        This experiment was carried out to develop a new technique of identifying XX or XY-bearing bisected embryos prior to implantation by immunological method. H-Y antiserum prepared in inbred Wistar female rats by repeated immunization with spleen cells from males of the same strain. The reactivity of H-Y antibody was confirmed by culturing mouse embryos in the medium containing H-Y antiserum and complement obtained from the guinea pig. The optimal condition for the activity of H-Y antibody was also investigated by culturing embryos under the concentration or affected H-Y antibody and culture rate. The bisection rate of mouse embryo at 4-cell stage(64.6%) was significantly(P<0.05) low than 8-cell(76.0%), morula(81.2%) or blastocyst(81.3%). And not significantly(P<0.05) different from 4 to 8-cell and morula to blastocyst stage embryos. When the bisected embryos without any damage from microsurgery were cultured in vitro up to twin blastocyst or twin reformated blastocyst, the in vitro development of demi-embryos bisected at morula to blastocyst was 81.6 to 84.7%. However, the in vitro development of demi-embryos bisected at 4-cell(31.6%) to 8-cell(37.8%) stage was significantly(P<0.05) lower. The lysis-rate of bisected 4- to 8-cell and morula to blastocyst embryo was 42.7 and 58.6%. The morphology embryos of zona-free and intact embryos showed the embryos lysis rate of 48.5 and 42.7% respectively(P<0.05).

      • 卵用鷄의 初産日齡이 産卵能力과 卵重에 미치는 影響

        본 試驗은 初産日齡이 産卵能力과 卵重에 미치는 영향을 살펴보고자 1986년 2월 1일부터 11월 30일까지 晋州農林專門大學 부속 動物飼育場에서 실시하였다. 初産日齡이 166~168일의 닭들이 165일 이전 또는 169일 이후의 개체들보다 産卵能力이 우수하였고 平均卵重은 初産日齡間에 차이가 없이 비슷하였다. 90%이상 産卵지속은 초산일령이 166~168일의 닭들이 26주령시부터 48주령시까지 지속되어 가장 우수하였다. For the purpose of identifing the reationship between the first egg day and other economic traits; egg production and egg weight, this experiment was divided 160 birds(DEKLB-Warren Sex-Sal-Link) into four first egg day groups and observed egg number and gee weight every day during 10 months form 1st February, 1986 to 30st November, the same year at Animal farm, Chin Ju Agricultural and Forestry Junior Technical College. Bird showing 166-168 day in first egg day layed the most eggs among four groups and they produced similar to average egg weight between the four groups. In egg production above 90%, 166-168 day group showed the best record as a 26-48 week.

      • 烏骨鷄의 特性에 關한 硏究 : 第Ⅱ報 烏骨鷄精液의 液狀保存과 性狀에 관한 硏究 Ⅱ. Studies on the Characteristic and liquid semen of korean Native ogolgye semen

        This experiment was carried out to develop dilutor for the usage of liquid semen and characteristic of Korean native Ogolgye semen. The results obtained in this experiment was summarized as follows, 1. The characteristic of Ogolgye semen used in this experiment showed of 5 month age 0.32ml(0.13∼0.46ml) average semen volume, 14.9×10 exp(8)) sperm number per ml, 18% (15∼21%) sperm abnormality, 82 live sperm index, 7.5pH, 96.57μ spermatozoa size, 12.44μ head size and 0.98μ head wide, and average of 6 month age 0.39ml(0.32∼0.45ml) semen volume. 17.8×10 exp (8)(12.2∼21.1×10 exp (8)) sperm number per ml. 14%(11∼20%) sperm abnormality, 83 live sperm index, 7.5pH, 100.05μ spermatozoa size, 12.65μ head size and 1.00μ head wide. 2. Diluted semen with 0.9Nacl and 5% glucose solution showed high sperm motility same whole semen just after dilution. 3. Ogolgye semen diluted in 0.9Nacl 5% glucose and mixture solution(0.9Nacl 1: 5% glucose 1) and storaged for 6hr showed no difference in sperm motility index between storage temperatures at 5 and 25℃. 4. Diluted semen with the mixture of 0.9% Nacl 5% glucose with same volume showed high sperm motility just after dilution. 5. It is known that Ogolgye whole semen has comparatively strong resistance against cold shock at 5℃ for min. 6. The semen diluted with 5% glucose solution showed better sperm motility than the semen diluted with 0.9% Nacl solution. 7. Semen extended with dilutor containing 0.8mg hostacillin and lmg streptomycin per ml of dilutor showed higher sperm motility index than that without antibiotics during storage.

      • 돼지의 분만유기에 관한 연구

        본 연구는 분만을 유기하기위하여 분만유도제인 tiaprost 0.3∼0.6mg을 임신 111일째 투여한후 20시간째 carazol과 oxytocin의 수준을 달리하여 분만유기를 시도한 실험1에서 144두중 133두가 실험2에서 140두 중 135두가 분만유기되어 92%와 96%의 높은 성적을 얻었으며 carazol 수준 1.5mg과 3.0mg을 처리했을 때 분만유기 및 분만완료까지의 시간도 단축되었으나 oxytocin의 수준간에는 차이를 나타내지 않았다. The present study was carried out to establish a practice for farrowing induction using tiaprost, carazol and oxytocin through conception stage in sows. After administration of 0.3-0.6mg tiaprost of farrowing induction in conception 111 days, Total 133 heads out of 144 heads in experiment 1 and Total 135 heads out of 140 heads in experiment 2 which were the treated difference interval carazol and oxytocin to farrowing in 20th were farrowed and obtained high result of 92% and 96% and treatment of carazol interval 1.5mg and 3.0mg, time of farrowing induction and farrowing completion were shorten but oxytocin interval did not show difference.

        This experiment was conducted to investigate the effect of separation of chick sex on growth with the data obtained from 250 pieces Broiler (Hubbard) fed and alloted 3 treatment at the females(♀) + males(♂) (A), separation of chick sex at 4 week of age (B) and separation of chick sex at 3 week of age (C) of during the experimental period of 35 days from May 10, 1983. to June 13. the same year at brooder house. Jin Ju Agricurtural and Forestry Junior Technical College. The results obtained were summarized as follows; 1. Body weight gain was highest in the C plot and there were significant (p<0.01) differences between the others except A and B. 2. Feed conversion at each experimental plot was 2.07(B). 2.095(C) and 2.10(A) in order. 3. Point spread at each experimental plot was 305.65%(C). 302.05(B) and 295.6%(A). but the difference was not significant. 4. In economic analysis of broiler production, the highest income was C(846.8 won) and B (829.0 won), A (808.9 won) in order.

      • 한우 체외수정란의 동결보존 및 핵이식에 관한 연구 : Ⅱ. 체외수저란의 동결방법이 동결.융해후 생존율에 미치는 영향 Ⅱ. Effects of Survival Rates of Frozen-Thawed Embryo Frozen Methods

        본 연구는 수정란의 동결기법을 확립하고자 배반포기에 있는 한우 체외수정란을 이용하여 유리화 및 직접이식 동결방법으로 동결을 실시하여 동결·융해후 생존성, 동해방지제의 종류, 평형 및 할구의 손상여부 등에 대한 영향을 조사한 결과는 다음과 같다. 유리화 동결방법중 GESD-VS와 EFS 용액을 동해방지제로 사용하였을 때 동결·융해 수정란의 생존율은 각각 69.8 및 37.3%로써 이들간에 유의적(P<0.05)인 차이가 있었으며, 동결기를 이용한 직접이식 동결법은 생존율이 56.0%로써 유리화 동결방법과는 유의적(P<0.05)인 차이가 없었다. 동결·융해후 생존한 수정란의 발달율은 GESD-VS용액을 사용하였을때가 53.4%로써 EFS용액을 사용하였을때의 24.0%보다는 유의적(P<0.05)으로 높게 나타났다. 1 및 2단계 평형시 단계별 생존율은 각각 11.7 및 23%로써 이들간에 유의적(P<0.05)인 차이가 없었으며, 동결·융해 수정란의 발달율도 각각 10 및 16.8%로써 평형방법간에 유의적(P<0.05)인 차이가 없었다. 신선수정란, 유리화 동결수정란 및 동결비를 이용한 직접이식 동결법으로 동결하였을 때 동결·융해 수정란의 할구수는 각각 98.7±2.3개, 69.1±2.6 및 84.2±2.7개로써 동결방법들간에 유의적(P<0.05)으로 할구수의 차이가 많았다. This study was carried out to investigate the effect of cryoprotectants, equilibration step, number of blastomeres on survival and developmental rates of bovine in vitro fertilized blastocysts(Korean native cattle) frozen/thawed by vitrification and frozen for direct transfer(1.8M ethylene glycol). The bovine oocytes were collected from 2-5mm follicles, matured for 24hrs in 5% CO_2 incubator and then fertilized with cauda epididymis sperm. Blastocyst at day 7-9 ater IVF were frozen in vitrification and frozen for direct transfer(1.8M ethylene glycol). Survival and developmental rates of frozen-thawed IVF embryos were examined. The results were as follow ; The survival rate of IVF blastocyst after vitrification in GESD-VS, EFS and 1.8M ethylene glycol solution were 69.8, 37.3 and 56.0%, respectively(P<0.05). When equilibrated in 1 step(11.7%) was similar to the result from 2 step(23.0%), and no significantly(P<0.05) different between the equilibrated steps. The number of blastomere at blastomere at blastocyst of bovine embryos frozen/thawed in fresh, vitrification and freezing for direct transfer were 98.7±2.3, 69.1±2.6 and 84.2±2.7, respectively, and were significantly(P<0.05) different between the freezing methods.

      • 생육시기별 홀크롭 맥주보리 Silage의 품질 평가

        This experiment was conducted to investigate the effect of maturity stages on the quality of brewer's barley silage. The silage was made at heading, milky, dough and yellow ripe stages, and the quality of the silage was evaluated on the bases of the chemical composition, pH, ammonia nitrogen and micro-organisms. The results obtained were summarized as follows : 1. Moisture, crude protein, crude fat and crude fiber contents of raw brewer's barley were decreased with advancing the maturity. NDF(neutral detergent fiber) content was significantly higher (P<0.05) in milky stage (70.35%), but ADF(acid detergent fiber) content was higher (P<0.05) in heading stage (33.19%) than other maturity stages. 2. Moisture, crude protein, crude fiber and crude ash contents of silage were decreased with advancing the maturity. NDF content was significantly higher (P<0.05) in milky stage (68.29%), but ADF content was higher (P<0.05) in heading stage (36.29%) than other maturity stages. 3. pH values of silage were increased with advancing the maturity. Total nitrogen content was significantly higher (P<0.05) in yellow ripe stage (474.3mg/100g), but ammonia nitrogen content was higher (P<0.05) in heading stage (14.91mg/100g) than other maturity stages. 4. The number of total bacteria and latic acid bacteria were increased with advancing the maturity, and were estimated 5×10 exp(2)∼2.5×10 exp(6) for total bacteria and 1×10 exp(2)∼3.2×10 exp(5)/g for lactic acid bacteria, respectively.

      • 한우 체외수정란의 동결보존 및 핵이식에 관한 연구 : Ⅰ. 계절 및 배양조건이 한우 체외수정란의 생산에 미치는 영향 Ⅰ. Effects of season and culture condition on production of bovine in vitro fertilization embryos

        본 연구는 도살장에서 도살되는 한우의 난소를 적출하여 난소로부터 미성숙 난포란을 회수하는데 있어서 계절별 난포란의 quality과 수정율의 차이점에 대한 규명과 체외성숙-체외수정한 난자를 체외배양기법을 확립하고자 본 실험을 수행하였다. 도살된 한우의 난소로 부터 난포의 크기별로 난포란을 회수하여 체외성숙을 유도하였다. 체외성숙란을 정소상체 미부정자를 이용하여 체외수정을 시킨 후 난관상피세포에서 공배양을 실시하여 수정란의 체외발달을 유도하였으며, 난포란의 계절별 회수율, 체외수정율 및 배양체계에 따른 체외발달율을 조사한 결과는 다음과 같다. 봄(426개), 여름(433개), 가을(419개) 및 겨울(428개) 등 계절별로 분류하여 모두 1,706개의 난소로부터 14,674개의 난포란을 채취하였다. 계절에 따른 1등급 회수율은 22.2(봄), 6.5(여름, 20.6(가을) 및 18.1%(겨울)로써 이들간에 유의적(P<0.05)인 차이는 없었으나 겨울철에 회수한 난자들이 다소 낮게 나타났다. 회수된 2등급 난자의 비율도 봄, 여름, 가을 및 겨울이 각각 43.7, 41.6, 44.1 및 35.2% 로써 계절에 따른 유의적(P<0.05)인 차이가 없었다. 봄, 여름, 가을 및 겨울 등 계절별 체외수정율은 각각 76.0, 80.6, 75.7 및 72.1% 로써 계절에 따른 수정율은 유의적(P<0.05)인 차이가 없었으며, 배반포기 단계까지 발달한 체외수정란은 TCM-199 및 Hams-F10 배양액에 10% FCS와 난관상피세포를 공배양을 실시하였을때는 각각 23.3 및 21.7%의 발달율을 보였으나, TCM-199 배양액에 10%FCS만을 첨가하였을 때는 전혀 발달이 되지 않았다(P<0.05). 본 연구에서는 계절별 회수율의 차이는 많은 요인이 있을 수 있겠으나 주로 계절별 사양관리의 차이 및 계절에 따른 조사료의 공급수준에 의한 영양수준의 차이 등에서 기인되는 것으로 생각된다. 체외수정란의 배반포기로의 발달을 위한 배양조건은 TCM-199 또는 Hams-F10배양액에 10% FCS를 첨가한 배양액으로 난관상피세포와 공배양을 실시하는 것이 효과적인 방법이라고 생각된다. The present study was carried out to determin the seasonal effects of oocyte collection and culture condition of Korean native cows and in vitro developmental rate of 1VF bovine embryos. The follicular oocytes were collected from small, medium and large follicles, and classified into 4 grades by the morphology of cumulus cells attached and homogenous cytoplasm. They were fertilized in vitro by epididymis spermatozoa treated with heparin for 24 hrs., and then the zygotes were co-cultured in vitro with bovine oviductal epithelial cells for 7 to 9 days. The results obtained were summarized as follows : The follicular oocytes recovered were classified into grade I 22.2% as spring, 26.5% as summer, 20.6% as autumn and 18.1% as winter. The cleavage rates of the embryos obtained from 76.0(spring), 80.6(summer), 75.5(autumn) and 72.1(winter) respectively(P<0.05). The developmental rates(23.3 and 21.6%)of embryos cultured in TCM 199+10% FCS-OCEM and Harms-F10+10%FCS+OECM up to blastocyst stage were higher(P<0.05) than the TCM-199+10%FCS culture condition(P<0.05)

      • 소 난포란의 체외성숙도가 수정 및 체외발달에 미치는 영향

        본 연구소는 소 난포란을 이용하여 체외수정시에 체외성숙 시간이 체외수정 및 체외발달에 미치는 영향을 조사하고자 실험을 수행하였다. 도살장에서 도살된 한우의 난소를 채취하여 실험실로 2시간 이내에 운반하여 난소를 깨끗이 세척한 후 18 gauge needle로 난포의 크기별로 난포란을 회수하여 10% FCS가 첨가된 TCM-199 배양액에 FSH(35 ㎍/ml), LH(10㎍/ml), estradiol-17β(1㎍/ml)가 첨가된 체외성숙용 배양액에서 20-28시간 동안 체외성숙을 유도하였다. 체외성숙란을 한우 동결정액을 이용하여 체외수정을 시킨 후 난관상피세포에서 공배양을 실시하여 수정란의 체외발달을 유도하였으며, 난포란의 등급별 회수율과 체외성숙시간에 따른 수정율 및 체외발달율을 조사한 결과는 다음과 같다. 난소에서 난포의 크기에 따른 Grade I의 회수율은 소, 중 및 대난포에서 각각 31.3, 27.8 및 3.7%였으며, 48개의 난소로 부터 모두 398개의 미성숙 난자를 회수하여 난소당 8.3개의 미성숙 난자를 회수하였다. 등급별로는 2.3, 2.5, 2.3 및 1.2%의 회수율을 보였다. 체외수정란의 배반포로의 발달율은 24시간 성숙시켰을 때 27.2%로서 20(11.6%) 및 28(9.0%)시간 성숙시켰을 때 보다 유의적(P<0.05)으로 높았다. 그러나 22, 24 및 26시간 성숙시켰을 때는 성숙시간에 따른 발달율은 유의적(P<0.05)인 차이가 없었다. The ovaries of Korean native cow or heifers were obtained from an abattoir and kept on 28-30℃ and transported to laboratory within 2 hrs. The follicular oocytes were collected from small, medium and large follicles, and classified into 3 grades by the morphology of cumulus cells attached and homogenous cytoplasm. The oocytes were matured in vitro for 24 hrs. in TCM-199 supplemented with 35㎍/ml FSH, 10㎍/ml LH, 1㎍/ml estradiol-17β and granulosa cells at 39℃ under 5% CO_2 in air. They were fertilized in vitro by freezing sperm treated with heparin for 24 hrs., and then the zygotes were co-cultured in vitro with bovine oviductal epithelial cells for 7 to 9 days. The follicular oocytes recovered were classified into 41.7% as gradae Ⅰ, 51.5% as grade Ⅱ and 6.8% as grade Ⅲ. The number of oocytes recovered per ovary was averaged 8.3 and they were classified into 2.3 as grade Ⅰ, 2.5 as grade Ⅱ and 2.3 as grade Ⅲ. The cleavage rates of the embryos obtained from 73.3, 78.4, 80.4, 77.3 and 72.7% in the groups of maturation age of 20, 22, 24, 26 and 28 hours respectively. The in vitro culture of bovine embryos with maturation age from 22-26 hours improved significantly(P<0.05) the subsequent development to transferable embryos(20.4-25.0%), compared with 20 hours(11.6%) or 28 hours(9.0%).

      • Rat H-Y 抗體가 생쥐 수정란의 발달단계에 따른 체외배양에 미치는 영향

        本 硏究는 免疫學的 방법에 의하여 性을 調節할 수 있는 방법을 개발하고자 近郊系統의 Wistar흰쥐의 수컷비장을 摘出하여 均質化시킨 다음 상충액을 취하여 同種의 암컷 腹膣內에 接種함으로서 H-Y抗原에 대한 H-Y 抗 血淸을 제조하였으며, 제조된 H-Y抗 血淸과 함유한 培養液에 ICR系統의 생쥐로부터 각 발달단계별로 채란한 수정란을 NaHCO3-BMOC-3 배양액에 H-Y항 혈청과 보체를 첨가하여 CO_2 incubator에서 24∼30시간 배양을 실시하였다. 본 연구에서 얻은 결과는 다음과 같다. H-Y 抗體의 形成은 rat 10마리중 8마리에서 抗體가 형성되어 80%의 形成率을 보였으며, 427개의 수정란을 H-Y 抗體를 處理하여 이중 182개(43%)가 파괴 되었다. H-Y 抗血淸 및 補體의 存在下에서 培養된 2-, 4-, 8-, 16-세포기, 상실배 및 胞胚期 수정란의 파괴율은 각각 0.0, 42.9, 54.1, 47.0, 48.1 및 47.5%로서 2-세포기와 4-세포기 以上間에는 유의적 (P<0.05)인 差異가 있었으나, 4-세포기 이상과 胞胚期 受精卵間에는 H-Y 항체가 미치는 영향은 유의적(P<0.05)인 차이가 없었다. When the embryos were in vitro cultured in the medium of H-Y antiserum and complement the lysis rate of 2-, 4-, 8-, 16-, morula and blastocyst was 0.0, 42.9, 54.1, 47.0, and 47.5% respectively, but significantly(P<0.05) low than that of 2-cell stage.
