Ⅰ. 연구개발의 목적 및 필요성: Odin 유전자의 발현이 제거된 유전자 적중 마우스 모델과 Odin의 dominant negative mutant인 Odin-dPTB가 뇌실막 세포에서 특이적으로 발현하는 형질전환마우스등을 질환동물모델로 활용하여 Odin의 기능이 뇌실막세포의 분화에 미치는 영향을 분석한다. 그 결과 선천성 뇌수종 발병의 원인기전을 이해하고 뇌수종 예방에의 활용성 가능성을 제시하고자 한다.
Ⅱ. 연구개발의 내용 및 범위
1차년도 연구내용: Odin-dPTB 형질전환 마우스모델을 이용한 Odin 유전자의 뇌실막 세포에서의기능 규명을 통하여 뇌수종 발병의 원인을 분석한다.
2차년도 연구내용: Odin knock-out 마우스와 Odin-PTB 발현 형질전환 마우스 모델을 이용하여 Odin 유전자의 기능을 규명하고 뇌수종 발병의 원인을 분석한다.
3차년도 연구내용: Odin의 프로모터에 의하여 Cre-ER을 발현하는 형질전환 마우스를 이용하여 reporter 발현을 뇌실막세포에서 유도하는지 분석하고 Odin 유전자와 상호작용하는 유전자들의 연구 기반을 구축한다.
Ⅲ. 연구개발결과
Odin-dPTB 마우스는 주로 중뇌에서 발현을 유도하였으며, 그 결과 중뇌에 있는 SCO 발달이 잘 되지 않고, ependymal cell들의 발달이 원활하게 일어나지 않아서 뇌수종이 발병하는 것으로 증명되었음. Odin-KO 마우스는 대뇌의 ependymal cell의 분화가 적절하게 일어나지 않고 type B세포상태로 미분화되어 있고 cilia 형성도 원활하게 일어나지 않았기 때문에 뇌수종이 발병되는 것으로 증명되었음. Odin-CreER 마우스는 tamoxifen 투여에 따라 reporter발현이 뇌실막세포에 특이적으로 유도됨을 증명하였음.
Ⅳ. 연구개발결과의 활용계획
1. 다수의 뇌수종 마우스 동물 모델은 이 분야 연구 활성화에 기여할 것임.
2. Odin 유전자의 기능과 메커니즘이 밝혀짐에 따라 뇌수종의 정확한 예방과 진단에 활용될 것임.
3. 뇌실막세포의 다양한 바이오 마커 개발과 뇌실막 세포 특이적 적중마우스 제작에 기여할 것임.

I. Research Purpose: This research utilizes two different mouse disease models, Odin null mutant mouse and Odin-dPTB expressing transgenic mouse, to determine the functional role of Odin in the proper differentiation of ependymal cells. This research is aimed to understand the etiology of congenital hydrocephalic brain disease and suggest its application for preventing the development of the congenital hydrocephalus.
II. The Contents of Research Project
First year: We aimed to intensively characterize Odin-dPTB transgenic mice to investigate whether Odin has an essential role in the development of congenital hydrocephalus through interfering with the proper differentiation of ependymal cells. Second year: We aimed to intensively characterize Odin null mutant mice to investigate whether Odin has an essential role in the development of congenital hydrocephalus through interfering with the proper differentiation of ependymal cells. Third year: We aimed to characterize Odin-CreER transgenic mice to investigate whether reporter expression is specifically induced in the ependymal cells. In addition, this mouse model will be useful to conditionally delete the genes related with the development of the congenital hydrocephalus.
III. Result
In vivo expression of Odin-dPTB was specifically induced in the midbrain. As a result, the SCO agenesis and immature development of ependymal cells were observed, indicating that Odin has an essential role in the development of hydrocephalus. Odin-KO mice was extremely useful to demonstrate that the ependymal cells in the lateral ventricle were not fully differentiated and remained as type B-like cells, the major cause of hydrocephalus. Lastly, Odin-CreER mice was shown to induce LacZ reporter expression in response to tamoxifen, indicating that this mouse line will be useful for the research of hydrocephalus.
IV. Application
1. The mouse models will contribute to better understanding of the hydrocephalus etiology.
2. Our research will contribute to developing the prevention and diagnosis of the congenital hydrocephalus.
3. Our research will be useful for the development of various biomarkers related with hydrocephalus and conditional knock-out mouse research.
