    화학작용제(Chemical Warfare Agents, CWAs)와 독성 산업화학 물질(Toxic Industrial Chemicals, TICs)은 전 지구적 안보, 환경 안전, 그리고 공중보건에 지속적인 위협을 가하고 있다. 그러나 이들 물질은 극도의 독성과 엄격한 규제적 제약으로 인해 직접적인 실험 연구가 제한되며, 그 결과 신뢰성 있는 독성 및 검출 데이터가 부족한 실정이다. 본 학위논문은 이러한 한계를 극복하기 위하여 계산화학, 정량적 구조–활성 상관관계(QSAR) 모델링, 그리고 기계학습 기법을 통합하여 화학위협의 예측, 검출, 대응을 포괄하는 평가 체계를 제시하고자 한다.
    먼저, 유기인계 신경작용제의 급성 경구 독성(LD50)을 예측하기 위한 기작 기반 계산 프레임워크를 개발하였다. 밀도범함수이론(DFT) 계산, 분자 도킹, 통계적 모델링을 결합함으로써 분자 수준의 상호작용 에너지와 거시적 독성 지표 간의 정량적 연관성을 규명하였다. 제한된 데이터셋으로 인해 예측 일반화 성능에 한계는 존재하였으나, 아세틸콜린에스터레이스 저해 기작에 대한 기계적 이해를 제공하고 데이터 기반 독성 예측을 위한 재현 가능한 기반을 확립하였다.
    다음으로, 데이터 부족 문제를 극복하기 위해 16,000종 이상의 구조적 유사체를 통합한 대규모 QSAR 앙상블 모델을 구축하여 독성 산업화학물질의 다중 종말점 위험성을 예측하였다. NFPA 및 GHS 분류 체계와 정합된 위험도 산출을 통해 주요 종말점 전반에서 80% 이상의 균형 정확도를 달성하였으며, 이를 통해 화학작용제와 산업화학물질을 효과적으로 구분할 수 있음을 확인하였다.
    마지막으로, 상용 형광 염료를 이용한 이중 모드 컬러리메트릭 센서 어레이를 개발하여 가시광 및 자외선 조건에서 화학작용제의 신속 검출을 구현하였다. 기계학습 분류 기법, 특히 Random Forest 모델을 적용한 결과 90.0%의 분류 정확도를 달성하였으며, 이를 통해 저비용·현장형 화학위협 식별 기술로서의 가능성을 입증하였다. 또한 신경작용제 모사체의 피롤 기반 제거 반응과 VX에 의한 항공우주 소재 부식 현상에 대한 실험–계산 융합 연구를 추가적으로 수행하였다.
    종합적으로 본 논문은 기작 기반 모델링과 대규모 데이터 분석을 유기적으로 결합한 통합적 화학위협 평가 프레임워크를 제시함으로써, 현대 화학방어를 위한 예측 가능하고 해석 가능하며 현장 적용이 가능한 기술 개발에 기여한다.

    Chemical warfare agents (CWAs) and toxic industrial chemicals (TICs) pose persistent threats to global security, environmental safety, and public health. However, their extreme toxicity and strict regulatory restrictions hinder direct experimental investigation, resulting in a scarcity of reliable toxicity and detection data. This dissertation addresses these challenges by integrating computational chemistry, quantitative structure–activity relationship (QSAR) modeling, and machine learning approaches to advance chemical threat assessment through prediction, detection, and mitigation.
    First, a mechanistically informed computational framework was developed to predict the acute oral toxicity (LD50) of organophosphorus nerve agents. By combining density functional theory (DFT) calculations, molecular docking, and statistical modeling, the study quantitatively linked molecular-level interaction energies with macroscopic toxicological outcomes. Although the limited dataset constrained predictive generalization, the framework provided mechanistic insight into acetylcholinesterase inhibition and established a reproducible foundation for data-driven toxicity prediction.
    Second, to overcome data scarcity, a large-scale QSAR ensemble model was constructed for multi-endpoint hazard prediction of TICs. By integrating more than 16,000 structural analogs and aligning hazard outputs with NFPA and GHS systems, the model achieved over 80% balanced accuracy across key endpoints and successfully differentiated CWAs from industrial chemicals.
    Finally, a dual-mode colorimetric sensor array employing 29 commercial fluorescent dyes was developed for rapid CWA detection under visible and UV illumination. Machine learning classification, particularly Random Forest, achieved 90.0% accuracy, enabling lowcost,field-deployable chemical threat identification. Complementary experimental–computational studies further explored pyrrole-based remediation of nerve agent surrogates and VX-induced corrosion on
    aerospace materials.
    Collectively, this dissertation presents an integrative framework that bridges mechanistic modeling and large-scale data analytics, contributing to the development of predictive, interpretable, and deployable technologies for modern chemical defense.
