Experimental validation and network pharmacology evaluation to decipher the mechanism of action of Erythrina variegata L. bark against scopolamine-induced memory impairment in rats

        Biradar Prakash,Patil Vishal Supda,Joshi Hanumanthachari,Khanal Pukar,Mallapur Shamanand 경희대학교 융합한의과학연구소 2022 Oriental Pharmacy and Experimental Medicine Vol.22 No.1

This paper aimed to elucidate the effect of Erythrina variegata L. bark on scopolamine-induced memory impairment in rats and to decipher the molecular mechanism of phytoconstituents via the utilization of gene set enrichment analysis, network pharmacology coupled with in silico docking study. First, three models i.e. Morris Water Maze (MWM), Elevated Plus Maze (EPM), and Passive Avoidance Paradigm (PA) were utilized to elucidate the memory function. Second, the level of biomarkers i.e. acetylcholinesterase enzyme, reduced glutathione, and lipid peroxidation level were measured in brain tis- sues. Third, the key bioactive phytoconstituents targeting potential protein targets and pathways were identified through gene set enrichment analysis and network pharmacology. Lastly, the interaction between bioactive phytoconstituents with their respective targets was confirmed by molecular docking analysis. The MWM, EPM, and PA activity showed, scopolamine administration increased Escape Latency Time (ELT), Transfer Latency (TL), and Step Through Latency (STL) respectively on day 0th, 7th, 14th, and 21st, whereas treatment with E. variegata extract significantly reverse the ELT, TL and STL activ- ity. The decreased level of Acetylcholinesterase (AChE) and MDA level in treated animals reflected the enhanced memory and was found to be comparable withclinically proven drug i.e. donepezil. Sixty compounds were identified in EV bark, among which twenty-two compounds are predicted to modulate potential targets involved in AD and considered potentially bioactive. Further, the docking study revealed, Alpinumisoflavone, Auriculatin, Osajin, and Scandenone to have the highest binding affinity with Tau protein, Whereas Donepezil and Glucoerysodine with acetylcholinesterase enzyme.

Comparative Analysis of Despeckling Filters for Continuous Wave Doppler Images

Nagashettappa Biradar,Mohan Lal Dewal,Manoj Kumar Rohit 대한의용생체공학회 2015 Biomedical Engineering Letters (BMEL) Vol.5 No.1

Purpose The problems of speckle noise, low contrast, andother artifacts in continuous wave Doppler (CWD) basedechocardiographic images are identical to the issues prevailingin the B-mode images. The median filter and Gaussian lowpass filter are more commonly used for reduction of noise inCWD images. The applications of state-of-the-art despecklingfilters and their impact on delineation are not extensivelyanalyzed using CW Doppler images. Methods This paper analyses the noise reduction, edge andstructure preservation characteristics of ten filters for CWDimages in terms of 15 image quality metrics along with visualquality assessment. Further, three segmentation techniqueswere analyzed using original and noisy CWD images. Threeexisting delineation techniques are modified for tracing theouter boundaries of CWD spectrum. The Gaussian filter usedreaction-diffusion based active contour and thresholdingbased segmentation techniques are replaced by despecklingfilter based on anisotropic diffusion and local statistics. Results The noise is suppressed with edges and structurewell preserved on application of filters such as specklereducing anisotropic diffusion filter and local statics (meanand variance) based filter. The number of iterations requiredin complete boundary tracing is reduced on replacing theGaussian filter with despeckling filter in reaction diffusionbased active contour segmentation. Conclusions The analysis reveals that the despecklingtechniques enhance the visual quality of images and play avital role in delineation process. The reaction diffusion basedactive contour embedded with speckle reducing anisotropicdiffusion filter results in accurate contouring of the CWDimages.

A Novel Hybrid Homomorphic Fuzzy Filter for Speckle Noise Reduction

        Nagashettappa Biradar,M. L. Dewal,Manoj Kumar Rohit 대한의용생체공학회 2014 Biomedical Engineering Letters (BMEL) Vol.4 No.2

Purpose The presence of speckle noise and artifacts makedelineation of transthoracic echocardiographic (TTE) imagesquite difficult and challenging; the edges are to be preservedwhile removing noise. To address issues, a novel specklereduction technique is being proposed and analyzed. Methods Three fuzzy filters based on median and movingaverage concepts are experimented in homomorphic domainand the best one is further fine tuned for applications on TTEimages. The proposed hybrid homomorphic Fuzzy (HHF)filter is the sequential integration of homomorphic fuzzyfilter with anisotropic diffusion. The denoising characteristicsof HHF filter are compared with ten existing techniquestested in homomorphic and other seven in non-homomorphicdomain using seven different performance parameters alongwith visual quality assessment. Experimentations are performedon TTE images acquired in two parasternal and three apicalviews, since image in one view may not very precisely speakof underlying valvular abnormality. Results The edge preservation capability is increased bymany fold (around 8 times) upon integration of homomorphicfuzzy filter with the anisotropic diffusion filtering technique. Beta metric, figure of merit and structure similarity indicesare all greater than 0.97 for proposed HHF filter. Conclusions The performance of proposed HHF filter issuperior in comparison to seventeen state-of-art denoisingtechniques in terms of all seven performance parameters. Noise is reduced with the edges and structure of TTE imageswell preserved.

      • Cardioprotective effect of Argyreia speciosa (Burm. f) Boj. extracts against Isoproterenol- induced myocardial infarction in rats

        Thakker, Shalin,Biradar, S.M.,Habbu, P.V.,Mahadevan, K.M.,Thippeswamy, B.S.,Veerapur, V.P. Kyung Hee Oriental Medicine Research Center 2010 Oriental pharmacy and experimental medicine Vol.10 No.4

The present study was undertaken to evaluate the protective effect of ethanol (EtAS), ethyl acetate (EAAS) and aqueous (AQAS) extracts of Argyreia speciosa (AS) roots against Isoproterenol (ISO)-induced myocardial infarction in rats. The animals were exposed to isoproterenol (200 mg/kg. s.c) twice at an interval of 24 hrs. Cardioprotective effect was assessed by observing ECG parameters, serum marker enzymes and histopathology of the heart. Pretreatment of EAAS, and EtAS (200 mg/kg) resulted in a significant (P < 0.001) increase in P wave, QRS complex and R-R interval, whereas heart rate, QT interval and cardiac cycle were maintained near to normal values. EtAS and EAAS showed significant (P < 0.05; P < 0.001) reduction in all the tested diagnostic markers compared to ISO treated group. Histological studies on the structural changes of heart tissue supported the protective activity of AS. The result suggest that treatment of AS prior to ISO has a significant role in protecting the animals from ISO induced myocardial infarction.

      • Fluorescence Quenching of Bis-msb by Carbon Tetrachloride in Different Solvents

        Thipperudrappa, J.,Biradar, D.S.,Lagare, M.T.,Hanagodimath, S.M.,Inamdar, S.R.,Kadadevaramath, J.S. Korean Society of Photoscience 2004 Journal of Photosciences Vol.11 No.1

Fluorescence quenching of l,4-bis [2-(2-methylphenyl) ethenyl]-benzene (Bis-MSB) by carbon tetrachloride in five different solvents namely hexane, cyclohexane, toluene, benzene and dioxane has been carried out at room temperature with a view to understand the quenching mechanisms. The Stern-Volmer plot has been found to be non-linear with a positive deviation for all the solvents studied. In order to interpret these results we have invoked the Ground state complex and Sphere of action static quenching models. Using these models various rate parameters have been determined. The magnitudes of these parameters imply that sphere of action static quenching model agrees well with the experimental results. Hence the positive deviation in the Stem-Volmer plots is attributed to the static and dynamic quenching. Further, with the use of Finite Sink approximation model, it was possible to check whether these bimolecular reactions as diffusion limited and to estimate independently distance parameter R' and mutual diffusion coefficient D. Finally an effort has been made to correlate the values of R'and D with the values of the encounter distance R and the mutual diffusion coefficient D determined using the Edwardis empirical relation and Stokes-Einstein relation.

      • A Novel Design of Bidirectional WDM-PON by using Mode Locked Laser and RSOA

Kavita Awade,Rajkumar Biradar 보안공학연구지원센터 2015 International Journal of Future Generation Communi Vol.8 No.4

This paper presents Long Reach Wavelength Division Multiplexing Passive Optical Network (WDM-PON) system capable of delivering downstream 40 Gbit/s data and upstream 40 Gbit/s data on a single wavelength. The optical source for downstream data and upstream data is mode locked laser at central office and reflective semiconductor optical amplifier (RSOA) at each optical network unit. We use four RSOAs at each optical network unit for the 40-Gb/s upstream transmission. The operating wavelengths of these RSOAs are separated by the free-spectral range of the optical demultiplexer at the central office and remote node (RN) for demultiplexing the WDM channels. We extend the maximum reach of this WDM PON to be 117 km by using Erbium-doped fiber amplifiers at the RN. Bit error rate, were measured to demonstrate the proposed scheme. In this paper Long reach and large data service aspects of a WDM-PON is presented. The results show that the error-free transmission can be achieved for all WDM channels with sufficient power margins.

      • Unicast Routing for CWSB Pest Identification in Coffee Plantation using WSN: Energy Aware Approach

        Roshan Zameer Ahmed,Rajashekhar C. Biradar 보안공학연구지원센터 2016 International Journal of u- and e- Service, Scienc Vol.9 No.12

This paper proposes a Unicast Routing in Coffee Plantation (URCP), an energy aware approach for forwarding the information of Coffee White Stem Borer existence in Coffee Plantation using Wireless Sensor Networks. Acoustic signals that are made with biting sound by the pests inside the stem are captured by the nodes, and the information is aggregated at Cluster-Head (CH) which is to be conveyed to sink node in an energy-efficient way. Event-driven routing of the aggregated data is carried out by the CH which follows a Poisson distribution and maintains energy conservation by reducing the number of control packets for route establishment. Simulation analysis of URCP is compared with data routing for in-network aggregation and energy-efficient hierarchical clustering routing protocol in terms of packet delivery ratio, control overhead and energy consumption. The URCP simulation results outperform set alongside the corresponding techniques.
