Mechanism study of dTDP-D-Glucose 4,6-Dehydratase by Site Directed Mutagenesis

dTDP-D-glucose 4,6-dehydratase는 dTDP-glucose에서 dTDP-4-keto-6-deoxy-D-glucose로 전환시키는 중요한 산화환원 효소이다. 이 효소의 반응 경로는 NAD' 조효소 와 수소 전달에 관여하는 두 개의 염기에 의한 산화, 탈수, 환원반응으로 보여지며, dehydratase와 UDP-D-glucose epimerase의 뵙티드 비교 분석을 바탕으로 active site domain의 두 염기를 His-85 와 His-300으로 가정하였다. His85는 Leu로 His300은 Ala으로 site-directed mutagenesis 방법에 의해 변이 되었다. 이것의 효소 반응성을 분석한 결과 His85L은 반응성이 없고, His300A는 Wild-type와 같은 반응성을 보였다. 결과적으로 active-site domain의 두 염기중 하나의 염기는 His85L임을 밝혀냈다. dTDP-D-glucose 4,6-dehydratase as an oxidoreductase, is important enzyme to catalyze dTDP-D-glucose to dTDP-4-keto-6-deoxy-D-glucose. The reaction mechanism is proposed to oxidation, dehydration and reduction by two base mediating proton transfer and NAD' cofactor. The base in the active site domain were proposed to His-85 and His-300 from the comparision of the peptides of the dehydratase and UDP-D-glucose epimerase. His85L and His300A were mutated by the site directed mutagenesis. The analyzed protein acitvity showed that His 85L has no activity and His300A has a activity like as wild-type. This result show that the his-85 participates in the reaction mechanism with and the oxidation-reduction mechanism.
Microsurgical Treatment and Outcome of Pediatric Supratentorial Cerebral Cavernous Malformation

Objective : The purpose of this study was to investigate the clinical features and outcomes of pediatric cavernous malformation (CM) in the central nervous system. Methods : Twenty-nine pediatric patients with supratentorial CM underwent microsurgical excision. In selected cases, transparent tubular retractor system (TTRS) was used to reduce retraction injury and intraoperative neuromonitoring (IONM) was held to preserve functioning cortex. Patients' demographics and symptoms were reviewed and surgical outcomes were discussed. Results : The main initial clinical manifestations included the following : seizures (n=13, 45%), headache (n=7, 24%), focal neurological deficits (n=3, 10%), and an incidental finding (n=6, 21%). Overt hemorrhage was detected in 7 patients (24%). There were 19 children (66%) with a single CM and 10 (34%) children with multiple CMs. In 7 cases with deep-seated CM, we used a TTRS to minimize retraction. In 9 cases which location of CM was at eloquent area, IONM was taken during surgery. There was no major morbidity or mortality after surgery. In the 29 operated children, the overall long-term results were satisfactory : 25 (86%) patients had no signs or symptoms associated with CMs, 3 had controllable seizures, and 1 had mild weakness. Conclusion : With the assistance of neuronavigation systems, intraoperative neuromonitoring, and TTRS, CMs could be targeted more accurately and excised more safely. Based on the satisfactory seizure outcome achieved, complete microsurgical excision in children is recommended for CMs presenting with seizures but removal of hemosiderin-stained areas seems to be unnecessary.
Pancreatic Neuroendocrine Tumor Presenting with Arthritis and Panniculitis

Pancreatic neoplasm is complicated and can be preceded by extra-pancreatic manifestations, such as cutaneous and musculoskeletal symptoms. Awareness of these associations is important for timely diagnosis and appropriate treatment. We report a case of pancreatic neuroendocrine tumor (NET) presenting with arthritis and panniculitis. The patient had a two month history of right knee pain and subcutaneous nodules in both legs. Synovial fluid analysis from the right knee joint revealed a mildly increased white blood cell count without crystallization. A skin biopsy of a subcutaneous nodule revealed lobular panniculitis. The initial treatment with empirical antibiotics did not alleviate the symptoms; however, the right knee arthritis and skin nodules improved with steroid treatment. On the eighth day of hospitalization, the patient complained of abdominal discomfort. Abdominopelvic computed tomography scanning revealed a 14-cm sized pancreatic mass with peritoneal metastasis. Percutaneous needle biopsy confirmed the diagnosis of pancreatic NET. (J Rheum Dis 2017;24:313-317)

식물에게 있어 화분매개는 필수적인 요소 중 하나인데, 화분매개를 하는 식물 중 50%이상이 곤충에 의해 화분매개가 이루어지고 있다. 화분매개를 하는 곤충에 대한 조사는 주로 농업과 관련되어 있는 과수작물 주변의 화분매개곤충에 대해 조사가 되어있지만, 정작 양봉과 관련되어 있는 밀원식물 주변의 화분매개곤충은 조사된 바가 없다. 이에 연구진은 밀원식물 중 국내에서 가장 많은 양봉생산물을 만드는 아까시나무(Robinia pseudoacacia L.)의 개화시기에 맞춰서 화분매개곤충을 조사하였다. 조사지역은 총 6군데로, 백두대간을 중심으로 RCP 기후변화 시나리오에 의해 지정되었다. 조사 결과, 전체적으로 6목 60과 183종 1,555개체의 화분매개곤충이 채집되었다. 이중, 가장 많이 채집된 종은 노린재목의 애긴노린재(Nysius plebejus)로 약 21.30%가 채집되었다. 채집된 종을 군집분석한 결과, 강릉지역이 가장 안정적인 생태계를 유지하고 있으며, 완주지역이 가장 불완전한 생태계를 유지하고 있는 것으로 확인되었다.

Global navigation satellite systems (GNSS) use dual frequency signals to remove ionosphere delay effect. GNSS receivers have their own biases, called inter-frequency bias (IFB) between dual frequencies due to differential signal delays in receiving each frequency codes. The IFB degrades pseudo-range and ionosphere delay accuracies, and they must be accurately estimated. Simultaneous estimation of ionosphere map and IFB is applied in order to analyze the IFB estimation accuracy and variability. GPS network data in Korea is used to compute each receiver`s IFB. Accuracy changes due to ionosphere model changes is analyzed and the effect of external GNSS satellite IFB on the receiver IFB is analyzed.
Molecular and neural mechanisms on the avoidance behaviors of Culex pipiens to ultrasonic sounds

Mosquitoes are transmit many dangerous disease such as malaria, yellow fever and dengue fever. So far, chemical insecticides such as DEET have been mainly used to control mosquitoes, but there are many side effects. This study used ultrasonic sounds as an alternative to chemical insecticides. We found that Culex pipiens, which are common in Korea, exhibit avoidance behavior in a specific ultrasonic frequency. Through electrophysiological recording, we have inferred that avoidance behavior is caused by different from each other mechanisms depending on the ultrasonic frequency. Using immunohistochemical staining, we analyzed the expression pattern of auditory related genes in the chordotonal organ. Quantitative real time-PCR was used to compare the expression levels of auditory related gene depending on the time of exposure to ultrasonic sounds.
