        한약재 마늘(Allium sativum L.)의 식별을 위한 유전자 감식연구

        Objectives : This study was carried out to identify DNA markers of "Allium sativum" be circulated from Korea and China, which is difficult to discriminate from morphological characters because of fragmental materials of bulb. That is, all these studies focused on the discrimination of Allium sativum L. But these day, Chinese A. sativum was in circulated Korean A. sativum in Korean medicine markets. Therefore, the purpose of our study was to develop molecular markers for discrimination between Korean A. sativum and imports from China. Methods : Materials were collected randomly from a markets in Korea and China and be analyzed with matK, ndhF and trnL-F regions of chloroplast DNA (cpDNA). We collected 45 A. sativum individuals from Korean and Chinese medicine markets, in 2013. Results : As a results, matK and ndhF regions of cpDNA was shown to be identify, Species that grow from warm place and cold place can divide as five SNP (Single nucleotide polymorphisms) markers in matK and ndhF genes. Also, in trnL-F regions, found one SNP that can divide Korean A. sativum and Chinese A. sativum. Conclusions : From the analysis of matK and ndhF regions of cpDNA, we presumed that three markers of cpDNA were found by useful marker that can distinguish Korean, Chinese, Warm place type, and Cold place type. Individual differences of Korean and Chinese was thought that appear in geographical difference and genetic difference by environment for long hour even if same species.

      • 진생베리 열수 추출물의 랫트 실험적 신경병성 통증 완화효과

        PURPOSE : we intended to observe the possibilities that standardized GB aqueous extracts have favorable analgesic effects on the CCI, a model of neuropathic pain in Sprague -Dawley (SD) rats, as compared to those of amitriptyline hydrochloride, a serotonin and norepinephrine reuptake inhibitor offer antidepressants for management of chronic pain, 10 mg/kg intraperitoneally treated rats as reference drug treated group. METHODS : GB aqueous extract 500, 250 and 125 mg/kg were orally administered, once a day for 14 days from 24 hrs after CCI surgery. Then the changes on the body weights and gains during experimental periods, analgesic activities on mechanical and cold allodynia, spinal cord lipid peroxidation and antioxidant defense systems - glutathione (GSH) contents, superoxide dismutase (SOD) and catalase (CAT) activities were monitored with Mrna expressions of ionized calcium-binding adapte rmoleculel (Iba-1), a microglia activation marker, glial fibrillary acidic protein (GFAP), an astroglia activation marker, inflammatory markers such as tumor necrosis factor (TNF) - α amd inducible nitric oxide synthase (iNOS). The results of GB aqueous extracts were compared with those of amitriptyline 10 mg/kg intraperitoneally treated rats. RESULTS : these CCI-induced inflammatory and oxidative stress-related neuropathic pains were favorably and dose-dependently inhibited by 14 days continuous oral administration of three different dosages of GB aqueous extracts - 500, 250 and 125 mg/kg as comparable to those of amitriptyline 10 mg/kg intraperitoneally injected rats in GB aqueous extracts 250 mg/kg orally administered CCI rats, at least in a condition of the present CCI rat model. CONCLUSION : The results obtained in this study suggest that GB aqueous extract has sufficient beneficial analgesic activity to inhibit CCI-induced chronic pain compared to amkriptyline 10 mg/kg intraperitoneal injection at 250 mg/kg oral dose.

        천궁의 기원과 식별을 위한 분자마커

        Objectives: This study was carried out to discriminate origin and molecular marker of oriental medicine Cnidii Rhizoma be circulated between Korea and China, which is difficult to discriminate from morphological distinction because of a fragmental materials of roots. Methods: Materials were collected randomly from a medicinal herb markets in Korea and China and be analyzed with ITS (internal transcribed spacer) regions of nuclear ribosomal DNA (nrDNA). Results: As a results, ITS regions of nrDNA was shown to be identify as three molecular markers. Cnidii Rhizoma was made up syster group of the genus Ligusticum L. and divided into three groups with Tou-chun-gung, IL-chun-gung and China-chun-gung. Conclusions: From the analysis of ITS regions of nrDNA, we presumed that it is the same origin of Cnidii Rhizoma from Korea and China because of phylogenetic tree consisted of sister groups with the genus Ligusticum than the genus Cnidium.
