    Glucocorticoids (GCs) are one of major stress hormones that protect the organisms from specific challenges. Voltage-gated potassium channels play a key to end action potentials in excitable cells, to control electrical signaling, and regulate ion flux and calcium transients in non-excitable cells. In the present study, I investigated the effect of cortisone and hydrocortisone on various voltage-gated potassium channels including hERG, human Kv1.5, and human Kv1.3 channels using a Xenopus oocyte expression system and a two-microelectrode voltage clamp amplifier to determine the electrophysiological effect of cortisone and hydrocortisone. Cortisone and hydrocortisone reduced the amplitude of the Kv1.3, Kv1.5, and hERG currents in concentration-dependent manner. Both cortisone and hydrocortisone inhibit the hERG channels in the closed, open, and inactivated state. The inhibitory effect of both cortisone and hydrocortisone on Kv1.3 and Kv1.5 channel current is acute and irreversible. The inhibition degree of cortisone on Kv1.5 decreased progressively from -10 mV to +30 mV, while that of hydrocortisone did not change at the same range of voltage. Cortisone shifted steady-state activation curves of Kv1.3 channel currents to the right direction, while hydrocortisone inhibited Kv1.3 channel steady-state currents more than peak currents and in a pattern of use-dependent block, which suggested an open-block manner. We concluded that Both cortisone and hydrocortisone blocked human Kv1.3, human Kv1.5 and hERG channel expressed in Xenopus oocytes, providing an possible electrophysiological mechanism for effect of cortisone and hydrocortisone on cardiovascular system and immune system.

Voltage-gated potassium channels play a key to end action potentials in excitable cells, to control electrical signaling, and regulate ion flux and calcium transients in non-excitable cells.     현대사회의 사람들은 빠른 생활패턴으로 인하여 많은 스트레스를 받고 있다. 지난 90년대의 연구자들은 장기적인 스트레스가 사람의 정신뿐만 아니라 신체에도 악영향을 미치며, 이로 인해 사망에 이를 수 있다고 밝혔다. Hydrocortisone은 Glucocorticoids 의 한 종류로, 중요한 스트레스 호르몬 중에 하나이다. 급작스런 스트레스 상황 시, 신경과 내분비 신호가 시상하부 - 뇌하수체 - 부신 축에 자극을 주게 되면, 부신피질에서 hydrocortisone이 분비된다. 이 호르몬은 심장 근육기능은 촉진시켜 심박 수를 증가시키고, 혈압 및 혈당을 상승시키며, 세기관지 확장, 면역과 소화기능을 억제 시킨다.
    Voltage-gated K+channel은 K+를 이동시키는 막 단백질이다. 이 channel은 막 전압에 따라 개폐되며, 활동전압에서 안정 막 전압으로 회복시킨다. 또한 전기신호와 Ca2+transients를 조절하여 우리 몸의 수많은 활동을 조절한다. 우리는 cortisone과 hydrocortisone이 xenopus oocyte에 발현시킨 hERG, Kv1.5, Kv1.3 current에 대해, 어떠한 영향을 미치는지 two-microelectrode voltage clamp 시스템을 통하여 측정하였다. 실험 결과, cortisone과 hydrocortisone 모두 농도 의존적으로 hERG, Kv1.5, Kv1.3 전류를 감소시켰다.
