Gelatin containing vaccines occasionally induce allergic reactions including anaphylaxis, and anti -gelatin IgE has been detected in sera obtained from patients with systemic immediate-type reactions. Furthermore, all the patients sensitized by gelatin develop gelatin specific lymphocyte proliferation activity. Even though the patients with systemic nonimmediate-type reactions do not produce any anti-gelatin IgE, some of them have identified to produce gelatin specific IgG. In this study we tried to setup a sensitive method to detect anti-gelatin antibodies in human sera. Since the levels of anti-gelatin antibodies are little in sera, large amount of sera should be used. Anti-gelatin IgG could be measured by an indirect ELISA using a colorimetric (OPD) substrate. However, anti -gelatin IgE could not be detected using the colorimetric substrate, therefore the chemiluminescent substrate was used. The Japanese reference serum (JR) obtained from a patient with gelatin allergy was used as a positive control. It found to contain high concentrations of both anti-gelatin IgG and IgE, indicating that our system is useful for detection of anti -gelatin antibodies. Of 26 antisera two patient sera, DS-3 and CS-6, also were also proved to have relatively higher concentration of IgG and IgE specific to gelatin. Anti -gelatin IgGs in 5 sera showed low affinities to gelatin, but anti-gelatin IgEs in those sera were found to have relatively higher affinities to gelatin. Two antibodies in two sera, DS-8 and CS-3, showed nonspecific binding patterns. Both wells with and without gelatin coating showed relatively high reactivity to both IgG and IgE in those sera. The other sera were found to contain little anti-gelatin IgGs and IgEs. Since there are very small amount IgE in sera. we tried to make a system for detection of anti-gelatin IgE. After capturing the human IgE using anti-human IgE mouse monoclonal antibody, biotinylated gelatins bound to the human IgE. However. no specific binding was found in all the samples tested including DS · :3. The gelatins on the glass slide were found to use for detecting anti- gelatin antibody in sera with high sensitivity.
