약독화 두창 백신 개발을 위한 Research Bank 구축 = Establishment of Research Bank for attenuated smallpox vaccine development

Smallpox is a serious, contagious, and sometimes fatal infectious disease. There is no specific treatment for smallpox disease, and the only prevention is vaccination. The pox part of smallpox is derived from the Latin word for “spotted” and refers to the raised bumps that appear on the face and body of an infected person. Smallpox is an acute contagious disease caused by variola virus, a member of the orthopoxvirus family. Smallpox, which is believed to have originated over 3,000 years ago in India or Egypt, is one of the most devastating diseases known to humanity. For centuries, repeated epidemics swept across continents, decimating populations and changing the course of history. The disease, for which no effective treatment was ever developed, killed as many as 30% of those infected. Between 65–80% of survivors were marked with deep pitted scars (pockmarks), most prominent on the face. In early 19th century, Smallpox vaccine was developed and eradicated smallpox on the world. But the previously developed vaccine has a lot of side effect to vaccinated human. So, the development of next generation vaccine was required with safe and efficacious properties. For economical development of vaccinia virus, we constructed research cell and virus bank. We hope to establish the basis for developing new smallpox vaccine. This study can be summarized as below
1) Vero cell bank adapted to serum-free media was constructed for adapting vaccinia virus to serum free media.
2) Research vaccinia virus bank preparation.
3) Characterization of research cell bank and virus bank.

국문 초록 (Abstract)

천연두 (두창)는 두창 바이러스 (variolar virus)가 원인이 되는 심각한 바이러스 감염 질환으로 인류 역사 초기부터 인간을 괴롭혀왔다. 인류역사상 전쟁과 다른 전염병으로 죽은 사람들을 모두 합친 것보다 많은 5억여 명이 이 질병으로 희생된 것으로 추정하고 있다. 남미 아즈텍문명과 잉카문명이 천연두의 등장으로 붕괴되었고 미국은 천연두를 전염시켜 인디언들을 멸망시켰다. 우리나라도 6.25 전쟁 중이던 1951년 한 해에 4만 여명이 천연두에 걸렸다. 기존 두창백신은 효능은 뛰어나나 양적으로 부족하며 접종 받은 사람에게 심각한 부작용을 일으키는 등 여러 가지 문제점이 보고되고 있다. 이에 전 세계적으로 안전하고 효능이 우수한 차세대 두창 백신 연구가 활발히 진행 중이다. 현재 국립보건연구원에서 개발 중인 약독화 두창 백신 후보주의 실용화 연구를 위하여 확보된 백신 후보주를 bank화 하여 안정적인 약독화 백시니아 바이러스를 확보하고자 하며, 이를 위해 Research Bank를 확립함으로서 차세대 두창백신 개발을 위한 기반을 마련하고자 한다. 본 연구개발결과로서는 다음과 같이 요약 할 수 있다.
1) 백신 후보주를 bank화 하기 위하여 무혈청 배지에 숙주세포를 adaptation 시킨 후 cell bank화 하였다.
2) 백신 후보주를 무혈청 배지에 adaptation 된 cell에 연속하여 계대 배양 함으로서 7 x 10⁵의 titer를 보이는 virus strain을 확보하였으며, 선별된 백신 후보주를 사용하여 research virus bank를 제작하였다.
3) Research Cell Bank와 Research Virus Bank에 대한 characterization 시험으로 무균시험, 순도시험, 동정시험을 하여 실시하였다.
