본 연구는 말 인플루엔자 바이러스 (A/equine/kyonggi/SA1/2011 (H3N8), EIV)에서 유래한 자연적으로 절단된 NS1 단백질을 발현하는 비구조 유전자를 이용해 개 인플루엔자 생백신을 개발하는 데 중점을 두었습니다. 절단된 NS 유전자는 역유전학을 통해 개 인플루엔자 바이러스 (A/canine/korea/01/2007 (H3N2)) 유전체에 삽입되었으며, 생성된 재조합 바이러스는 세포 및 유정란에서 원래 바이러스와 유사한 증식능을 보여 대량 생산 가능성을 확인했습니다. 본 연구의 주요 목적은 효과적인 개인플루엔자 생독백신을 개발한 후, 이 백신의 면역원성과 안전성을 평가하며, 새로운 투여 경로의 가능성을 탐색하여 개 인플루엔자에 대한 보호 효과를 극대화하는 것입니다. 이를 위해 마우스와 개를 대상으로 백신의 약독화 기전, 면역 반응, 보호 효능을 포괄적으로 분석하고, 특히 개에서의 설하 필름 제형 투여를 호흡기 점막 면역 유도 전략으로서 실현 가능한 대안으로 제시했습니다.
마우스 실험에서는, 절단된 비구조 단백질이 RIG-I 기반 선천 면역 반응을 억제하지 않음을 확인하였고, 이에 따라 바이러스의 폐 병변과 바이러스 역가가 감소하며 약독화가 일어남을 입증하였습니다. 생독 백신은 불활화 백신에 비해 월등히 높은 체액성 및 세포성 면역 반응을 유도하였고, 더 강력한 보호 효능을 보였습니다. 특히 비강을 통해 접종된 생독 백신은 근육접종 불활화 백신으로는 유도되지 않는 점막 면역을 유도하였습니다.
개 실험에서는 마우스에서 안전성 및 효능이 확인된 생독 백신을 활용하여 비강접종시 점막면역 유도 및 바이러스 전파 감소 효과를 보일 수 있을지 실험을 진행하였습니다. 우선, 같은 역가의 야생형 개인플루엔자 바이러스 비해 안전성을 보이는 것을 확인하였으며, 이 백신에 접종된 개는 상부호흡기에 높은 점막면역을 유도할 수 있음을 확인하였습니다. 또한 근육접종 사백신 접종보다 높은 체액성면역을 보이는 것을 확인하였으며, 높은 역가의 야생형 개인플루엔자 바이러스를 공격접종하여 보호능을 확인해보고자 하였습니다. 결과, 점막면역 유도에 따라 바이러스 전파율이 낮아질 것이 예측되었으며, 생독백신이 사독백신 근육접종에 비해 개인플루엔자에 대해 개체에서의 방어능이 더 높은 것을 확인하였습니다. 이를 통해 비강접종 용동의 개인플루엔자 생독백신 후보주를 도출하였습니다.
비강접종의 점막 면역 유도를 유지하면서도 백신의 안전성을 높이기 위해, 마우스를 대상으로 설하 투여의 안전성과 효능을 평가하였습니다. 그 결과, 설하 접종 후 상·하기도에서 바이러스가 검출되지 않았으며, 조직 분석을 통해 비강 경로보다 더 뛰어난 안전성을 나타냈습니다. 비강 접종 또한 효과적이고 안전했지만, 설하 경로는 호흡기계 내 바이러스 확산 위험을 제거하고 정량 투여의 장점까지 갖추며 추가적인 안전 이점을 제공하였습니다. 특히, 설하 백신은 마우스에서 계대된 개인플루엔자 바이러스의 공격접종 후 100% 생존율을 달성했으며, 기존의 근육주사 방식보다 우수한 보호 효과를 보였습니다. 또한, 1회의 설하 생백신 접종만으로도 2회 근육 접종보다 뛰어난 면역 및 보호 능력을 유도했습니다. 개를 대상으로 한 실험에서 설하 필름 제형의 생독 백신은 높은 체액성, 세포성 및 점막 면역을 유도하였으며, 바이러스 배출이나 다른 부작용을 나타내지 않는 것을 확인하였습니다. 이 마우스 실험과 개실험을 통해 설하접종을 통한 호흡기 점막면역 유도 개인플루엔자 생독 백신 후보주 도출을 완료하였습니다.

The disease poses significant health risks, particularly in crowded environments such as kennels and dog parks. As a persistent threat, canine influenza can result in widespread outbreaks, affecting both the health of individual dogs and the overall canine population. The virus's ability to mutate complicates control efforts and necessitates the development of effective preventive measures. Current vaccines have limitations in providing comprehensive protection against various viral strains, highlighting the urgent need for improved vaccine options. Developing a more effective vaccine is crucial to enhance immunity and reduce the incidence of canine influenza outbreaks. This study focuses on the development of a Live Attenuated Influenza Vaccine (LAIV) for canine influenza, utilizing a naturally truncated nonstructural (NS) gene identified from an equine influenza virus (A/equine/kyonggi/SA1/2011 (H3N8), EIV). The LAIV candidate was engineered using reverse genetics by inserting this truncated NS gene into a CIV backbone (A/canine/korea/01/2007 (H3N2)). The resulting recombinant virus demonstrated a growth rate in cell cultures and embryonated eggs comparable to the parental CIV, confirming its potential for large- scale vaccine production. The primary objectives of this research were to develop an effective LAIV, evaluate its efficacy and safety profile, and explore novel administration routes for enhanced protection against canine influenza. Through comprehensive testing in both mouse models and dogs (beagles), the study aimed to assess the vaccine's attenuation mechanism, immunogenicity, and protective efficacy, while also investigating the potential of Sublingual (SL) film formulation in dogs as an innovative mucosal delivery method. In mice, vaccination confirmed that the truncated NS1 protein did not suppress the RIG-I-mediated innate immune response, resulting in viral attenuation as evidenced by reduced lung lesions and viral titers. In comparison with inactivated vaccines, the LAIV elicited significantly higher levels of humoral and cellular immunity, along with enhanced protective efficacy. Notably, the intranasally vaccinated LAIV induced mucosal immunity, which is not typically elicited by inactivated vaccines. To further improve safety along with inducing mucosal immunity, safety and efficacy evaluations of the SL administration route were conducted in mice. Results showed that no viral detection occurred in the upper or lower respiratory tracts following SL vaccination of LAIV, and tissue analysis support that SL vaccination provided greater safety than the intranasal (IN) route. While IN vaccination was also effective and safe, the SL route conferred additional safety advantages by eliminating any risk of viral dissemination in respiratory tracts and ensuring more precise dosing. Importantly, it achieved 100% survival against 20 MLD50 of Mouse-adapted canine influenza virus (MACIV) and outperformed intramuscular (IM) delivery in protective efficacy. Specifically, a single dose of SL vaccination induce superior immune and protective capabilities than two doses of IM vaccination. Finally, in beagle dogs, SL administration of the LAIV film elicited robust humoral, cellular and mucosal immunity without causing any detectable viral shedding or clinical symptoms. This study demonstrates the successful development of a highly effective LAIV for canine influenza, offering superior immunogenicity and protection compared to traditional inactivated vaccines. This is the first demonstration of combining an NS1- truncated live influenza virus with a SL film delivery platform for mucosal immunization in veterinary medicine. This novel approach represents a significant advancement in vaccine technology and could potentially revolutionize canine influenza prevention strategies. The findings of this research not only provide a promising new vaccine candidate but also open up new avenues for vaccine delivery and immune response optimization in veterinary medicine. Future studies may focus on large-scale field trials and elucidating the precise mechanisms by which SL vaccination induces mucosal immunity, paving the way for more effective and user- friendly vaccination protocols in real-world settings. This SL film vaccine platform offers an unprecedented combination of mucosal immunogenicity, safety, dose precision, and ease of administration, making it a leading candidate for next- generation influenza control strategies in dogs.
