본 연구과제는 스포츠 현장 및 일반 대중들이 운동과 식이조절을 통해 주로 행하고 있는 다양한 체중조절 방법이 생체 내 생리 스트레스와 대사기전에 어떠한 영향을 미치는가를 알아보기 위해 시행되었다. 이를 위해 1차 년도 연구에서는 실험동물을 이용하여 단기간의 극단적인 식이제한과 운동부하에 대한 연구를 시행하였으며, 2차 년도 연구에서는 6주간의 반복적인 식이제한 및 운동부하 병행 적용을 통해 그에 따른 대사 스트레스 반응 및 대사성 증후군 위험요인과의 상관성을 살펴보았다. 또한 1, 2차 년도의 연구결과를 토대로 3차 년도에서는 실제 임상실험을 적용함으로써 식이를 극단적으로 제한하는 단기간의 절식 후와 장기간의 회복기에 대한 체중감량 효과와 생리 변화를 대사적·분자생물학적 분석을 통해 살펴보았다.
그 결과, 1차 년도 연구(동물실험)에서 일시적이고 극단적 절식에 의해 혈액성분 중 혈당, 인슐린은 운동부하 및 절식기간에 비례하여 매우 강한 정적인 상관성을 확인할 수 있었으며, 유리지방산의 경우 절식유무에 대해서만 유의성을 확인 할 수 있었다. 또한 대사스트레스 반응 단백질 분석결과 특이적으로 ATF4 에서만 운동부하 및 절식시간에 비례한 유의적 상관성을 확인 할 수 있었고, 세포사멸 및 염증관련 단백질에서는 CHOP와 IL-1β에서만 운동부하 및 절식시간에 비례한 유의적 상관성을 확인 할 수 있었다.
2차 년도 연구(동물실험)에서 반복적 절식으로 인해 혈중 유리지방산의 경우 정상운동군군에서 가장 낮은 것으로 나타났으며, 절식군에서 가장 높은 수치를 보여주었으며, 혈중 인슐린의 경우 반복절식 및 운동병행군에서 가장 높은 수치를 보여주었다. 한편, 대사스트레스 반응 단백질의 경우 Bip의 경우 반복절식군과 운동병행군에서 모두 유의한 발현증가를 보여주었으며, ATF4과 PERK mRNA의 경우 반복절식 및 운동병행군에서 가장 높은 발현을 보여주었다. 또한 세포사멸 및 염증관련 단백질 분석결과 CHOP만이 이와 같은 양상으로 보여주어 조직내 PERK mRNA와의 연관성이 제기되었다. 또한 골격근 지방산화율 분석 결과 운동군에서 유의한 지방산 산화율 증가양상을 보여주어 반복절식 및 운동병행군이 다른 실험집단에 비해 가장 높은 산화율의 증가현상을 보여주었다.
3차 년도 연구에서는(임상실험) 1일간의 감식기와 10일간의 절식기 및 신체활동 적용에 따라 체중, 체지방율, 복부지방에서 감량기에 해당하는 절식기 10일에서 감량전에 비해 유의한 감소가 나타났으며 회복기 10주 후에도 체중 및 체지방에서 유의한 감소를 보여주었다. 또한 혈액성분 분석결과 혈중 중성지방(TG), 유리지방산(F

This study was carried out to investigate the effect of diverse body weight control methods on the physiological stress and metabolic pathway in the sports field and general public through exercise and dietary control.
