The effectiveness of ginseng saponin on the extraction of an electron carrier, cytochrome b?, from the outer membrane of bovine heart mitochondrion was compared with those of digittonin and dodecyl maltoside.
The mitochondrial outer membrane treated with 1% digitonin or dodecyl maltoside released a lot of cytochrome b? into the supernatant after centrifugation, whereas the same membrane treated with 2% or 5% ginseng saponin did not.
DEAE-cellulose colunn chromatography was performed for the cytochrome b?-containing supernatant solution.
A single band of pure cytochrome b? was obtained after SDS-polyacrylamide gel elctrophoresis of the eluent, and its molecular Weight was estimated to be 13,000∼20,000.
