Although D(-)-α-aminobenzyl penicillin(ampicillin) is relatively stable to acid it is absorbed inefficiently by the oral route as indicated, e.g., by the fact that much higher blood levels and urinary recovery are attained after parenteral than after oral administratration.
Of a series of acyloxymethyl esters of ampicillin which are more completely absorbed than the parent compound, the hydrochloride salt of pivampicillin was selected for extended studies. Comparative studies of pivampicillin and smpicillin in 20 healthy rabbits revealed that pivampicillin is better absorbed from the gastro-intestinal tract than ampicillin, as reflected in considerably higher peak ampicillin concentration in blood.
