Complementation cloning of the hisI gene in Corynebacterium glutamicum was done by transforming the genomic DNA library into the corresponding histidine auxotrophs of Escherichia coli. Recombinant plasmids containing 7.8 kb and 6.0 kb fragments were also able to complement the E. coli hisI mutant. The insert DNA fragments in the recombinant plasinids, named puIl and pHI2, were physically mapped with several restriction enzymes. By further subcloning the entire DNA fragment containing the function and by complementation analysis, we located the hisI gene of C. glutamicum on the 1.8kb PstI-HindIII pHI12 fragment.
