Abstract : Polymerase chain reaction (PCR) is the one of the most widely used molecular biological techniques because of it's simplicity, flexibility, sensitivity, and robustness. Although the principle of the PCR is conceptually simple, many attempts to perform PCR fail to yield meaningful results, In those cases of PCR failure, performing control reactions will establish whether or not the PCR conditions are functioning appropriately. PCR optimization and other efforts to seek the causes of the PCR problems and adequate treatments for them will lead to PCR success.
Optimization of PCR conditions includes design of nice primers, and fine tuning of the concentrations of MgCl₂, annealing temperature, pH of the reaction, dNTP, and Taq polymerase. For troubleshooting of PCR, some commonly observed problems in the period of setting up PCRs for the first time or in amplification reactions were described. Also the explanation about the possible causes and suggested remedies for them were provided, In addition, methods for improving PCR such as hot start PCR, nested PCR, touch down PCR, and booster PCR were introduced.
