Parabens are substances that inhibit and reduce the growth of harmful bacteria and fungi and are widely used worldwide as preservatives in cosmetics and food products due to their odourless and tasteless properties, high chemical stability and low production costs. However, a number of studies have reported adverse effects on human health through endocrine disruption. In addition, several previous studies have raised concerns about the neurotoxicity of parabens, but the overall understanding of this issue is still incomplete. In this study, we aimed to analyze the multiple in vivo toxicity of parabens and to investigate their association with the development of neurodegenerative diseases using C. elegans as an experimental model. Methylparaben and butylparaben, the two most commonly used parabens, were selected for analysis. The results showed growth retardation, motor impairment, dopaminergic neuronal damage and altered alpha-synuclein aggregation in the paraben-treated group, and the expression changes of the genes involved confirmed that parabens cause multiple toxicities. Furthermore, as dopaminergic neuronal damage and alpha-synuclein aggregation are typical hallmarks of Parkinson's disease patients, this study is ultimately expected to contribute to the elucidation of the in vivo multi-toxicity of parabens and to elucidate the link between parabens and Parkinson's disease.
