To investigate the effects of ginseng saponin and 3´-methyl-4-dimethyl aminoazobenzene(3´-Me-DAB) on the expression of transferrin receptor(TfR) in the regenerating liver, we have studied [³H]thymidine uptake, ^125I-transferrin binding, and mRNA expression of the TfR from normal regenerated rat liver after partial hepatectomy, or from the liver of gingseng saponin and 3´-Me-DAB treated rat. Results are as follows;
1.With the administration of 3´-Me-DAB for 8 weeks, the liver tissue showed marked morphologic changes of regenerative change of hepatocytes, lymphocyte infiltration, atypical proliferation of bile ducts and advanced bridging necrosis, but these changes were little affected by partial hepatectomy or ginseng saponin.
2.In the [³H]thymidine incorporation assay, DNA synthesis was increased 6 and 7 folds by the 1st day after partial hepatectomy of normal and 3´-Me-DAB treated rat, compared to those of normal rat liver. And it was decreased slightly by the treatment of ginseng in the both groups.
3.Transferrin binding values in plasma membrane from the regenerated liver of normal rat were increased by 3rd day and diminished at 5th day after partial hepatectomy. With the treatment of 3´-Me-DAB, transferrin binding sites were higher than those of normal rat liver and further increased by 3rd day after partial hepatectomy. Ginseng saponin decreased about 35% by 3rd day after partial hepatectomy of 3´-Me-DAB treated rat, but there was little effect on transferrin binding sites of the plasma membrane of normal rat.
4.The mRNA expression of TfR in the nomal regenerated rat liver was increased at 8hr and peak at 24hr and abolished to control level at 72hr after partial hepatectomy, and that in the liver of 3´-Me-DAB treated rat was higher than that in normal regenerated rat liver. Ginseng saponin decreased mRNA expression of TfR by 8hr and 24hr day after partial hepatectomy of 3´-Me-DAB treated rat, but there was no significant change on the normal regenerated rat liver.
These results suggest that the increase of TfR binding sites in the regenerated rat liver with 3´-Me-DAB treatment might be correlated with transcriptional regulation of TfR gene, and ginseng saponin could inhibit the expression of TfR gene at the proliferative state.
