      • Performance advances in ion mobility spectrometry through combination with high speed vapor sampling, preconcentration and separation trchniques

        Rugged, low weight, hand-held ion mobility spectrometry devices, initially developed far chemical warfare detection purposes, possess attractive characteristics as field-portable instruments for paramilitary (treaty verification, chemical demilitarization, drug interdiction, counterterrorism operations) and civilian (environmental monitoring, forensic characterization, process control) applications. Generally, However, such devices tend to exhibit limited resolution, narrow dynamic range, nonlinear response and long clearance times which severely limit their usefulness for qualitative and quantitative analysis of mixtures. To overcome these restrictions a prototype combined gas chromatography-ion mobility spectrometry (GC-IMS) unit was constructed by replacing the membrane inlet of a military IMS device known as the CAM (chemical agent monitor) with suitable front-end modules. These modules enable high speed automated vapor sampling (AVS), microvolume preconcentration/thermal desorption, and isothermal GC preseparation of analytes using a short capillary column while operating the BMS source and cell at subambient pressures as low as 0.5 atm. The AVS-GC-lMS methodology sharply reduces competitive ionization and facilitates identification of mixture components, thereby enabling quantitation of volatile and semivolatile com-pounds over a broad range of concentrations in air, At higher concentration levels (c.g. > 1 ppm) using the AVS inlet in automatic attenuation control (AAC) mode maintains excellent linear response. At ultralow concentration levels, e.g. < 10 ppb, a microvolume, trap-and-desorb type preconcentration module, maintains adequate signal to noise levels, thereby expanding the effective dynamic range of the method to approπ. 6 orders of magnitude (100 ppt-100 ppm). The resulting "hyphenated" GC-IMS technique has the potential of evolving into the first hand-portable, combined chromatography-spectroscopy instruments for field screening applications.

        Exploring spatial patterns of trends in monthly rainfall and temperature in the Philippines based on Climate Research Unit grid

        This study assessed spatial pattern of trends in monthly rainfall and temperature in the Philippines using Climate Research Unit time series data. Based on the results, there are significant trends in monthly rainfall and temperature in the country. On the average, monthly rainfall in the country is increasing by 0.34 mm/year. In the case of monthly temperature, the average increases per year were 0.008 and 0.019 C, for maximum and minimum temperature, respectively. In terms of proportion, larger portion of the country showed significant trends in monthly temperature ([80%) compared to rainfall (\10%). Shift in wettest, driest, warmest, and coldest months were also observed between the periods of 1951–1980 to 1986–2015.

        Banana suitability and Fusarium wilt distribution in the Philippines under climate change

        Climate change is expected to affect crop production directly and indirectly due to changes in crop suitability, decrease in productivity, and higher incidence of pest and diseases. In the case of banana, change in suitability and distribution of Fusarium wilt due to climate change can pose a major threat on its production system. Being a major dollar earner for the Philippines, these threats can greatly affect the country’s economic and food production system. This study assessed banana suitability and the potential distribution of Fusarium wilt in the Philippines under current and future climate condition using fuzzy logic and maximum entropy approach. Based on the results, climate change might have limited impact on banana suitability in the country. But the projected changes in rainfall in the future can increase the areas that are favorable for Fusarium wilt occurrence. From 21% under baseline climate condition, favorable areas for Fusarium wilt in the Philippines is estimated to increase to 27% covering 91.2% and 28.5% of the country’s highly and moderately suitable areas for banana, respectively. Such coverage accounts for approximately 67% of the country’s total harvested area for banana.

      • Real-time Vehicle Classification on Phnom Penh Road in Video Surveillance

        In this work we tackle traffic analysis problems for Phnom Penh roads. Unlike in most developed countries such as Australia and USA, the traffic conditions in developing countries such as Cambodia pose unique challenges. For instance, motorcycles are popular vehicles. This means, during peak hours, where the traffic for cars is congested, the traffic for motorcycles could still be flowing. Thus, any approaches using the whole video to determine the traffic condition are not suitable. To that end, this work provides the first stepping-stone by addressing the Phnom Penh vehicle classification problem. We propose a novel dataset capturing CCTV of Phnom Penh roads in 5 locations from 7am until 5pm on five sunny days. Then, we perform the study by proposing a baseline method comprising three steps: (1) background subtraction; (2) bag-of-word histogram feature extraction; and (3) classification using Support Vector Machine (SVM). Evaluation results show promising results with accuracy 0.76, 0.63 and 0.40 for light, medium and heavy traffic.

        Nutritional Studies on Production of Antibacterial Activity by the Zebra Mussel Antagonist, Pseudomonas fluorescens CL0145A

        Pseudomonas fluorescens strain CL0145A was discovered at the New York State Museum Field Research Laboratory as an effective agent against the environmentally destructive zebra mussel, which has contaminated US waters. Dried cells of the microbe are being commercialized as an environmentally friendly solution to the problem. We found that antibiotic activity against the Gram-positive bacterium Bacillus subtilis is produced and excreted by this strain. We have carried out studies to optimize production of the antibiotic. Studies were begun in a complex corn meal medium. Activity was found in both cells and culture supernates and was maximal after one day of fermentation. Static fermentation conditions were found to be superior to shaken culture. Production of extracellular antibiotic in complex medium was found to be dependent on the content of sucrose and enzymehydrolyzed casein. Indeed, production was greater in sucrose plus enzyme-hydrolyzed casein than in the complex medium. Of a large number of carbon sources studied as improvements over sucrose, the best was glycerol. An examination of nitrogen sources showed that production was improved by replacement of enzymehydrolyzed casein with soy hydrolysates. Production in the simple glycerol-Hy-Soy medium was not improved by addition of an inorganic salt mixture or by complex nitrogen sources, with the exception of malt extract. In an attempt to keep the medium more defined, we studied the effect of amino acids and vitamins as replacements for malt extract. Of 21 amino acids and 7 vitamins, we found tryptophan, glutamine, biotin, and riboflavin to be stimulatory. The final medium contained glycerol, Hy- Soy, tryptophan, glutamine, biotin, and riboflavin.

        Quantitative Skeletal Muscle MRI: Part 1, Derived T2 Fat Map in Differentiation Between Boys With Duchenne Muscular Dystrophy and Healthy Boys.

        <P>OBJECTIVE. The purpose of this study was to validate derived T2 maps as an objective measure of muscular fat for discrimination between boys with Duchenne muscular dystrophy (DMD) and healthy boys. SUBJECTS AND METHODS. Forty-two boys with DMD (mean age, 9.9 years) and 31 healthy boys (mean age, 11.4 years) were included in the study. Age, body mass index, and clinical function scale grade were evaluated. T1-weighted MR images and T2 maps with and without fat suppression were obtained. Fatty infiltration was graded 0-4 on T1-weighted images, and derived T2 fat values (difference between mean T2 values from T2 maps with and without fat suppression) of the gluteus maximus and vastus lateralis muscles were calculated. Group comparisons were performed. The upper limit of the 95% reference interval of T2 fat values from the control group was applied. RESULTS. There was no significant difference in age or body mass index between groups. All healthy boys and 19 boys (45.2%) with DMD had a normal clinical function scale grade. Grade 1 fatty infiltration was seen in 90.3% (gluteus maximus) and 71.0% (vastus lateralis) of healthy boys versus 33.3% (gluteus maximus) and 52.4% (vastus lateralis) of boys with DMD. T2 fat values of boys with DMD were significantly longer than in the control group (p < 0.001). Using a 95% reference interval for healthy boys for the gluteus maximus (28.3 milliseconds) allowed complete separation from boys with DMD (100% sensitivity, 100% specificity), whereas the values for the vastus lateralis (7.28 milliseconds) resulted in 83.3% sensitivity and 100% specificity. CONCLUSION. Measurement of muscular fat with T2 maps is accurate for differentiating boys with DMD from healthy boys.</P>
