    HDAC6 is a class IIb histone deacetylase and deacetylates lysine residues in non-histone proteins. HDAC6 is primarily expressed in the cytoplasm and acts as a regulator of microtubule-mediated processes such as cell migration, autophagy, stress granule formation, and misfolded protein degradation. HDAC6 has been proposed as a therapeutic target for various cancers, neurodegenerative diseases, and innate immune responses. However, the molecular mechanism of HDAC6 in hypersensitivity immune reactions has not yet been identified. To determine the role of HDAC6 in allergic inflammation, two models of allergic inflammation were used: anaphylaxis and atopic dermatitis.
    In anaphylaxis models, early growth response 3 gene (EGR3) is most expressed in antigen-stimulated RBL2H3 cells and regulates the expression of HDAC6 by binding to the HDAC6 promoter. HDAC6 mediates in vitro allergic inflammation reactions and in vivo anaphylaxis models. In anaphylaxis models, IL-27 is an inflammatory cytokine directly regulated by HDAC6 and mediates allergic inflammation in vitro and in vivo. In the inflammatory tumor microenvironment, IL-27 is present in exosomes secreted from activated mast cells and mediates intercellular interactions to promote tumorigenic and invasive ability of cancer cells and TAM polarization of macrophages.
    Atopic dermatitis is a chronic allergic inflammatory disease mediated by mast cells, immune cells, and skin component cells. In atopic dermatitis, inhibition of HDAC6 attenuates the symptoms of atopic dermatitis and reduces the recruitment of mast cells to the inflammatory site and the expression of inflammatory mediators in tissues. HDAC6 regulates the expression of sirtuin 1 (SIRT1) by regulating the expression of miR-9. SIRT1 and miR-9 regulate autophagy and reactive oxygen species (ROS) generation accompanied by AD in keratinocytes. CXCL13 is an inflammatory cytokine and regulated by HDAC6 in atopic dermatitis. CXCL13 mediates skin inflammation and is present in exosomes secreted by activated skin mast cells. CXCL13 regulates the intercellular interactions among skin mast cells, keratinocytes, and dermal fibroblasts, and acts as a major mediator of atopic dermatitis.
    Taken together, these findings indicate that the EGR3-HDAC6-IL27 molecular network modulates allergic inflammatory responses and the inflammatory tumor microenvironment, and HDAC6 mediates atopic dermatitis by regulating miR-9 and CXCL13. This provides evidence that HDAC6 can be a target for anti-allergy or anti-atopic drugs by understanding the molecular networks of allergic inflammation and atopic dermatitis.

HDAC6 is primarily expressed in the cytoplasm and acts as a regulator of microtubule-mediated processes such as cell migration, autophagy, stress granule formation, and misfolded protein degradation. HDAC6 has been proposed as a therapeutic target for various cancers, neurodegenerative diseases, and innate immune responses. This provides evidence that HDAC6 can be a target for anti-allergy or anti-atopic drugs by understanding the molecular networks of allergic inflammation and atopic dermatitis.

    HDAC6는 히스톤 탈 아세틸화 효소 중 하나로서, 비히스톤 단백질의 탈 아세틸화를 통해서 다양한 생물학적 과정을 조절함이 알려져 있다. HDAC6는 주로 미세소관이 매개하는 생물학적 과정의 주 조절자로서 작용하며, 다양한 암 질환, 신경 퇴행성 질환 및 선천적 면역 반응의 치료 표적으로서 제시된다. 본 연구에서는 과민성 면역반응인 알레르기성 염증 및 아토피 피부염에서 HDAC6의 역할 및 분자적 조절 메커니즘을 밝히기 위해 연구를 수행하였다.
    RBL2H3 세포에 항원을 자극하여 알레르기성 염증 반응을 유도한 뒤, 가장 많이 발현이 변화하는 유전자를 RNA-seq 및 차등 발현 유전자(DEG) 분석을 통하여 발굴하였다. EGR3는 유사 분열 자극에 의해 유도되는 초기 성장 반응 유전자 패밀리 중 하나이며, 전사 조절자로서 널리 알려져 있다. 본 연구에서는 EGR3가 HDAC6유전자의 프로모터에 결합하여 HDAC6의 발현을 전사 수준에서 조절함을 밝혔다. EGR3는 HDAC6의 발현을 조절함으로써 시험관 및 생체 내 아나필락시스 모델을 매개하는 주 조절자로서 작용하였다. 아나필락시스 모델에서 HDAC6의 조절은 알레르기성 염증반응에 의해 증가되는 사이토 카인인 IL-27의 발현을 조절하였다. IL-27은 염증성 또는 항 염증성 성향을 가지고 있는 사이토 카인으로, 면역계에서T세포의 분화를 유도하며 IL-10의 발현을 조절함이 알려져 있다. 본 연구에서 IL-27은 항원에 의해 발현이 중가 하며, 시험관 및 생체 내 알레르기 염증반응을 매개하였다. 또한, 암세포의 종양 형성 및 침습 능력과 대식세포의 분극화를 조절함으로써 염증성 종양 미세환경의 주요 조절 매개체로 작용하였다.
    만성 알레르기성 염증질환 모델인 아토피 피부염은 각질세포, 진피 섬유아세포 및 비반 세포를 포함하는 다양한 면역세포들이 관여하는 염증반응으로 심한 가려움증, 피부 건조증 및 습진 등의 증상이 유발되는 대표적인 알레르기 질환이다. 본 연구에서는 아토피 피부염 모델에서 아토피 피부염 유발물질인 DNCB에 의해 HDAC6의 발현이 유도되며, HDAC6의 작용 및 발현 저해는 아토피 피부염의 임상 증상을 약화시키고 피부 염증 부위로 비반 세포 및 면역 세포의 모집을 감소시킴을 밝혔다. MicroRNA array 및 Cytokine array를 통한 HDAC6의 분자적 기전 연구는 HDAC6가 miR-9의 발현을 조절하여 SIRT1의 발현을 직접적으로 조절하며, 염증성 사이토 카인인 CXCL13의 발현 및 분비를 조절함을 밝혔다. MiR-9는 SIRT1을 표적으로 조절하여 아토피 피부염 증상을 약화시켰고, CXCL13은 아토피 피부염을 매개하는 주 조절자로서 작용하였다. 또한, SIRT1과 miR-9는 각질세포에서 손상으로 인하여 발생하는 자가포식 반응을 조절하고, CXCL13은 비반 세포에서 분비하는 엑소좀에 존재하여 비반세포, 대식세포, 각질세포 및 진피 섬유아세포 사이의 세포간 상호작용을 매개함으로써 아토피 피부염의 주 매개자로서 작용하였다.
    본 연구에서는 알레르기성 염증 및 염증성 종양 미세환경에서 EGR3-HDAC6-IL27 분자 네트워크와 아토피 피부염에서 miR-9 및 CXCL13을 조절하는 HDAC6의 역할을 조사하였다. 이러한 발견은 HDAC6이 항 알레르기 또는 항 아토피 약물의 표적이 될 수 있음을 시사한다.
