The aim of this study was to evaluate new oral insulin preparation using newly developed drug absorbtion enhancer(Procarrier). Rats was divided into two groups, one of them was normal control and the other was diabetic disease model. Each group was administered with insulin or insulin/Procarrier solution and then we measured serum insulin level and blood glucose level using radio immunoassay and glucomerter, respectively. Insulin/Procarrier significantly enhanced the oral absorbtion of insulin compraed to insulin. Moreover, Insulin/Procarrier effectively decreased blood glucose level in diabetic disease model. These results suggest that Insulin/Procarrier may be effective in the treatment of diabetic mellitus via the increase in the oral absorbtion of insulin. In contrast, Insulin/Procarrier did not affect blood glucose level in normal control with stable blood glucose level after Insulin/Procarrier administration. This finding indicate that there may be little risk of hypoglycemia with Insulin/Procarrier. The hypoglycemia is the most common adverse effect of oral insulin preparations. In conclusion, Insulin/Procarrier with economic cost and convenience in use might lead to new drug delivery system of oral insulin or other peptide drugs.
