In this study, we optimized the production process of growth-promoting factors from Mycobacterium kansasii and produced growth-enhancing media for M. tuberculosis culture. Then the media were evaluated in BacT/Alert culture system using M. tuberculosis H37Rv, K, HN878 strains, 41 clinical isolates, and 30 sputum collected from tuberculosis patients.
○ Production and quantification of growth-promoting factors for M. tuberculosis culture
- Large scale production of M. kansasii cells by surface culture or shaking culture
- Production of growth-promoting factors from M. kansasii cells according to the method established in the 1st year
- Optimization of the production process of growth-promoting factors from M. kansasii by adding lysozyme or glycine to the culture / by disrupting the harvested cells with sonicator or French press
