I. 연구개발의목적 및 필요성
- 인체의 뼈는 조골세포에 의해서 형성되고 파골세포에 의해서 파괴되는 과정이 균형을 이뤄 평생 동안 유지됨. 최근 우리나라는 고령인구의 증가 및 식습관의 변화로 골다공증이 증가되고 있음. 특히 활동이 적은 노인에게서 골다공증이 빈번히 나타나지만 골다공증과 운동의 영향 연구는 거의 없음. 따라서 본 연구는 immbobilization 마우스 모델을 이용하여 운동부족이 ①뼈 밀도에 미치는 영향 규명 ②파골세포 및 조골세포의 분화와 기능에 미치는 영향을 검증 ③운동 부족으로 인한 뼈 밀도 감소 원인 인자 규명 및 작용기전 연구로 뼈 밀도 및 골다공증에 운동의 영향을 정량화 하고 원인을 규명하여 골다공증을 예방할 수 있는 단서를 제공하는 것이 궁극적인 목표임.
II. 연구개발의 내용 및 범위
o 골다공증과 운동의 상관관계 규명
- 새로운 immobilization 동물 모델 확립
- immobilization이 뼈 밀도에 미치는 영향 확인
o immobilization 동물 모델에서 조절되는 유전자 발굴
- immobilization 동물 모델에서 파골세포 관련 유전자의 발현 확인
- immobilization 동물 모델에서 조골세포 관련 유전자의 발현 확인
- immobilization에 의해 조절되는 후보 유전자로서 HO-1의 발현 확인
o 파골세포에서 HO-1의 영향 확인
- HO-1 과발현 및 knock-down 후 파골세포 분화 확인
- HO-1 과발현 및 knock-down 후 파골세포 관련 유전자의 발현 확인
o 조골세포에서 HO-1의 영향 확인
- HO-1 과발현 및 knock-down 후 조골세포 분화 확인

III. 연구개발 결과

o HO-1 siRNA에 의한 파골세포 분화 증가 확인
o HO-1 과발현에 의한 파골세포 분화 감소 확인
o HO-1 과발현에 의한 NFATc1의 발현 억제 확인
o HO-1이 c-fos의 전사활성을 억제함으로써 NFATc1의 발현을 억제할 가능성 확인
o HO-1의 과발현 및 knock-down은 조골세포의 분화 및 기능에는 별다른 영향을 미치지 않음
IV. 연구개발결과의 활용계획
- 골다공증은 노화와 관련된 주요 질환 중 하나로 파골세포와 조골세포의 불균형에 의해 야기되는 질환임. mechanical stimuli의 결여가 증가된 골흡수와 감소된 골형성을 야기함이 알려져 있지만, 어떤 조절기전을 통해 작용하는지는 잘 알려져 있지 않음.
본 연구는 mechanical stimuli가 골밀도에 어떻게 작용하는지에 관한 기초적 지식을 제공하고, 더 나아가서는 골다공증 치료제 개발을 위한 중요 정보를 제공할 수 있음

I. Purpose
o Disuse osteoporosis occurs commonly in elderly population. However, the mechanisms underlying such unloading-induced bone loss have not yet been fully understood. Thus, the purpose of our study is ① to develop a new immmmobilization mouse model in order to verify involvement of disuse in reduction of bone density, ② to identify the effects of immobilization on differentiaion and function of the osteoclast and osteblast, ③ to find a gene which is able to regulate bone mass in an immobilization model. We will try to find the action mechanism of disuse osteoporosis and offer the basic information for a therapeutic agent development for osteoporosis.
II. Experimental contents
o Study the effects of prolonged lack of movement on bone mass
- develop a new immobilization mouse model
- evaluate the effects of immobilization on the bone density using the new developed immboilization mouse model
o Study the effects and action mechanism of the gene regulated by immobilization-mediated bone loss
- screen the differently expressed genes between control mouse and immobilization mouse
- evaluate the role of HO-1 in osteoclast differentiation using overexpression or knock-down system.
o Study the effects of HO-1 on differentiation and bone nodule formation of osteoblast.
- evaluate the effects of HO-1 on osteoblast differentiation using overexpression or knock-down system.
- examine the effects of HO-1 on bon nodule formation using overexpression or knock-down system.
III. Experimental results
o established a new immobilization mouse model with low bone mass
o Prolonged lack of movement leads to osteoporosis
o Immobilization mouse showed osteopenic bone mass parameters after 15 days of immobilization
o The expression levels of osteoclast-related genes were increased and osteoblast-related genes were decreased in immobilization mouse
o The expression level of HO-1 was decreased in immobilization mouse
o siHO-1 increased RANKL-induced osteoclast differentiation
o Overexpression of HO-1 inhibited RANKL-induced osteoclast differentiation
o HO-1 inhibited NFATc1 expression by suppressing c-Fos transcriptional activity
o Knock-down or overexpression of HO-1 failed to regulate osteoblast differentiation.
IV. Expected contribution
This study will help to understand the biological function of mechanical stimuli (movement) for bone metabolism maintained by osteoclasts and osteoblasts and offer the information of therapeutic agents development for osteoporosis
