Prostate, whether benign or malignat, is hormone, especially androgen dependant and various kninds of hormones are used for the treatment of prostatic canecer. Harris' report on hypotalamo-pituitary gonadal axis in 1949, the hypothalamus had been believed as the exclusive organ producing GnRH. After Gibbon's report on the detection of GnRH or GnRH like substance from the placenta, many reported the detection of the GnRH or GnRH-like material from various human and animal organs meaning some organs produce the extrahypothalamic GnRH or GnRH like materials.
In theis study, author tried to confirm the production of the GnRH and GnRH receptor from the human prostate tissue and prostate cancer cell lines and localized them electron microscopically to access the function of the prostatic GnRH.
In this study, three normal human prostate tissues, three human prostate cancer tissues and four prostate cancer cell lines, LNCaP, DU-145, PPC-1 and TSU-Pr1 were used. For the detection of the expression of GnRH and GnRH receptor mRNA, In Situ Hybridization histochemistry using avidine-biotin complex method were used for individual prostate cancer cells. For the detection of the GnRH and GnRH receptor produced from prostate cancer cells, the immunohistochemical electorn microseopy(EM) was done.
The GnRH and GnRH receptor mRNAs were expressed in almost all examined prostate cancer cells. The signals of the GnRH receptor mRNA were more than those of GnRH. The GnRH were localized within nucleus and cytoplasm and GnRH receptors were localized at the nuclear membrane at EM.
Author hypothesize the role of prostatic GnRH with previous results as follow Among the GnRHs which produced from the cytoplasm of the prostatc cancer cells, some would be secreted into the outside of the cell and others would be transferred into the nucleus after complexing with GnRH receptors located at the nuclear membrane and translocated into the nucleus. The GnRH- GnRH receptor complex in nucleus would be separated by unknown mechanism With this hypothesis, author suppose that the role of the prostatic GnRH will be a intracrine.
Author concludes that the prostate cancer produces the GnRHs and GnRH recepfors and the prostate cancer cells have the independent exrtahypothalamic GnRH system.
