유가식 배양에서 재조합 대장균으로부터 Interferon ${\alpha}$-1 생산에 산소 공급이 미치는 영향

산소를 많이 소비하는 발효공정일수록 배양액중의 용존산소의 농도가 목적생산물의 생산성에 많은 영향을 주는 경우가 많다. 때문에 고농도 발효에 앞서, 발효조의 sparging hole로부터 임펠러 높이에 따른 산소전달 능력을 알아본 결과 공기공급이 1 vvm, 교반속도가 600 rpm에서 산소전달계수($K_La$)는 2.67($min^{-1}$)으로 가장 높았다. 배양 시 용존산소 농도를 20% 이상 유지시켰을 때 온도에 따른 k6ub/IFN-${\alpha}1$ 생성은 $30^{\circ}C$에서 세포증식을 하고 $25^{\circ}C$에서 IPTG로 Induction 하였을 때 발현율이 6.43mg/ml로 total protein의 37%로 가장 많은 양이 발현되는 것을 알 수 있었다. 용존산소 농도에 따른 k6ub/IFN-${\alpha}1$의 발현양은 용존산소 농도가 35%일 때 가장 높은 수율을 나타냈다. 용존산소량은 산소소비 속도를 측정함으로써 정확한 임계점을 찾을 수 있었는데 용존산소량이 35% 유지될 때 산소 전달 속도와 비교하여 가장 적당한 산소공급량임을 확인할 수 있었다. In order to achieve high-level expression of interferon-${\alpha}1$ (IFN-${\alpha}1$) during fed-batch fermentation of recombinant E. coli, effects of oxygen supply and induction temperature on the expression of recombinant proteins were evaluated. Supplementation of oxygen and its transfer into cells is one of the most important parameters involved in the design and operation of mixing-sparging equipment for bioreactors. Generally, higher oxygen supply stimulates cell growth of aerobic microorganism and consequently the amount of products is increased. In this study, the optimum aeration strategy for the higher production of IFN-${\alpha}1$ during fed-batch fermentation of recombinant E. coli was surveyed. The growth of the cells was also monitored with four different concentrations of dissolved oxygen (DO; limiting, 20%, 35%, 50%) conditions. The DO was controlled by varying aeration rates of air and pure oxygen. Oxygen uptake rate (OUR) and specific oxygen uptake rate (SOUR) were evaluated and compared for the enhanced growth and induction of the cells and IFN-${\alpha}1$, respectively. We confirmed that increased DO by additional oxygen supply, up to 35%, can improve the production of IFN-${\alpha}1$ during the fermentation.
악취 Sampling Bag의 황 화합물질 안정성 연구

In this study, stability of 4 sulfur compounds (H<sub>2</sub>S, CH<sub>3</sub>SH, DMS, and DMDS) was investigated for up to 96 h (at 6, 12, 24, 48, 72, and 96 h time points). For these experiments, each sulfur standard was prepared at 10 nmol/mol in a 10 L sampling bag (Teldlar bag). To investigate the stability of the sulfur compounds, regression analysis was applied to the data obtained by the concentration reduction over time at the 95% confidence level. According to regression analysis, the concentration of sulfur compounds in 48 h had statistically significant difference (P-value < 0.05) from stability of 4 sulfur compounds (H<sub>2</sub>S, CH<sub>3</sub>SH, DMS, and DMDS). The results also indicated that the heavy sulfur compounds (DMS and DMDS) tend to show more rapid concentration reduction than lighter sulfur compounds (H<sub>2</sub>S and CH<sub>3</sub>SH). This study can be used to improve the reliability of analysis by judging quantitative concentrations of sulfur compounds when controlling odor-polluted areas.
Intranasal delivery of siRNA targeting NR2B attenuates cancer-associated neuropathic pain

Purpose This study aimed to reduce cancer-associated pain by blocking pain signals through the intranasal administration of siRNA targeting the NMDA subunit NR2B (siNR2B). Methods Cancer pain models were established by injecting 3 × 105 B16F10 melanoma cells into the left hind paws of C57BL/6 mice. To evaluate pain reduction, 600 pmol of siNR2B was complexed with the RVG9R peptide at a 20:1 molar ratio, or 5 mg/kg NR2B receptor antagonist Ro25-6981 was used as a positive control. Melanoma-xenografted mice were intranasally administered the peptide/siRNA complex or intraperitoneally inoculated with Ro25-6981 three times a week for 3 weeks. The mechanical withdrawal threshold was determined using an electronic von Frey apparatus. Results The therapeutic effect of intranasally administered siNR2B was observed 21 days after cancer cell implantation in a hind paw melanoma model. NR2B expression in the cancer model was approximately twice that in the normal animals. The groups treated with siNR2B or Ro25-6981 exhibited approximately 60 and 50% of NR2B expression in the thalamus, respectively. This reduced pain signaling in the thalamic region, as evidenced by a decrease in phosphorylated extracellular signal-regulated kinase. In addition, the siNR2B-treated group displayed significant behavioral improvements, a marked reduction in cancer-induced pain, compared with controls. siNR2B treatment in a cancer-induced murine model did not affect the general cognitive function. Conclusion This study demonstrated that the intranasal delivery of siNR2B in a murine model effectively reduced cancerinduced neuropathic pain by downregulating overexpressed NMDA receptor-mediated pain signaling in the thalamus.

This study is to find out influence on the physical fitness by reducing body weight. The 24 subjects selected were Judo players attending Pyung Taek Boy's High School. The processes of degree about reducing body weight are 2%, 2.5%, 2.9%, 3.3%, 4.2%, of one's normal body weight. The method used was to reduce and increas the exorcises. The objects used for physical fitness test tore gripping power, pull-ups, vertical jump, 1,000m running, sit-ups and flexibility of waist. The influences on the physical fitness by gradual degree of reducing body weight were as follows. 1) The record of gripping power went down when one reduced his weight by 2.5% . 2) The record of pull-ups went up when one reduced his weight by 2% and 2.5%, and went down when one reduced his weight by 3.3%. 3) The record of vertical jump went up when one reduced his weight by 2%, and went down hen one reduced by 2.9% 4) The record if 100m running showed no difference until one reduced his weight by 4.2% 5) The record of 1,OOOm running went down when one reduced his weight by 2.5% . 6) The record of sit- ups went down when one reduced ruts weight by 2.9% . 7) The record of waist flexibility went up when one reduced his weight from 2.5% to 4.2%.
