Nowadays, Magnoliae Cortex used herbal medicine differ extremely significantly by region, leaf shape, DBH, tree height, crown size, age, bark thickness, color of bark powder, oiliness, grounding nature, bark type, position of sampling, etc., of which region, leaf shape, powder color, bark thickness and DBH are the most influential factors.
In this study, we analyzed and quantified the hexane fraction from the leaves of Magnolia species by HPLC. HPLC methods for the determination of the contents of magnolol (Ⅰ), honokiol (Ⅱ), obovatol (Ⅲ) of the hexane fraction from the leaves of Magnolia species were developed and validated by ICH guide lines comparing the linearity, intraday precision, inter-day precision. Those compounds were detected with optimal condition; wavelength: 288 nm, column: Zorbax Eclipse XDB-C18 (2.1 × 150mm, 5 μm), mobile phase: Acetonitril - 0.1% H3PO4 (45 : 55, v/v) buffer, flow rate: 0.5 ml/min.
The leaves of M. officinalis contained 1.08% Ⅰ, 2.37% Ⅱ, 0.57% Ⅲ and that of M. biloba contained 0.50% Ⅰ, 0.03% Ⅱ, 1.54% Ⅲ and that of M. obovata contained 0.37% Ⅰ, 0.03% Ⅱ, 0.76% Ⅲ.
The contents of effective ingredients were higher in the leaves of M. officinalis and M. biloba than M. obovata. Especially, the content of obovatol in the leaves of M. biloba was high remarkably. This show that the shapes of leave affect the contents of obovatol.
