Vol.43 No.12

As we all know, another trend in contemporary documentary photography is to express the photographer’s self-independent emotion and worldview, which brings new topics and higher pursuits to photographers. In this paper, by studying the Chinese documentary photographer Yang Yankang, taking his series of works “Mind Image” as an example, this article analyzes the photographer’s emotional state during shooting, and explores the ways in which he expresses religious thoughts in his photos, in order to make a difference to the photographer’s creation. This article adopts semiotic analysis methods, starting from the three semiotic structural levels of “linguistic message signs”, “coded message signs”, and “metaphoric signs”, deconstructing photographers conveying different levels of information signs to readers through photos, interpreting messages and readers the way to create an emotional connection. The article selects typical photos, specifically analyzes the construction process of Tibetan Buddhism-related information symbols in Yang Yankang’s photos, analyzes the content of the work and the expression of religious thoughts, finds the deep meaning of the photos from the information elements of the photos, and thinks based on this Documentary photography has more possibilities in the future.

      • Complete chloroplast genome assembly of Brassica species from Next Generation Sequencing-based whole genome resequencing data and comparative analysis

Vol.2014 No.07

Chloroplasts are plant-specific organelles, which have their own genome. Most of the plant chloroplast genomes (CP genome) are highly conserved in terms of its gene contents and genome structures, and they exist in cells with abundant copy numbers. Because of numerous copy numbers, the complete chloroplast sequence assembly pipeline with small amount of whole genome resequencing data, produced by NGS technique, was established in our laboratory. From 14 accessions of cabbage (Brassica oleracea L.) resequencing data produced by Illumina Hi-seq 2000, CP genomes were assembled and compared to each other. 18 sequence variance regions were detected, and 6 HRM(High Resolution Melting curves) markers were developed. Approximately 1 Gb of whole genome sequencing data of 10 Brassica rapa and 2 Brassica napus were also obtained from Institute of Vegetables and Flowers, Chinese Academy of Agricultural Science. With these resequencing data, all CP genomes from these accessions were assembled. Total 27 complete CP genomes of B.oleracea, B.rapa, B.napus, and brassico-raphanus which is a novel allotetraploid species between B.rapa and Raphanus sativus, were compared in sequence level. Phylogenetic analysis based on the comparison revealed that B.rapa could be the maternal species when rapeseeds and brassico-raphanus became allotetraploid species. Additionally, CP genome of B.napus cv.M083 is closer to B.rapa accessions than the other B.napus accessions, thus B.napus could have several different origins.

      • Diversity and Inheritance of Intergenic Spacer Sequences of 45S Ribosomal DNA among Accessions of <i>Brassica oleracea</i> L. var. <i>capitata</i>

        Yang, Kiwoung,Robin, Arif Hasan Khan,Yi, Go-Eun,Lee, Jonghoon,Chung, Mi-Young,Yang, Tae-Jin,Nou, Ill-Sup MDPI 2015 INTERNATIONAL JOURNAL OF MOLECULAR SCIENCES Vol.16 No.12

<P>Ribosomal DNA (rDNA) of plants is present in high copy number and shows variation between and within species in the length of the intergenic spacer (IGS). The 45S rDNA of flowering plants includes the 5.8S, 18S and 25S rDNA genes, the internal transcribed spacer (ITS1 and ITS2), and the intergenic spacer 45S-IGS (25S-18S). This study identified six different types of 45S-IGS, A to F, which at 363 bp, 1121 bp, 1717 bp, 1969 bp, 2036 bp and 2111 bp in length, respectively, were much shorter than the reported reference IGS sequences in <I>B. oleracea</I> var. <I>alboglabra</I>. The shortest two IGS types, A and B, lacked the transcription initiation site, non-transcribed spacer, and external transcribed spacer. Functional behavior of those two IGS types in relation to rRNA synthesis is a subject of further investigation. The other four IGSs had subtle variations in the transcription termination site, guanine-cytosine (GC) content, and number of tandem repeats, but the external transcribed spacers of these four IGSs were quite similar in length. The 45S IGSs were found to follow Mendelian inheritance in a population of 15 F1s and their 30 inbred parental lines, which suggests that these sequences could be useful for development of new breeding tools. In addition, this study represents the first report of intra-specific (within subspecies) variation of the 45S IGS in <I>B. oleracea.</I></P>

      • Identification of various complete circle form of mitochondrial genome sequences using WGS in Brassica oleracea

        Kiwoung Yang,Nasar Uddin Ahmed,Jonghoon Lee,Junki Lee,Ill-Sup Nou,Tae-Jin Yang 한국육종학회 2014 한국육종학회 심포지엄 Vol.2014 No.07

Mitochondria are essential organelles of eukaryotic cells and plant cells contain varying numbers of mitochondrial genome sequences. Sizes and shapes of mitochondria differ within a tissue or in the same cells. Previously sequenced complete mitochondrial genome (NC_016118) of Brassica oleracea size was 360,271 bp, where segmental duplication (repeat block) was 141,800 bp. In this study, we resequenced this whole mitochondrial genome by using WGS (whole genome sequencing) and assembled organelles genome method (unpublished). Newly sequenced mitochondrial genome length was 219,975 bp and circle form. A new sequence segment of approximately 4,800 bp was obtained compared to the previous genome sequence without any large repeat block. Newly obtained mitochondria genome sequence was compared with recently reported mitochondria genome sequences of various species (B. oleracea, B. juncea, B. rapa, B. napus and B. carinata) and subspecies (cabbage, cauliflower, brussels sprouts, kohlrabi, broccoli and kale) by PCR using primers specifying different region of genome sequences. PCR analysis results have also confirmed the variation between previous and newly sequenced mitochondrial genome circles form. Thus, the results suggest new B. oleracea mitotype, including evolutionary events such as inheritance, rearrangement, genome compaction, and diversity

      • Construction of high resolution genetic map and QTL mapping for clubroot resistance using genotyping-by-sequencing analysis in cabbage

        Jonghoon Lee,Nur Kholilatul Izzah,Beom-Soon Choi,Ho Jun Joh,Sang-Choon Lee,Sampath Perumal,Joodeok Seo,Kyounggu Ahn,Eun Ju Jo,Gyung Ja Choi,Ill-Sup Nou,Yeisoo Yu,Tae-Jin Yang 한국육종학회 2015 한국육종학회 심포지엄 Vol.2015 No.07

Clubroot is a devastating disease caused by Plasmodiophora brassicae and results in severe losses of yield and quality in Brassica crops including Brassica oleracea. Therefore, it is important to identify resistance gene for CR disease and apply it to breeding of Brassica crops. In this study, we applied genotyping-by-sequencing (GBS) technique to construct high resolution genetic map and mapping of clubroot resistance (CR) genes. A total of 18,187 GBS markers were identified between two parent lines resistant and susceptible to the disease, of which 4,103 markers were genotyped in all 78 F2 plants generated from crossing of both parent lines. The markers were clustered into nine linkage groups spanning 879.9 cM, generating high resolution genetic map enough to refine reported reference genome of cabbage. In addition, through QTL analysis using 78 F2:3 progenies and mapping based on the genetic map, two and single major QTLs were identified for resistance of race 2 and race 9 of P. brassicae, respectively. These QTLs did not show collinearity with CR loci found in Chinese cabbage (Brassica rapa) but roughly overlapped with CR loci identified in cabbage for resistance to race 4. Taken together, genetic map and QTLs obtained in this study will provide valuable information to improve reference genome and clubroot resistance in cabbage.

2006 Korean journal of medical education Vol.18 No.3

Purpose: This study was designed to investigate the effects of information sharing between students on results of clinical performance examination (CPX). Methods: 143 third-year students, who completed their core clinical clerkship, were randomly assigned to one day of a 3-day, 6 station CPX. Station checklists, scored by the SPs, consisted of history taking, physical examination, information sharing, clinical courtesy and patient-physician interaction. We compared the CPX station scores, CPX domain scores, self assessment scores and GPA of the three groups tested on different days with ANOVA, and analyzed the differences in CPX scores, after controlling for GPA, with ANCOVA. The correlations among the CPX total and domain scores, self assessment and the GPA of the 3rd year students were calculated. Data regarding information sharing between students and self-assessment were obtained through student questionnaires. Results: CPX total scores, self assessment scores and GPA of 3rd year students from day-2 were significantly higher than scores from day-1 or day-3. Checklist analysis showed that PE, CC and PPI scores were significantly higher on day-2, but Hx and IS scores were not significantly different. Results from questionnaires showed that 43% of students shared evaluation information. And CPX total scores had little correlation with self assessment scores. GPA of 3rd year students showed a similar pattern with the CPX totalscore. Conclusion: There is information sharing occurring between students during clinical performance examinations. Although there are significant differences in CPXscores for the treatment group, it seemed that CPX scores are not affected by information sharing between students. Reliability of information sharing from other students is low. The generalization of this study should be interpreted carefully.

Vol.34 No.1

This study examined the effects of paternal participation in child care and social support on planning for a second childbirth focusing on the mediating effect of the mother`s parenting stress. The 887 subjects studied in this paper were mothers who have a first child and were selected from a sample of the 2nd Panel Study of Korean Children(PSKC) by the Korea Institute of Child Care and Education(KICCE) conducted in 2009. Data was analyzed using a Structural Equation Model(SEM) to identify a structure and relationships among factors. The significance of indirect effects to test the mediating effect of maternal parenting stress was indentified via bootstrapping. Results indicated that maternal parenting stress partially mediated the relationship between the father`s participation in child care and planning for a second childbirth and absolutely mediated the relationship between social support and planning for second childbirth. The findings reveal the importance of reducing parenting stress given its substantial impact on planning for a second childbirth.
