Five cosmid clones containing the biosynthetic gene cluster of the ansamycin antibiotic rubradirin jointly extended over a 165 kb which was restriction enzyme mapped. Approximately 90 kb of the DNA were sequenced and 45 open reading frames were identified. Seven genes known to be involved in the biosynthesis of 3-amino-5-hydroxybenzoic acid (AHBA) were identified and showed 50 to 70% identity in amino acid sequence to the proteins reported for the AHBA biosynthesis of the rifamycin. Two genes were determined to encode the modular type I PKS that uses AHBA as the starter unit and contains six modular sets. Genes reported to be involved in deoxysugar biosynthesis and regulatory functions were also identified.
