Heparin anticoagulation, is utilized during and after cardiopulmonary bypass procedures in experimental animals to reduce the rlsk of acute or chronic thromboembolic problem. A total of 6 healthy sheens were used and assigned to 3 groups (A, B, and C) according to their body weight. Body weight of group A, B and C was less than 40kg, 40 to 80kg and more than 80kg, respectively. All animals were treated with heparin (300 lU/kg BW) through two routes, IV bolus or SC injection and then measured for APTT value, platelet count, fibrinogen concentration before and every hour after treatment.
After IV bolus and SC injection, APTT values increased significantly (P<0.05) and then returned baseline values 6 and 10 hours, respectively. The APTT values of group C was consistently higher than those of group A, B after heparin treatment by two routes. After IV bolus and SC injection, APTI ratio became sub-therapeutic at 5, 7 hours, respectively, and in the time maintaining APTT ratio's therapeutic range of IV and SC group was 1, 6 hours, respectively. After SC injection of heparin, the highest APTT ratio of group B, C significantly higher (P<0.05) than that of group A. Mean platelet count of group A, B and C before the injection was 337.4(31.2, 285.1(7.1, and 197.5(5.5 103/㎕, respectively. In the SC group showed the significant decrease (P<0.05) 2, 6, 9 hours after heparin injection, but that In the IV bolus group gradually decreased without significant variation. There were no significant differences in the fibrinogen between the treatments used.
From the above results we obtained elementary data for monitoring of heparin therapy with APTT in sheep, and these results suggest that at 5 hours, heparin must be administrated by SC, in order to maintain the therapeutic range after the IV bolus initial dosing.(Keywords:APTI,'heparin monitoring, therapeutlc range, sheep)
