    아프리카돼지열병(African swine fever, ASF)은 사육돼지와 야생멧돼지에서 발생하는 치명적인 바이러스성 질병으로, 아프리카돼지열병바이러스(African swine fever virus, ASFV) 감염에 의해 유발되며 주로 돼지 단핵구/대식세포 계열 세포에 대한 높은 세포 특이성을 보이며 세포주에서 잘 증식하지 않는 특성을 나타낸다. 이러한 특성은 ASFV 대량 생산을 어렵게 만들어 연구 및 백신 개발에 큰 제약이 되고 있다. 본 연구에서는 한국에서 분리된 ASFV를 Vero 세포에 적응시키는 초기 단계에서 바이러스 클론들을 분리하고, 이들의 세포 내 특성을 분석하였다. 분리된 클론들 중 상이한 유전자 돌연변이 패턴을 나타내는 클론을 선별하여, 이를 기반으로 생백신 후보주로서 안전성과 유효성을 평가하였다.
    높은 병원성을 갖는 유전형 Ⅱ 형 ASFV 국내분리주를 Vero 세포에 적응시킨 뒤, 초기 적응 단계에서 플라크 정제(plaque purification)를 수행하여 유전적으로 다양한 바이러스 클론들을 확보하였다. 이 중 3개의 Vero 세포 적응 ASFV 클론을 최대 30번째 계대까지 각각 독립적으로 계대 배양하였으며, 모든 클론은 30번째 계대까지 돼지 폐포 대식세포(porcine alveolar macrophage, PAM) 및 Vero 세포에서 안정적인 증식을 유지함을 확인하였다. 전장유전체 분석 결과, 공통 및 고유의 단일 염기 다형성(single nucleotide polymorphisms, SNPs)과 더불어, 두 클론에서는 30번째 계대 시점에서 좌측 가변영역 (Left variable region, LVR)에 큰 유전자 결실이 발생하였다. 반면, VaCln3 클론은 대규모 유전자 결실 없이 12개의 비동의적 (non-synonymous) SNPs을 보였으며, 이 중 ASFV의 면역억제 기전과 연관된 것으로 알려진 MGF300-2R 및 MGF505-7R 유전자에 조기 종결코돈(stop codon) 형성이 확인되었다.
    VaCln3 클론의 13번째 계대 바이러스인 VaCln3 P13의 생백신 후보로서 평가를 위해 8주령 사육돼지에 102.0, 104.0, 106.0 TCID₅₀의 용량으로 근육 주사하고 체온, 임상증상, 바이러스혈증 등을 확인하였다. 접종 후 일부 돼지에서 일시적인 발열과 경미한 임상증상이 관찰되었으며, 고용량 접종 그룹(106.0 TCID50)의 돼지 1두는 Streptococcus suis에 의한 2차 감염으로 폐사하였고, ASF 특이 병변은 나타나지 않았다. 이 경우 일시적인 면역억제로 인한 기회감염 가능성을 배제할 수 없다. 모든 백신 접종군에서 ASFV 항체가 양성으로 확인되었으며, 고병원성 ASFV 국내분리주인 Paju1/19 공격접종에서 완전한 생존과 보호 효과가 확인되었다. 반면, 백신미접종 대조군은 모두 10일 이내에 폐사하였고, 육안 및 조직병리학적 분석 결과에서도 접종군에서 제한적인 병변만이 관찰되었다. 염기서열분석 결과에 따르면 일부 백신 접종군에서는 조직에서 Paju1/19유전자가 부분적으로 검출되었으나, 백신 바이러스에 의한 방어가 주된 역할을 한 것으로 확인되었다.
    결론적으로 본 연구에서는 Vero 세포 적응 초기 단계에서 Plaque purification을 통해 유전적으로 다양한 ASFV 클론을 확보할 수 있음을 확인하였다. 이 클론들 중 VaCln3 은 30번째 계대까지 대규모 유전자 결실 없이 안정적인 증식능을 나타냈으며, VaCln3 P13은 현저한 병원성 소실과 높은 방어능을 나타내어 향후 세포 적응 생백신 개발에 주요한 후보로 활용될 것으로 기대된다. 또한 본 연구 결과는 ASFV의 세포 적응 기전에 대한 이해를 확장하며, 안정적인 ASF 생백신 생산을 위한 기초자료로 활용될 것으로 기대된다.

    African swine fever (ASF) is a highly virulent disease affecting domestic pigs and wild boars. The virus that causes ASF is referred to as the African swine fever virus (ASFV), a complex and large DNA virus with a double-stranded genome and notable genomic variability. Despite the urgent need for effective vaccines, ASF vaccine development has been hindered by the lack of a continuous cell line suitable for large-scale virus propagation. ASFV exhibits a strong tropism for primary porcine macrophages, and attempts to adapt the virus to continuous cell lines often result in significant genomic deletions, which compromise the immunogenicity and protective efficacy of vaccine candidates.
    The primary objective of this study was to adapt a highly virulent Korean ASFV isolate (Korea/Pig/Paju1/2019) to Vero cells and to isolate genetically stable viral clones through early-stage plaque purification. Three Vero-adapted ASFV clones were obtained and independently passaged up to 30 times. All clones exhibited efficient replication in both porcine alveolar macrophages (PAMs) and Vero cells. Whole-genome sequencing revealed multiple shared and unique single-nucleotide polymorphisms (SNPs), with large deletions in the left variable region occurring in two clones by passage 30. Notably, one clone, VaCln3, retained genomic integrity without major deletions but carried 12 non-synonymous SNPs, including premature stop codons in the MGF300-2R and MGF505-7R genes, which have been identified as suppressors of host immune responses.
    Based on its favorable genetic and replication profiles, the VaCln3 clone at passage 13 (VaCln3 P13) was further evaluated as a live attenuated vaccine candidate in domestic pigs. Intramuscular vaccination with doses of 102.0, 104.0, or 106.0 TCID50 per pig showed that the virus was substantially attenuated, with only transient fever and mild clinical signs observed. One pig in the highest-dose group succumbed to a secondary Streptococcus suis infection; however, no ASF-specific pathology was detected. The death was likely due to opportunistic infection, although transient immunosuppression cannot be ruled out. All vaccinated pigs developed strong antibody responses and were completely protected against challenge with the virulent parental Paju1/19 strain. In contrast, all unvaccinated pigs developed severe ASF and died within ten days post-challenge. Clinical observation and gross and histologic examination revealed minimal lesions in the vaccinated pigs, while all pigs in the mock group died of typical ASF symptoms. Although the vaccine strain was identified in the tissues confirmed by Sanger sequencing, indicating persistence, challenge virus was still detected in some pigs, and this was more pronounced in the low-dose vaccinated group.
    This study suggests that ASFV adaptation in Vero cells may allow for the isolation of genetically distinct viral clones during early plaque purification. Of these, VaCln3 P13 showed stable replication without extensive gene deletions, an attenuated phenotype, and effective protection in pigs. These results contribute to the understanding of in vitro adaptation of ASFV and suggest a practical approach for the development of scalable vaccines using stable cell lines.
