating its important role in CNS development. Together MsHID is likely to be an important cell death inducer during embryonic and post-embryonic development in this species. In addition, we found 2-fold induction of MsHid expression in UV-irradiated embryos, indicating a possible role for MsHid in UV-induced apoptosis.

        Quantum Packet for the Next Generation Network/ISDN3

        This paper proposes a novel method for transporting various types of user traffic effectively over the next generation network called integrated services digital network 3 (ISDN3) (or quantum network) using quantum packets. Basically, a quantum packet comprises one or more 53-byte quanta as generated by a "quantumization" process. While connection-oriented traffic is supported by fixed-size quantum packets each with one quantum to emulate circuit switching, connectionless traffic (e.g., IP packets and active packets) is carried by variable-size quantum packets with multiple quanta to support store-and-forward switching/routing. Our aim is to provide frame-like or datagram-like services while enabling cell-based multiplexing. The quantum packet method also establishes a flexible and extensible framework that caters for future packetization needs while maintaining backward compatibility with ATM. In this paper, we discuss the design of the quantum packet method, including its format, the "quantumization" process, and support for different types of user traffic. We also present an analytical model to evaluate the consumption of network resources (or network costs) when quantum packets are employed to transfer loss-sensitive data using three different approaches: cut-through, store-and-forward and ideal. Close form mathematical expressions are obtained for some situations. In particular, in terms of network cost, we discover two interesting equivalence phenomena for the cut-through and store-and-forward approaches under certain conditions and assumptions. Furthermore, analytical and simulation results are presented to study the system behavior. Our analysis provides valuable insights into the. design of the ISDN3/quantum network.

        Physical and Physiological Dormancy in Black Henbane (Hyoscyamus niger L.) Seeds

        Aim of this study was to investigate the nature of dormancy in black henbane (Hyoscyamus niger) seeds which have low germination rate under normal laboratory conditions. To do this, before placing the seeds in Petri dishes, they were soaked in 5, 10 and 15 ㎎/L GA; 1, 2 and 3% H₂SO₄,15 ㎎/L GA + 1% H₂SO₄, 0.01 M KNO₃ solutions, tap water, 40, 50 and 60°C hot water for 30 min. The study was performed under both continuous illumination and darkness in growth chambers to evaluate the effect of light on germination rate. The results showed that H₂SO₄ and GA treatments were the most important factors affecting seed germination and their germination enhancing effects were more evident in darkness. The results also suggested that black henbane seeds exhibit double dormancy involving a hard seed coat and a partially dormant embryo and have a partial dark requirement to germinate.

        The Local Web Buckling Strength of Stiffened Coped Steel I-Beams

        In steel construction the flange of a steel I-beam is usualy coped to allow clearance at the connection. The presence of acope in a beam wil reduce the strength of the beam in the coped region. Local web buckling at the coped region may occurwhen the cope length is long and/or the cope depth is large, provided that lateral-torsional buckling of the beam is prevented.coped I-beams although no experimental evidence was provided to support the recomendation. In order to verify suchrecomendation, an experimental and numerical investigation of coped I-beams with stiffeners at the coped region wasconducted and reported in this paper. The study showed that current recommendations did not consider the web distortionproperly, i.e. local web buckling could not be prevented efficiently if only horizontal stiffeners were provided at the copedregion. Both the test and the numerical results showed that the horizontal stiffeners at the cope displaced lateraly due to grossweb distortion. Based on the results of the parametric study of coped beams with diferent configurations of horizontal andvertical stiffeners, it was found that for cope depth to beam depth ratio (dc/D)≥0.3, both horizontal and vertical stiffeners arerequired in order to prevent local web buckling at the cope region. A preliminary recomendation for the design of copedbeams with both horizontal and vertical stiffeners was proposed according to the key findings of the investigation.

        Study of Anisothermal Ageing of CANTiM Shape Memory Alloys by Positron Annihilation

        The thermal-elastic property of high temperature CuAlNiMnTi shape memory alloys was found strongly affected by anisothermal ageing. With slow cycling (cycled at low heating and cooling rates), the transformation temperatures increase with the number of thermal cycle while the transformation enthalpy decreases and finally drop to zero, which indicated a total deterioration of the shape memory effect. Martensite stabilisation was found to be the cause of such deterioration. Positron Annihilation technique was employed to study the properties of the vacancies during the anisothermal ageing process. This was compared with the isothermal aged results. We propose that the changes in the quantity and characteristics of vacancies during anisothermal ageing are only responsible for the kinetics of the martensitic transformation. The result is the shift of transformation temperatures. However, the presence of vacancy agglomerations may have assisted the diffusion of atoms during ageing. Other factors like microstructural changes and atomic reordering will lead to the irreversible martensite stabilisation and make the transformation thermodynamically unfavourable.

      • Genome-wide association study in a Chinese population identifies a susceptibility locus for type 2 diabetes at 7q32 near <i>PAX4</i>

        <P><B>Aims/hypothesis</B></P><P>Most genetic variants identified for type 2 diabetes have been discovered in European populations. We performed genome-wide association studies (GWAS) in a Chinese population with the aim of identifying novel variants for type 2 diabetes in Asians.</P><P><B>Methods</B></P><P>We performed a meta-analysis of three GWAS comprising 684 patients with type 2 diabetes and 955 controls of Southern Han Chinese descent. We followed up the top signals in two independent Southern Han Chinese cohorts (totalling 10,383 cases and 6,974 controls), and performed in silico replication in multiple populations.</P><P><B>Results</B></P><P>We identified <I>CDKN2A/B</I> and four novel type 2 diabetes association signals with <I>p</I> < 1 × 10<SUP>−5</SUP> from the meta-analysis. Thirteen variants within these four loci were followed up in two independent Chinese cohorts, and rs10229583 at 7q32 was found to be associated with type 2 diabetes in a combined analysis of 11,067 cases and 7,929 controls (<I>p</I><SUB>meta</SUB> = 2.6 × 10<SUP>−8</SUP>; OR [95% CI] 1.18 [1.11, 1.25]). In silico replication revealed consistent associations across multiethnic groups, including five East Asian populations (<I>p</I><SUB>meta</SUB> = 2.3 × 10<SUP>−10</SUP>) and a population of European descent (<I>p</I> = 8.6 × 10<SUP>−3</SUP>). The rs10229583 risk variant was associated with elevated fasting plasma glucose, impaired beta cell function in controls, and an earlier age at diagnosis for the cases. The novel variant lies within an islet-selective cluster of open regulatory elements. There was significant heterogeneity of effect between Han Chinese and individuals of European descent, Malaysians and Indians.</P><P><B>Conclusions/interpretation</B></P><P>Our study identifies rs10229583 near <I>PAX4</I> as a novel locus for type 2 diabetes in Chinese and other populations and provides new insights into the pathogenesis of type 2 diabetes.</P><P><B>Electronic supplementary material</B></P><P>The online version of this article (doi:10.1007/s00125-013-2874-4) contains peer-reviewed but unedited supplementary material, which is available to authorised users.</P>

      • DRIVERS OF GREEN PRODUCT ADOPTION - THE ROLE OF GREEN PERCEIVED VALUE, GREEN TRUST AND PERCEIVED QUALITY

        This study investigates the role of green perceived value, green trust and perceived quality on the adoption behavior of green consumers. A survey was conducted using a sample of 188 respondents and a partial least squares approach was used to validate the research model. The research results show that both green perceived value and perceived quality have a direct influence on green purchase intention. Green trust also mediates the relationship between green perceived value and green purchase intention, which in turn predicts the purchase behavior of green products. The research results indicate that the drivers of green product adoption are represented by green perceived value, green trust, and perceived quality. Marketers should be putting more emphasis in this relatively new approach of green marketing strategies to induce purchase and adoption of green products by creating competitive advantages for the company.
