    poly(A)+ RNA-결합 단백질인 Nab2는 mRNA의 3’ 말단 형성과 세포질로의 방출에 중요한 역할을 한다. mRNA의 가공과 방출에 관여하는 유전자들을 찾기 위해, 이전 연구에서 분열효모인 Schizosaccharomyces pombe에서 nab2 결핍과 합성치사를 보이는 3개의 돌연변이(SLN1~3)를 선별하였다. 이번 연구에서는 SLN3 균주의 생장과 mRNA 방출에 대한 결함을 부분적으로 상보하는 srp2 (SPAC16.02c) 유전자를 선별하였다. srp2 유전자는 생장에 필수적이고, pre-mRNA의 스플라이싱에 관여하는 SR 단백질로 알려져 있다. srp2를 과발현시키면 생장에 심각한 결함을 보였으며, 동시에 poly(A)+ RNA 가 핵 안에 강하게 축적되었다. 또한, Yeast two-hybrid 분석 실험에서 Srp2는 주요 mRNA 방출인자인 Mex67의 어뎁터로알려진 Mlo3와 상호작용하였다. 이러한 결과들은 분열효모에서도 srp2가 mRNA 스플라이싱 과정을 mRNA 방출과 연결하는 기능이 있음을 제시한다.

    The poly(A)+ RNA-binding protein Nab2 plays crucial roles in mRNA 3' end formation and mRNA nuclear export. To search for genes involved in mRNA processing and export, we previously isolated three synthetic lethal mutants (SLN1~3) that required nab2 for growth in fission yeast, Schizosaccharomyces pombe. In this study, we isolated srp2 (SPAC16.02c) gene as a multi-copy suppressor that partially complemented both growth and mRNA export defects of SLN3 mutant. The srp2 gene is known to be essential for growth and encodes a SR protein involved in pre-mRNA splicing. Overexpression of srp2 caused severe growth defect accompanied by intense accumulation of poly(A)+ RNA in the nucleus. Yeast two-hybrid analysis showed that Srp2 interacted Mlo3, which is known as an adaptor of major mRNA export receptor Mex67. These results suggest that the S. pombe srp2 may have functions in linking pre-mRNA splicing to mRNA export.

    The poly(A)+ RNA-binding protein Nab2 plays crucial roles in mRNA 3' end formation and mRNA nuclear export. To search for genes involved in mRNA processing and export, we previously isolated three synthetic lethal mutants (SLN1~3) that required nab2 for growth in fission yeast, Schizosaccharomyces pombe.
