The interactions of P-M system generate germ-line aberrations found in certain interstrain hybrid(M type female × P type male) in Drosophila melanogaster. We examined a natural population of Drosophila collected from Gwacheon city near Seoul in two consecutive years in order to determine the distribution of the chromosomal P factor activity and the extrachromosomal cytotype. No strong P factor activity for potential gonadal dysgenic sterility was found in this population. However, a search for the M and P cytotype that cause susceptibility and resistance to the P factor, respectively, showed polymorphic distribution for the extrachromosomal cytotypes. In the strain types resulted from gonadal dysgenesis of P-M system, Q and M' type were predominant as 98.13% in total 267 lines. In order to detect the quantity and variation of P element in total genomic DNA of Q and M' types, Southern blot analysis was performed with Pπ25.1 probe, an intact P element. According to the result, all lines appeared to contain a little complete P element as well as a plenty of defective P element, KP elements(0.48 Kb and 0.63 Kb). Also, it showed that the sizes of fragments were between 0.5 Kb and 1.8 Kb. Therefore, it was concluded that KP elements from Southern blot analysis of Q and M' lines must be reacted as a repressor which resists P-factor activity.
