Smallpox is a serious, contagious, and sometimes fatal infectious disease. There is no specific treatment for smallpox disease, and the only prevention is vaccination. The pox part of smallpox is derived from the Latin word for “spotted” and refers to the raised bumps that appear on the face and body of an infected person. Smallpox is an acute contagious disease caused by variola virus, a member of the orthopoxvirus family. Smallpox, which is believed to have originated over 3,000 years ago in India or Egypt, is one of the most devastating diseases known to humanity. For centuries, repeated epidemics swept across continents, decimating populations and changing the course of history. The disease, for which no effective treatment was ever developed, killed as many as 30% of those infected. Between 65–80% of survivors were marked with deep pitted scars (pockmarks), most prominent on the face. In early 19th century, Smallpox vaccine was developed and eradicated smallpox on the world. But the previously developed vaccine has a lot of side effect to vaccinated human. So, the development of next generation vaccine was required with safe and efficacious properties. For economical development of vaccinia virus, we constructed research cell and virus bank. We hope to establish the basis for developing new smallpox vaccine. This study can be summarized as below
1) Vero cell bank adapted to serum-free media was constructed for adapting vaccinia virus to serum free media.
2) Research vaccinia virus bank preparation.
3) Characterization of research cell bank and virus bank.
